Genetic Diversity of Bananas (Musa spp.) in Pesawaran District, Lampung Province Based on Sequence Related Amplified Polymorphism (SRAP) Marker
Banana (Musa spp.) is a leading commodity that makes a major contribution to national fruit production. One of the districts in Lampung Province which has a significant contribution to Indonesia's banana production is Pesawaran Regency. The molecular marker used in this study is Sequence-Related Amplified Polymorphism (SRAP). This study aims to determine the genetic diversity of bananas, determine the suitable combination of SRAP primers to determine the genetic diversity of bananas and determine the similarity relationship of bananas in the Pesawaran District. The total genomic DNA was extracted using DNeasy Plant Mini Kit (Geneaid) according to the protocol from the manufacturer followed by their amplification using SRAP primers. Similarity analysis was carried out based on similarity index values and dendrogram construction using the NTSYS 2.0 program. Research results show that a total of 204 loci (167 polymorphic loci and 37 monomorphic loci) were obtained from 9 selected primer combinations. The genetic diversity of 19 banana cultivars originating from Pesawaran Regency resulted in low genetic diversity values (0-0.27). Banana kinship based on Sequence Related Amplified Polymorphism (SRAP) markers divided 19 banana cultivar samples and 4 wild banana samples into 2 large clusters with similarity coefficient values between 0.70-0.97
ISOLATION AND CLONING OF cDNA OF GENE ENCODING FOR METALLOTHIONEIN TYPE 2 FROM MELASTOMA AFFINE
Metallothionein is an important protein for detoxifying heavy metal ions. This research was conducted to isolate and clone cDNA of gene encoding for metallothionein type 2 from Melastoma affine. Total RNA was isolated from young leaves. Total cDNA was synthesized from the total RNA by reverse transcription. The MaMt2 cDNA was successfully isolated by PCR technique. The MaMt2 cDNA was inserted into pGEM-T Easy and the recombinant plasmid was successfully introduced into Escherichia coli DH5α. DNA sequencing analysis showed that this cDNA is full length consisting of 246 pb encoding 81 amino acid residues. This cDNA is identical to mRNA of AtMt2 from Arabidopsis thaliana. It does not contain any restriction sites found in the cloning sites of pGEM-T easy. The deduced protein of MaMT2 contains 14 cysteine residues distributed in the Cys-Cys, Cys-X-Cys, and Cys-X-X-Cys motifs.  Key words: cDNA, metallothionein, Melastoma affine, cloning, cystein
Typification and taxonomic notes of Smilacaceae species in Java
Smilacaceae is known as a taxa with wide phenotypic variation and their taxonomical complexities remain unsolved. The three species of Smilacaceae housed in Java, are given nomenclature history. Since all the potential sources of original material have been investigated, but nothing has been identified, three neotypifications were designated here. Herbarium specimen of Koorders 34990β in Herbarium Bogoriense (BO) was chosen as the neotype of Smilax klotzschii. The de Groot & Wehlburg RD52 herbarium specimen in BO was chosen as the neotype of S. nageliana and Blume 463 herbarium specimen in L was chosen as the neotype of S. odoratissima
KERAGAMAN GENETIK Tacca leontopetaloides (L.) Kuntze (Taccaceae) DARI BEBERAPA PROVENANSI DI INDONESIA BERDASARKAN MARKA INTER SIMPLE SEQUENCE REPEATS (ISSR)
Kecondang or Taka (Tacca leontopetaloides), a member of Taccaceae, is one the carbohydrate alternatives to rice which can be developed in coastal area. With the global warming, the sea level has increased and small islands are more isolated. The people who lives in these islands should depend on their bioresources.As Taka inhabits coastal area, the development of this plant becomes critical.A total of 65 accessions of Taka from 9 provenances in Indonesia (Sukabumi, Madura, Kp.Siung (Yogyakarta), Gn. Kidung (Yogyakarta), and the small islands in Karimun Jawa namely Nyamuk, Katang, Cendekia, Kumbang, Seruni islands) were characterized using six ISSR markers [GSGG (TG5)T; CCAG(TGG)3TG; GGAG(TGG)3TG; BDBT(TCC4); (AC)8C; (AG)8G]. The scoring of the fragments resulted in 55 (69.6%) polymorphic bands. The size of the bands varied from 150bp to 1,6kb. The 65 accessions of Taka made several clusters according to their provenances. The range of individual genetic dissimilarity was from 0.012 to 0.186. The results showed that amongst nine population studied, Gn. Kidul, Yogyakarta population has the highest level of genetic variation with mean values of Na = 1.3544±0.4814, Ne =1.1784±0.3025, PLP = 35.44% and He = 0.1094±0.1681. Whereas, P. Nyamuk, Kep. Karimun Jawa population has the lowest level of genetic variation with mean values of Na = 1.0759±0.2666, Ne = 1.0523±0.2163, PLP = 7.59% and He = 0.0275±0.1097
The Diversity of Wild Banana Species (Genus Musa) in Java
The diversity of wild banana species (genus Musa, listed in Flora of Java) has been revised. The present taxonomic study is based on morphological characteristics observed in the herbarium specimens deposited at the Herbarium Bogoriense (BO), living collections in the Bogor Botanical Garden, the Cibodas Botanical Garden, and during the explorations done at Mt. Salak, West Java. Eight species of Musa (Musa acuminata, M. balbisiana, M. coccinea, M. ornata, M. salaccensis, M. sanguinea, M. textilis and M. velutina) and seven infraspecific taxa of M. acuminata are recognized in Java, of which two infraspecific taxa are endemic. West Java is the center of distribution for the wild banana species in Java. Taxonomic descriptions including an identification key are presented

Reinwardtia 16 (1): 19 - 24. - A new variety of Musa borneensis, M. borneensis var. donggalaensis Sulis. is proposed based upon specimens from Donggala, Central Sulawesi, Indonesia. Endemic status of M. borneensis was rejected. The description, distribution map and the identification key are provided.
