    Identifying the specific functional regulator of integrin family molecules in cancer cells is critical because they are directly involved in tumor invasion and metastasis. Here we report high expression of PLOD2 in oropharyngeal squamous cell carcinomas (SCCs) and its critical role as a stabilizer of integrin β1, enabling integrin β1 to initiate tumor invasion/metastasis. Integrin β1 stabilized by PLOD2-mediated hydroxylation was recruited to the plasma membrane, its functional site, and accelerated tumor cell motility, leading to tumor metastasis in vivo, whereas loss of PLOD2 expression abrogated it. In accordance with molecular analysis, examination of oropharyngeal SCC tissues from patients corroborated PLOD2 expression associated with integrin β1 at the invasive front of tumor nests. PLOD2 is thus implicated as the key regulator of integrin β1 that prominently regulates tumor invasion and metastasis, and it provides important clues engendering novel therapeutics for these intractable cancers

    The Dense Molecular Gas and Nuclear Activity in the ULIRG IRAS 13120-5453

    We present new ALMA Band 7 (340\sim340 GHz) observations of the dense gas tracers HCN, HCO+^+, and CS in the local, single-nucleus, ultraluminous infrared galaxy IRAS 13120-5453. We find centrally enhanced HCN (4-3) emission, relative to HCO+^+ (4-3), but do not find evidence for radiative pumping of HCN. Considering the size of the starburst (0.5 kpc) and the estimated supernovae rate of 1.2\sim1.2 yr1^{-1}, the high HCN/HCO+^+ ratio can be explained by an enhanced HCN abundance as a result of mechanical heating by the supernovae, though the active galactic nucleus and winds may also contribute additional mechanical heating. The starburst size implies a high ΣIR\Sigma_{IR} of 4.7×10124.7\times10^{12} LL_{\odot} kpc2^{-2}, slightly below predictions of radiation-pressure limited starbursts. The HCN line profile has low-level wings, which we tentatively interpret as evidence for outflowing dense molecular gas. However, the dense molecular outflow seen in the HCN line wings is unlikely to escape the galaxy and is destined to return to the nucleus and fuel future star formation. We also present modeling of Herschel observations of the H2_2O lines and find a nuclear dust temperature of 40\sim40 K. IRAS 13120-5453 has a lower dust temperature and ΣIR\Sigma_{IR} than is inferred for the systems termed "compact obscured nuclei" (such as Arp 220 and Mrk 231). If IRAS 13120-5453 has undergone a compact obscured nucleus phase, we are likely witnessing it at a time when the feedback has already inflated the nuclear ISM and diluted star formation in the starburst/AGN core.Comment: accepted for publication in ApJ, 21 pages, 11 figure

    Correlation of Heat Shock Protein Expression to Gender Difference in Development of Stress-Induced Gastric Mucosal Injury in Rats

    Recent studies have indicated that heat shock proteins (HSPs), which function as molecular chaperones, play important roles in cellular responses to stress-related events. However, the gender difference in the expression of HSP in the gastric mucosa remains unclear. In order to understand the mechanism of gender difference in the prevalence or severity of gastric mucosal lesions, the expression level of HSP and the correlation of estrogen to HSP induction in the gastric mucosa were evaluated in this study. The basal expression levels of HSP60 and HSP90 in the gastric mucosa were significantly higher in females than those in males. The gastric ulcer index was significantly higher in male rats compared to female rats observed after 12 h water immersion stress exposure. At this time point, the expression levels of HSP60 and HSP90 in the gastric mucosa were significantly higher in females than those in males. An estrogen-treatment significantly induced the expression of HSP60, HSP70 and HSP90 in the gastric mucosa. Inversely, an ovariectomy dramatically reduced the expression of HSP60, HSP70 and HSP90 in the gastric mucosa. Our results suggested that estrogen might play an important role in gastric mucosal protection with the induction of gastric mucosal HSPs

    In situ evaluation of podocin in normal and glomerular diseases.BackgroundMutations of the NPHS2 gene are responsible for autosomal-recessive steroid-resistant nephrotic syndrome. Its product, podocin, faces the slit diaphragm area with its two ends in the cytoplasm of foot processes.MethodsWe generated rabbit polyclonal antibodies against conjugated peptides from human podocin N- and C-termini, and studied podocin and synaptopodin using kidney tissues of normal humans and those with glomerular diseases.ResultsAntipodocin antibodies detected the original 42 kD fragment and an extra smaller fragment by Western blot analysis using human isolated mature glomeruli. RNA analysis showed two bands, the original and the other of a decreased length. Immunohistochemically, podocin was detected in a linear pattern along the glomerular capillary loop. Antipodocin antibody (C-terminal) stained the smooth muscles of renal arterioles and aorta. Among 42 patients, podocin was normally expressed in glomeruli in purpura nephritis, IgA nephropathy (IgAN), and minimal-change disease (MCD), while it was either decreased or absent in most subjects with focal segmental glomerulosclerosis (FSGS). The expression of synaptopodin was similar to that of podocin, although some discrepancy existed.ConclusionAlthough indirect, our data suggest the existence of a vascular isoform of podocin with a different molecular mass. We propose that examination of podocin expression may help differentiate MCD from FSGS

    Performance, egg quality and nitrogen balance of commercial laying hens fed diets with different levels of crude protein and lysine

    O experimento foi conduzido para avaliar os efeitos de rações com diferentes níveis de proteína bruta (PB) e lisina sobre as características de desempenho, a qualidade interna dos ovos e o balanço/retenção do nitrogênio. Foram utilizadas 160 poedeiras Hisex White com 48 semanas de idade, alojadas individualmente em delineamento inteiramente casualizado em esquema fatorial 4 × 2, com quatro níveis de PB (12, 14, 16 e 18%) e dois de lisina (0,85 e 1,00%), totalizando oito rações com cinco repetições de quatro aves. O consumo de proteína bruta, o peso dos ovos, a massa de ovos e a porcentagem de albúmen apresentam resposta linear crescente aos níveis de PB na dieta. O balanço de nitrogênio não é alterado pelos níveis de proteína das rações.The experiment was carried out to evaluate the effects of diets with different levels of crude protein (CP) and lysine (LYS) on characteristics of performance, egg internal quality, and nitrogen balance/retention. It was used 160 Hisex White laying hens at 48 weeks of age, individually accomodated in a complete random design in a 4 × 2 factorial scheme with four levels of crude protein (12, 14, 16 and 18%) and two levels of lysine (0.85 and 1.00%), totalizing eight treatments with five replicates of four birds each. Crude protein intake, egg weight, egg mass and percentage of the albumen show increasing linear response to the levels of crude protein in the diet.     Virological characterization of the 2022 outbreak-causing monkeypox virus using human keratinocytes and colon organoids

    ヒトiPS細胞由来大腸オルガノイドおよびヒトケラチノサイトを用いたmpoxウイルス2022年株の解析. 京都大学プレスリリース. 2023-06-07.The outbreak-causing monkeypox virus of 2022 (2022 MPXV) is classified as a clade IIb strain and phylogenetically distinct from prior endemic MPXV strains (clades I or IIa), suggesting that its virological properties may also differ. Here, we used human keratinocytes and induced pluripotent stem cell-derived colon organoids to examine the efficiency of viral growth in these cells and the MPXV infection-mediated host responses. MPXV replication was much more productive in keratinocytes than in colon organoids. We observed that MPXV infections, regardless of strain, caused cellular dysfunction and mitochondrial damage in keratinocytes. Notably, a significant increase in the expression of hypoxia-related genes was observed specifically in 2022 MPXV-infected keratinocytes. Our comparison of virological features between 2022 MPXV and prior endemic MPXV strains revealed signaling pathways potentially involved with the cellular damages caused by MPXV infections and highlights host vulnerabilities that could be utilized as protective therapeutic strategies against human mpox in the future
