    In vitro and in vivo evaluation of different measures to control Ascochyta blight in chickpea

    Ascochyta blight, an infection caused by Ascochyta rabiei is a destructive disease in many chickpea growing regions and it caused significant yield losses. To minimize the impact of Ascochyta blight, 5 fungicides viz., Aliette, Cabrio Top, Thiovit Jet, Cymoxanil and Difenoconazole, 5 plants extracts namely Azadirachta indica, Azadirachta azedarach, Datura stramonium, Chenopodium album and Allium sativum L. and two strains T-22 and E58 of bio-control agents (BCAs) Trichoderma viride and Aspergillus flavus were evaluated on the growth of A. rabiei under in vitro conditions by using the food poison technique. The colony growth of Ascochyta rabiei was inhibited at all concentrations of fungicides @ 0.07, 0.15, 0.21%, plants extracts @ 4, 6, 9% and bio-control agents @ 105, 106 and 107 conidia ml-1 respectively. Among all applied treatments, maximum inhibition colony growth of pathogen was recorded in the case of Aliette (83.4%), followed by Cabrio Top (74.3%), Azadirachta indica (50.3%) and Trichoderma viride (60.3%) at their high concentrations. Field trials showed that Aliette and Cabario Top significantly reduced the disease severity to 10 % and 24% respectively, followed by Azadirachta indica and Allium sativum which reduced the disease severity to 40% and 50% respectively. Bio-control agent Trichoderma viride proved less effective in controlling Ascochyta bight severity under field conditions. The present study showed that systemic and sulphur containing fungicides, plant extracts and bio-control agents (BCAs) have the potential to control Ascochyta blight in both in vitro and in vivo conditions

    Determination of Crop Coefficient of Hybrid Wheat under Arid Climate: A Pot Study

    Climate change increases vulnerabilities for crop productivity in Pakistan. Water crises are increasing with an increase in temperature and change in precipitation patterns due to climate change which ultimately imposed a threat to the food security of the country. Water is indispensable for all plants to complete life cycle as the unavailability of water at critical growth stages drastically affects the development of the plant. The present pot study was conducted for the estimation of crop coefficient of hybrid wheat for irrigation scheduling at Muhammad Nawaz Shareef University of Agriculture, Multan during two growing seasons 2018-19 and 2019-20. In this experiment, three wheat varieties were used were Hybrid-1 (R26-3-1/DH-16), Hybrid-2(AR 7-5 / ZWB-14), and Galaxy-2013 as treatment. The soil moisture content was maintained between 50 to 100 % available water content (AWC) during both growing seasons. The crop coefficient (Kc) and actual evapotranspiration (Eta) were maximum in galaxy-13 and minimum in hybrid wheat. The grain yield for Hybrid-1, Hybrid-2, and galaxy-13 was 1, 1.5, and 0.6 g plant-1, respectively while the straw output was 4.8, 4.3, and 3 g plant-1, respectively. The harvest index for Hybrid-1, Hybrid-2, and galaxy-13 were 20, 34, and 20% respectively. The water use efficiency (WUE) for Hybrid-1, Hybrid-2, and galaxy-13 was 0.2. 0.3 and 0.1 g plant-1mm-1, respectively. The Hybrid-1 and Hybrid-2 produced more grain yield, straw yield, more spikes, and more grains per spikes and showed more water use efficiency with short plant height as compared to galaxy-13. The results of the study revealed that Hybrid-2 is more water-efficient with low water requirement and it was followed by Hybrid-1. The growing of Hybrid-2 will enhance the wheat yield to meet the food requirements of the increasing population under the climate change scenario with less water

    Fabrication and Characterization of Sulfonated Graphene Oxide (SGO) Doped PVDF Nanocomposite Membranes with Improved Anti-Biofouling Performance

    Emergence of membrane technology for effective performance is qualified due to its low energy consumption, no use of chemicals, high removal capacity and easy accessibility of membrane material. The hydrophobic nature of polymeric membranes limits their applications due to biofouling (assemblage of microorganisms on surface of membrane). Polymeric nanocomposite membranes emerge to alleviate this issue. The current research work was concerned with the fabrication of sulfonated graphene oxide doped polyvinylidene fluoride (PVDF) membrane and investigation of its anti-biofouling and anti-bacterial behavior. The membrane was fabricated through phase inversion method, and its structure and morphology were characterized by Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-rays diffraction (XRD) and thermo gravimetric analysis (TGA) techniques. Performance of the membrane was evaluated via pure water flux; anti-biofouling behavior was determined through Bovine Serum albumin (BSA) rejection. Our results revealed that the highest water flux was shown by M7 membrane about 308.7 Lm−2h−1/bar having (0.5%) concentration of SGO with improved BSA rejection. Furthermore, these fabricated membranes showed high antibacterial activity, more hydrophilicity and mechanical strength as compared to pristine PVDF membranes. It was concluded that SGO addition within PVDF polymer matrix enhanced the properties and performance of membranes. Therefore, SGO was found to be a promising material for the fabrication of nanocomposite membranes

    Adaptation of the Wound Healing Questionnaire universal-reporter outcome measure for use in global surgery trials (TALON-1 study): mixed-methods study and Rasch analysis

    BackgroundThe Bluebelle Wound Healing Questionnaire (WHQ) is a universal-reporter outcome measure developed in the UK for remote detection of surgical-site infection after abdominal surgery. This study aimed to explore cross-cultural equivalence, acceptability, and content validity of the WHQ for use across low- and middle-income countries, and to make recommendations for its adaptation.MethodsThis was a mixed-methods study within a trial (SWAT) embedded in an international randomized trial, conducted according to best practice guidelines, and co-produced with community and patient partners (TALON-1). Structured interviews and focus groups were used to gather data regarding cross-cultural, cross-contextual equivalence of the individual items and scale, and conduct a translatability assessment. Translation was completed into five languages in accordance with Mapi recommendations. Next, data from a prospective cohort (SWAT) were interpreted using Rasch analysis to explore scaling and measurement properties of the WHQ. Finally, qualitative and quantitative data were triangulated using a modified, exploratory, instrumental design model.ResultsIn the qualitative phase, 10 structured interviews and six focus groups took place with a total of 47 investigators across six countries. Themes related to comprehension, response mapping, retrieval, and judgement were identified with rich cross-cultural insights. In the quantitative phase, an exploratory Rasch model was fitted to data from 537 patients (369 excluding extremes). Owing to the number of extreme (floor) values, the overall level of power was low. The single WHQ scale satisfied tests of unidimensionality, indicating validity of the ordinal total WHQ score. There was significant overall model misfit of five items (5, 9, 14, 15, 16) and local dependency in 11 item pairs. The person separation index was estimated as 0.48 suggesting weak discrimination between classes, whereas Cronbach's α was high at 0.86. Triangulation of qualitative data with the Rasch analysis supported recommendations for cross-cultural adaptation of the WHQ items 1 (redness), 3 (clear fluid), 7 (deep wound opening), 10 (pain), 11 (fever), 15 (antibiotics), 16 (debridement), 18 (drainage), and 19 (reoperation). Changes to three item response categories (1, not at all; 2, a little; 3, a lot) were adopted for symptom items 1 to 10, and two categories (0, no; 1, yes) for item 11 (fever).ConclusionThis study made recommendations for cross-cultural adaptation of the WHQ for use in global surgical research and practice, using co-produced mixed-methods data from three continents. Translations are now available for implementation into remote wound assessment pathways
