    Cryptotephra as a dating and correlation tool in archaeology

    A new development in archaeological chronology involves the use of far travelled volcanic ash which may form discrete but invisible layers within a site's stratigraphy. Known as cryptotephra, these horizons can provide isochrons for the precise correlation of archaeological records at single moments in time, removing, or at least significantly reducing, temporal uncertainty within inter-site comparisons. When a tephra can be dated elsewhere, its age can be imported between records, providing an independent check on other dating methods in use and valuable age estimates for difficult to date sequences. The use of cryptotephra layers to date and correlate palaeoenvironmental archives is well established and there exists a wealth of tephra compositional data and regional tephrostratigraphic frameworks from which archaeological cryptotephra studies can benefit greatly. Existing approaches to finding and analysing cryptotephra are easily adapted to archaeological sequences, so long as the often complex nature of archaeological stratigraphies and sediment taphonomy are borne in mind. © 2013 Elsevier Ltd

    The Late Quaternary tephrostratigraphy of annually laminated sediments from Meerfelder Maar, Germany

    © 2015 Elsevier Ltd.The record of Late Quaternary environmental change within the sediments of Meerfelder Maar in the Eifel region of Germany is renowned for its high precision chronology, which is annually laminated throughout the Last Glacial to Interglacial transition (LGIT) and most of the Holocene. Two visible tephra layers are prominent within the floating varve chronology of Meerfelder Maar. An Early Holocene tephra layer, the Ulmener Maar Tephra (~11,000 varve years BP), provides a tie-line of the Meerfelder Maar record to the varved Holocene record of nearby Lake Holzmaar. The Laacher See Tephra provides another prominent time marker for the late AllerÞd, ~200 varve years before the transition into the Younger Dryas at 12,680 varve years BP. Further investigation has now shown that there are also 15 cryptotephra layers within the Meerfelder Maar LGIT-Holocene stratigraphy and these layers hold the potential to make direct comparisons between the Meerfelder Maar record and other palaeoenvironmental archives from across Europe and the North Atlantic. Most notable is the presence of the Vedde Ash, the most widespread Icelandic eruption known from the Late Quaternary, which occurred midway through the Younger Dryas. The Vedde Ash has also been found in the Greenland ice cores and can be used as an isochron around which the GICC05 and Meerfelder Maar annual chronologies can be compared. Near the base of the annual laminations in Meerfelder Maar a cryptotephra is found that correlates to the Neapolitan Yellow Tuff, erupted from Campi Flegrei in southern Italy, 1200km away. This is the furthest north that the Neapolitan Yellow Tuff has been found, highlighting its importance in the construction of a European-wide tephrostratigraphic framework. The co-location of cryptotephra layers from Italian, Icelandic and Eifel volcanic sources, within such a precise chronological record, makes Meerfelder Maar one of the most important tephrostratotype records for continental Europe during the Last Glacial to Interglacial transition

    Resistance exercise and whey protein supplementation are effective strategies to activate muscle cell anabolic signaling and ultimately promote increases in muscle mass and strength. In the current study, 46 healthy older men aged 60–75 (69.0 ± 0.55 years, 85.9 ± 1.8 kg, 176.8 ± 1.0 cm) performed a single bout of unaccustomed lower body resistance exercise immediately followed by ingestion of a noncaloric placebo beverage or supplement containing 10, 20, 30, or 40 g of whey protein concentrate (WPC). Intramuscular amino acid levels in muscle biopsy samples were measured by Gas Chromatography–Mass Spectrometry (GC-MS) at baseline (before exercise and WPC supplementation) plus at 2 h and 4 h post exercise. Additionally, the extent of p70S6K phosphorylation at Thr389 in muscle biopsy homogenates was assessed by western blot. Resistance exercise alone reduced intramuscular branch chain amino acid (BCAA; leucine, isoleucine, and valine) content. Supplementation with increasing doses of whey protein prevented this fall in muscle BCAAs during postexercise recovery and larger doses (30 g and 40 g) significantly augmented postexercise muscle BCAA content above that observed following placebo ingestion. Additionally, the fold change in the phosphorylation of p70S6K (Thr389) at 2 h post exercise was correlated with the dose of whey protein consumed (r = 0.51, P < 001) and was found to be significantly correlated with intramuscular leucine content (r = 0.32, P = 0.026). Intramuscular BCAAs, and leucine in particular, appear to be important regulators of anabolic signaling in aged human muscle during postexercise recovery via reversal of exercise-induced declines in intramuscular BCAAs

    Effects of donor/recipient human leukocyte antigen mismatch on human cytomegalovirus replication following liver transplantation.

    Background Natural immunity against cytomegalovirus (CMV) can control virus replication after solid organ transplantation; however, it is not known which components of the adaptive immune system mediate this protection. We investigated whether this protection requires human leukocyte antigen (HLA) matching between donor and recipient by exploiting the fact that, unlike transplantation of other solid organs, liver transplantation does not require HLA matching, but some donor and recipient pairs may nevertheless be matched by chance. Methods To further investigate this immune control, we determined whether chance HLA matching between donor (D) and recipient (R) in liver transplants affected a range of viral replication parameters. Results In total, 274 liver transplant recipients were stratified according to matches at the HLA A, HLA B, and HLA DR loci. The incidence of CMV viremia, kinetics of replication, and peak viral load were similar between the HLA matched and mismatched patients in the D+/R+ and D−/R+ transplant groups. D+/R− transplants with 1 or 2 mismatches at the HLA DR locus had a higher incidence of CMV viremia >3000 genomes/mL blood compared to patients matched at this locus (78% vs. 17%; P = 0.01). Evidence was seen that matching at the HLA A locus had a small effect on peak viral loads in D+/R− patients, with median peak loads of 3540 and 14,706 genomes/mL in the 0 and combined (1 and 2) mismatch groups, respectively (P = 0.03). Conclusion Overall, our data indicate that, in the setting of liver transplantation, prevention of CMV infection and control of CMV replication by adaptive immunity is minimally influenced by HLA matching of the donor and recipient. Our data raise questions about immune control of CMV in the liver and also about the cells in which the virus is amplified to give rise to CMV viremia

    Layers of far-travelled volcanic ash (tephra) from explosive volcanic eruptions provide stratigraphic and numerical dating horizons in sedimentary and volcanic sequences. Such tephra layers may be dispersed over tens to thousands of kilometres from source, reaching far beyond individual volcanic regions. Tephrochronology is consequently a truly global dating tool, with applications increasingly widespread across a range of Quaternary and geoscience disciplines. This special issue of the International Focus Group on Tephrochronology and Volcanism (INTAV) showcases some of the many recent advances in tephrochronology, from methodological developments to diverse applications across volcanological, archaeological, and palaeoclimatological research

    X-linked incomplete achromatopsia with more than one class of functional cones

    Five affected males in the fifth generation of a large pedigree of X-chromosomal incomplete achromatopsia were tested. All had SWS cone function. A 19-year-old affected man was a classical blue cone monochromat on color matching and spectral sensitivity. A 16-year-old boy showed evidence of a long wavelength sensitive cone active in 8 degrees color matches. With a blue-green background, his cone spectral sensitivity function peaked near 550-560 nm. Three younger boys, aged 7-10 yrs were evaluated only with color matching. All showed evidence of long wavelength cone function with an 8 degree field and one showed long wavelength cones in 2 degree matches. An independent observation concerning the family was the finding that deuteranomaly was introduced in the third generation. The fourth generation women, all obligate carriers of X-linked achromatopsia, had a 0.5 chance to carry deuteranomaly. Neither carrier state per se is usually associated with expression of deuteranomaly. Three of the five tested expressed deuteranomaly. This finding of deuteranomaly in the carrier females might be a consequence of a double carrier state indicating association between the genes for deuteranomaly and X-linked achromatopsia

    The role of cryptotephra in refining the chronology of Late Pleistocene human evolution and cultural change in North Africa

    © 2014.Sites in North Africa hold key information for dating the presence of Homo sapiens and the distribution of Middle Stone Age (MSA), Middle Palaeolithic (MP) and Later Stone Age (LSA) cultural activity in the Late Pleistocene. Here we present new and review recently published tephrochronological evidence for five cave sites in North Africa with long MSA/MP and LSA cultural sequences. Four tephra horizons have been identified at the Haua Fteah (Cyrenaica, Libya). They include cryptotephra evidence for the Campanian Ignimbrite (CI) eruption dating to ~39 ka that allows correlation with other Palaeolithic sequences in the eastern Mediterranean and as far north as Russia. Cryptotephra have also been recorded from the Moroccan sites of Taforalt, Rhafas and Dar es-Soltane 1. At Taforalt the geochemical composition suggests a provenance in the Azores, while examples from Sodmein (Egypt) appear to derive from central Anatolia and another unknown source. In these latter examples chemical compositional data from relevant proximal volcanic centres is currently lacking so the identification of tephra in layers of known age and cultural association provides the first reliable age determinations for distal volcanic events and their geographical extent. The future potential for tephrochronological research in North Africa is also discussed

    Contamination of clinical specimens with MLV-encoding nucleic acids: implications for XMRV and other candidate human retroviruses

    Efforts to assess the prevalence of xenotropic murine leukemia virus-related virus (XMRV) in patients with prostate cancer and chronic fatigue syndrome have relied heavily on PCR-based testing of clinical samples and have yielded widely divergent findings. This week in Retrovirology, reports from four independent research groups illustrate the extreme care needed to exclude DNA or RNA contamination in PCR analyses of XMRV. In addition, phylogenetic evidence suggesting that previously-published XMRV sequences originated from a commonly-used prostate carcinoma cell line (22Rv1) is presented. These findings raise important questions regarding the provenance of XMRV and its potential connection to human disease
