    Experiência no curso de estudantes de 1º ano – um estudo no âmbito das tutorias de acompanhamento na Universidade de Évora

    O presente estudo pretende conhecer a experiência no curso de estudantes de 1º ano que ingressaram na Universidade de Évora no final do 1º semestre. Estudos realizados sobre a percepção dos estudantes relativa ao contexto de aprendizagem no ensino superior indicam forte relação com as abordagens à aprendizagem e apresentam forte relevância para a compreensão da forma como os estudantes acedem ao conhecimento e para a definição de processos de aprendizagem de elevada qualidade (Entwistle, 2009; Chaleta & Entwistle, 2011). Os dados foram obtidos através da aplicação do CEQP (Ramsden, 2005; 2006; Chaleta et al, 2012) com 565 estudantes de diferentes cursos e áreas científicas. Os resultados indicaram que a experiência no curso é positiva para o conjunto dos estudantes havendo necessidade de observar com mais atenção as questões relacionadas com a avaliação. A grande maioria dos estudantes revela também satisfação com o curso que frequenta. Palavras-Chave: Experiência no Curso; Tutorias de Acompanhamento; CEQP; Ensino Superior. Abstract This study examine the experience in the course of the 1st year students who entered at the University of Évora. Studies on the perception of students on the learning environment in higher education indicate a strong relationship with the approaches to learning and have strong relevance to the understanding how students access the knowledge and the definition of high quality learning processes (Entwistle, 2009; Chaleta & Entwistle, 2011). The data were obtained by applying the CEQP (Ramsden, 2005, 2006; Chaleta et al, 2012) with 565 students from different courses and scientific areas. The results indicated that the course experience is positive for all the students but we need to look more closely at the issues related to assessment.     Performance, environmental, and mobility analysis of large capacity fast rotorcraft configurations for the European regional air traffic market

    Fast, large rotorcraft are of interest in the future European air transport system due to their runway independent operation and potential mobility improvements for the passenger. Both a tiltrotor and a coaxial compound concept model were developed for a 70 passenger, 500 NM design mission that would compete with regional fixed-wing aircraft. These models were flown along virtual trajectories representing possible use-cases and assessed for environmental performance in comparison to an in-service baseline aircraft using comparable engine technology levels. Further, the travel time and mobility improvement available to the intermodal transport network through the inclusion of these concept rotorcraft was examined with promising results. Future work is suggested to address the shortfall in environmental performance

    Avian Primordial Germ Cells Contribute to and Interact With the Extracellular Matrix During Early Migration

    During early avian development, primordial germ cells (PGC) are highly migratory, moving from the central area pellucida of the blastoderm to the anterior extra-embryonic germinal crescent. The PGCs soon move into the forming blood vessels by intravasation and travel in the circulatory system to the genital ridges where they participate in the organogenesis of the gonads. This complex cellular migration takes place in close association with a nascent extracellular matrix that matures in a precise spatio-temporal pattern. We first compiled a list of quail matrisome genes by bioinformatic screening of human matrisome orthologs. Next, we used single cell RNA-seq analysis (scRNAseq) to determine that PGCs express numerous ECM and ECM-associated genes in early embryos. The expression of select ECM transcripts and proteins in PGCs were verified by fluorescent in situ hybridization (FISH) and immunofluorescence (IF). Live imaging of transgenic quail embryos injected with fluorescent antibodies against fibronectin and laminin, showed that germinal crescent PGCs display rapid shape changes and morphological properties such as blebbing and filopodia while surrounded by, or in close contact with, an ECM fibril meshwork that is itself in constant motion. Injection of anti-β1 integrin CSAT antibodies resulted in a reduction of mature fibronectin and laminin fibril meshwork in the germinal crescent at HH4-5 but did not alter the active motility of the PGCs or their ability to populate the germinal crescent. These results suggest that integrin β1 receptors are important, but not required, for PGCs to successfully migrate during embryonic development, but instead play a vital role in ECM fibrillogenesis and assembly

    Wallerian-Like Degeneration of Central Neurons After Synchronized and Geometrically Registered Mass Axotomy in a Three-Compartmental Microfluidic Chip

    Degeneration of central axons may occur following injury or due to various diseases and it involves complex molecular mechanisms that need to be elucidated. Existing in vitro axotomy models are difficult to perform, and they provide limited information on the localization of events along the axon. We present here a novel experimental model system, based on microfluidic isolation, which consists of three distinct compartments, interconnected by parallel microchannels allowing axon outgrowth. Neurons cultured in one compartment successfully elongated their axons to cross a short central compartment and invade the outermost compartment. This design provides an interesting model system for studying axonal degeneration and death mechanisms, with a previously impossible spatial and temporal control on specific molecular pathways. We provide a proof-of-concept of the system by reporting its application to a well-characterized experimental paradigm, axotomy-induced Wallerian degeneration in primary central neurons. Using this model, we applied localized central axotomy by a brief, isolated flux of detergent. We report that mouse embryonic cortical neurons exhibit rapid Wallerian-like distal degeneration but no somatic death following central axotomy. Distal axons show progressive degeneration leading to axonal beading and cytoskeletal fragmentation within a few hours after axotomy. Degeneration is asynchronous, reminiscent of in vivo Wallerian degeneration. Axonal cytoskeletal fragmentation is significantly delayed with nicotinamide adenine dinucleotide pretreatment, but it does not change when distal calpain or caspase activity is inhibited. These findings, consistent with previous experiments in vivo, confirm the power and biological relevance of this microfluidic architecture

    A selecção de um modelo que se ajuste adequadamente a um conjunto de dados é uma parte fulcral no processo da inferência estatística. O critério mais comum para seleccionar uma função de detectabilidade, que se ajuste a um conjunto de dados de distâncias, denomina-se critério de informação de Akaike (AIC). Para avaliar a performance da rotina da selecção de modelos implementada no programa DISTANCE (Thomas et al., 2006) realizaram-se simulações Monte Carlo para gerar dados de duas funções densidade de probabilidade frequentemente usadas. Aplicando esta metodologia, observou-se um enviesamento substancial na selecção do modelo, sendo os estimadores dos parâmetros baseados no modelo seleccionado frequentemente enviesados. Utilizaram-se duas abordagens na estimação dos parâmetros: selecção de modelos tradicional (ST) e ponderação de modelos (PM).     Decrease in Cell Volume Generates Contractile Forces Driving Dorsal Closure

    SummaryBiological tissues must generate forces to shape organs and achieve proper development. Such forces often result from the contraction of an apical acto-myosin meshwork. Here we describe an alternative mechanism for tissue contraction, based on individual cell volume change. We show that during Drosophila dorsal closure (DC), a wound healing-related process, the contraction of the amnioserosa (AS) is associated with a major reduction of the volume of its cells, triggered by caspase activation at the onset of the apoptotic program of AS cells. Cell volume decrease results in a contractile force that promotes tissue shrinkage. Estimating mechanical tensions with laser dissection and using 3D biophysical modeling, we show that the cell volume decrease acts together with the contraction of the actin cable surrounding the tissue to govern DC kinetics. Our study identifies a mechanism by which tissues generate forces and movements by modulating individual cell volume during development

    Fertility preservation after fertility-sparing surgery in women with borderline ovarian tumours

    International audienceObjective: To present a case series of women with borderline ovarian tumours (BOTs) who underwent oocyte vitrification in addition to fertility-sparing surgery.Study design: Observational study of all women referred to a French fertility preservation unit between 2015 and 2019 for counselling regarding a fertility preservation (FP) strategy after BOT fertility-sparing surgery. All eligible women underwent one or more cycles of controlled ovarian stimulation (COS) using an antagonist protocol, followed by oocyte retrieval. Metaphase II (MII) oocytes were vitrified.Results: Twenty-five women with BOTs were referred during the study period. Among them, 11 women underwent at least one cycle of COS. One hundred and seven MII oocytes were vitrified. The mean number of vitrified MII oocytes per woman was 9.7 (standard deviation 5.2). Five live births were reported during follow-up of four women with vitrified oocytes: three spontaneous pregnancies, one in-vitro fertilization cycle with fresh embryo transfer, and one live birth after return of vitrified oocytes.Conclusion: Conservative surgery for BOTs offers a high spontaneous pregnancy rate but has a higher risk of relapse than radical treatment. Furthermore, women who undergo conservative BOT surgery have a higher risk of surgery-induced premature ovarian failure. Oocyte cryopreservation after COS appears to be an effective technique after the conservative management of BOTs in women of reproductive age. Although the available short-term data are reassuring, further long-term studies evaluating the safety and cost-effectiveness of this systematic FP strategy after BOT fertility-sparing surgery are required
