    Eficiência da bioadição na remoção de matéria orgânica em sistemas aquaculturais

    Several techniques are currently used to treat effluents. Bioaugmentation is a new bioremediation strategy and has been employed to improve effluent quality by treating the water during the production process. This technology consists basically of the addition of microorganisms able to degrade or remove polluting compounds, especially organic matter and nutrients. The objective of this study was to assess the effects of bioaugmentation on some parameters of organic matter and on the performance of juvenile tilapias in an intensive aquaculture production system. The combination of two bacterial consortiums in a complete randomized design was employed in a factorial analysis with two factors. Statistical differences between treatments were analyzed by the analysis of variance (ANOVA) and Tukey test at the 5% level. One of the treatments, heterotrophic bacterial supplementation, was able to reduce biochemical oxygen demand (BOD) by 23%, dissolved organic carbon (DOC) by 83.7% and phytoplanktonic biomass by 43%. On the other hand, no damage was done to either the physical-chemical indicators of water quality or to the growth performance of juvenile tilapias assessed in this study.Existem diversas tecnologias para tratamento de efluentes, o processo de bioadição consiste em uma vertente da biorremediação e tem sido empregado na melhoria da qualidade dos efluentes através do tratamento da água de produção. Esta tecnologia consiste basicamente na adição de microrganismos com a capacidade de degradar ou remover compostos poluentes, especialmente matéria orgânica e nutrientes. Este estudo objetivou avaliar os efeitos da suplementação de composto bioativo sobre alguns parâmetros de matéria orgânica e de desempenho de juvenis de tilápias em um sistema intensivo de produção aquacultural. Foi empregada a combinação de dois consórcios bacterianos em delineamento inteiramente aleatorizado, em um esquema fatorial com dois fatores. As diferenças estatísticas entre os tratamentos foram analisadas por meio da análise de variância (ANOVA) e do teste de Tukey ao nível de 5%. Verificou-se neste estudo, que a bioadição heterotrófica foi capaz de reduzir em 23% a demanda bioquímica de oxigênio (DBO); em 83,7%, o carbono orgânico dissolvido (COD); e em 43%, a biomassa fitoplanctônica.     Content of Lipids, Fatty Acids, Carbohydrates, and Proteins in Continental Cyanobacteria: A Systematic Analysis and Database Application

    The lipid, fatty acid, protein, and carbohydrate contents in cyanobacterial strains and biomass can vary by orders of magnitude. Many publications (thousands of peer-reviewed articles) require more work to extract their precise concentration values (i.e., different units, inaccurate data), which makes them not easily exploitable. For this purpose, tables have been compiled from the literature data, including lipids, fatty acids, proteins, and carbohydrates composition and quantities in cyanobacteria. A lot of data (323) were collected after careful a literature search, according to selected criteria in order to distinguish separately cyanobacteria, and according to categories of genus and species and generate average values of the contents of these cell components. These data are exploited in a first systematic analysis of the content in types of strains. Our database can be a powerful tool for biologists, chemists, and environmental agencies to determine the potential concentration of high-value chemical building blocks directly from low-value bloom biomass, cell cultures, or debris in the sediment, offering the potential to minimize environmental waste and add value to the agro-industrial residues. The database can also support strategies for food manufacturers to develop new products with optimized properties for veterinarian applications

    Wide spectrum of NR5A1-related phenotypes in 46,XY and 46,XX individuals

    Steroidogenic factor 1 (NR5A1, SF-1, Ad4BP) is a transcriptional regulator of genes involved in adrenal and gonadal development and function. Mutations in NR5A1 have been among the most frequently identified genetic causes of gonadal development disorders and are associated with a wide phenotypic spectrum. In 46,XY individuals, NR5A1-related phenotypes may range from disorders of sex development (DSD) to oligo/azoospermia, and in 46,XX individuals, from 46,XX ovotesticular and testicular DSD to primary ovarian insufficiency (POI). The most common 46,XY phenotype is atypical or female external genitalia with clitoromegaly, palpable gonads, and absence of Müllerian derivatives. Notably, an undervirilized external genitalia is frequently seen at birth, while spontaneous virilization may occur later, at puberty. In 46,XX individuals, NR5A1 mutations are a rare genetic cause of POI, manifesting as primary or secondary amenorrhea, infertility, hypoestrogenism, and elevated gonadotropin levels. Mothers and sisters of 46,XY DSD patients carrying heterozygous NR5A1 mutations may develop POI, and therefore require appropriate counseling. Moreover, the recurrent heterozygous p.Arg92Trp NR5A1 mutation is associated with variable degrees of testis development in 46,XX patients. A clear genotype-phenotype correlation is not seen in patients bearing NR5A1 mutations, suggesting that genetic modifiers, such as pathogenic variants in other testis/ovarian-determining genes, may contribute to the phenotypic expression. Here, we review the published literature on NR5A1-related disease, and discuss our findings at a single tertiary center in Brazil, including ten novel NR5A1 mutations identified in 46,XY DSD patients. The ever-expanding phenotypic range associated with NR5A1 variants in XY and XX individuals confirms its pivotal role in reproductive biology, and should alert clinicians to the possibility of NR5A1 defects in a variety of phenotypes presenting with gonadal dysfunction

    Development and validation of a questionnaire to identify severe maternal morbidity in epidemiological surveys

    <p>Abstract</p> <p>Objective</p> <p>to develop and validate a questionnaire on severe maternal morbidity and to evaluate the maternal recall of complications related to pregnancy and childbirth. <it>Design: </it>validity of a questionnaire as diagnostic instrument. <it>Setting: </it>a third level referral maternity in Campinas, Brazil. <it>Population: </it>386 survivors of severe maternal complications and 123 women that delivered without major complications between 2002 and 2007.</p> <p>Methods</p> <p>eligible women were traced and interviewed by telephone on the occurrence of obstetric complications and events related to their treatment. Their answers were compared with their medical records as gold standard. Sensitivity, specificity and likelihood ratios plus their correspondent 95% confidence intervals were used as main estimators of accuracy. <it>Main outcomes: </it>diagnosis of severe maternal morbidity associated with past pregnancies, including hemorrhage, eclampsia, infections, jaundice and related procedures (hysterectomy, admission to ICU, blood transfusion, laparotomy, inter-hospital transfer, mechanical ventilation and post partum stay above seven days).</p> <p>Results</p> <p>Women did not recall accurately the occurrence of obstetric complications, especially hemorrhage and infection. The likelihood ratios were < 5 for hemorrhage and infection, while for eclampsia it almost reached 10. The information recalled by women regarding hysterectomy, intensive care unit admission and blood transfusion were found to be highly correlated with finding evidence of the event in the medical records (likelihood ratios ranging from 12.7-240). The higher length of time between delivery and interview was associated with poor recall.</p> <p>Conclusion</p> <p>Process indicators are better recalled by women than obstetric complication and should be considered when applying a questionnaire on severe maternal morbidity.</p

    From Classical Genetics to Quantitative Genetics to Systems Biology: Modeling Epistasis

    Gene expression data has been used in lieu of phenotype in both classical and quantitative genetic settings. These two disciplines have separate approaches to measuring and interpreting epistasis, which is the interaction between alleles at different loci. We propose a framework for estimating and interpreting epistasis from a classical experiment that combines the strengths of each approach. A regression analysis step accommodates the quantitative nature of expression measurements by estimating the effect of gene deletions plus any interaction. Effects are selected by significance such that a reduced model describes each expression trait. We show how the resulting models correspond to specific hierarchical relationships between two regulator genes and a target gene. These relationships are the basic units of genetic pathways and genomic system diagrams. Our approach can be extended to analyze data from a variety of experiments, multiple loci, and multiple environments

A well known but rare toxicity are hypersensitivity reactions, usually manifested by chest discomfort, dyspnoea, bronchospasm and hypotension. We report the details of a patient who developed hypersensitivity reactions to intravenous etoposide, but subsequently tolerated the administration of intravenous etoposide phosphate with no sequalae
