    A new device for the simultaneous recording of cerebral, cardiac, and muscular electrical activity in freely moving rodents

    AbstractWe present a new technique for the simultaneous capture of bioelectrical time signals from the brain and peripheral organs of freely moving rodents. The recording system integrates all systemic signals into an electrical interface board that is mounted on an animal's head for an extended period. The interface board accommodates up to 48 channels, enabling us to analyze neuronal activity patterns in multiple brain regions by comparing a variety of physiological body states over weeks and months. This technique will advance the understanding of the neurophysiological correlate of mind–body associations in health and disease

    Field Projection Device of Atomic Bomb Dome by Augmented Reality

    We developed a projection device with Head Mounted Display through which visitors to Atomic Bomb Dome can see the 3D image of what the dome used to be before the dropping of the bomb. This wearable device, based on the technology of augmented reality (AR), enables wearers to have a clear understanding of the effect of the bomb by showing the difference before and after the catastrophe, because the 3D image of Hiroshima prefectural products museum is projected exactly where Atomic Bomb Dome is seen in the wearer's field of view. This development was carried out in the wake of our field interview for foreign visitors. Many hoped that they would learn about Hiroshima prefectural products museum and some suggested that we build its life-size replica just beside the dome. Since building it physically is out of the question, we struck on the idea of building it virtually utilizing AR technology. Technically, our device is composed of a Head Mounted Display available on the market and an original application we programed. The difficulty of the development lay in the way of programing and installing the original software because its built-in installer could install only a few official applications for the hardware and therefore it might not execute any other original applications. Even if it could, controlling built-in hardware devices, a camera in this particular case, was not guaranteed. Through trial and error we found a way to install our software and execute it. Controlling the camera was also successful

    Mapping the Ethical Issues of Brain Organoid Research and Application

    脳オルガノイドの研究と臨床応用での倫理問題を体系化. 京都大学プレスリリース. 2021-04-07.Society is not ready to make human brains. 京都大学プレスリリース. 2021-03-26.In 2008, researchers created human three-dimensional neural tissue – known as the pioneering work of “brain organoids.” In recent years, some researchers have transplanted human brain organoids into animal brains for applicational purposes. With these experiments have come many ethical concerns. It is thus an urgent task to clarify what is ethically permissible and impermissible in brain organoid research. This paper seeks (1) to sort out the ethical issues related to brain organoid research and application and (2) to propose future directions for additional ethical consideration and policy debates in the field. Toward (1), this paper first outlines the current state of brain organoid research, and then briefly responds to previously raised related ethical concerns. Looking next at anticipated scientific developments in brain organoid research, we will discuss (i) ethical issues related to in vitro brain organoids, (ii) ethical issues raised when brain organoids form complexes or have relationships with other entities, and (iii) ethical issues of research ethics and governance. Finally, in pursuit of (2), we propose research policies that are mindful of the ethics of brain organoid research and application and also suggest the need for an international framework for research and application of brain organoids

    Moving toward generalizable NZ-1 labeling for 3D structure determination with optimized epitope-tag insertion

    タンパク質の抗体ラベリング技術を改良し、構造解析をアシスト --電子顕微鏡やX線結晶解析による構造決定を加速化--. 京都大学プレスリリース. 2021-04-20.Antibody labeling has been conducted extensively for structure determination using both X-ray crystallography and electron microscopy (EM). However, establishing target-specific antibodies is a prerequisite for applying antibody-assisted structural analysis. To expand the applicability of this strategy, an alternative method has been developed to prepare an antibody complex by inserting an exogenous epitope into the target. It has already been demonstrated that the Fab of the NZ-1 monoclonal antibody can form a stable complex with a target containing a PA12 tag as an inserted epitope. Nevertheless, it was also found that complex formation through the inserted PA12 tag inevitably caused structural changes around the insertion site on the target. Here, an attempt was made to improve the tag-insertion method, and it was consequently discovered that an alternate tag (PA14) could replace various loops on the target without inducing large structural changes. Crystallographic analysis demonstrated that the inserted PA14 tag adopts a loop-like conformation with closed ends in the antigen-binding pocket of the NZ-1 Fab. Due to proximity of the termini in the bound conformation, the more optimal PA14 tag had only a minor impact on the target structure. In fact, the PA14 tag could also be inserted into a sterically hindered loop for labeling. Molecular-dynamics simulations also showed a rigid structure for the target regardless of PA14 insertion and complex formation with the NZ-1 Fab. Using this improved labeling technique, negative-stain EM was performed on a bacterial site-2 protease, which enabled an approximation of the domain arrangement based on the docking mode of the NZ-1 Fab
