    A primitive equation barotropic stability analysis of the 700 mb zonal wind profile over the Arabian Sea was performed for each day between 10 and 14 June 1979, during the onset phase of the monsoon. The flow was found to become progessively more unstable with the passage of time. The most unstable inviscid primitive equation mode has an e-folding time of 3.1 days, a wavelength of 3500 km and a westward phase speed of 1.9 m s-1

    An Alternative Method to Niskin Sampling for Molecular Analysis of the Marine Environment

    The development of low-cost, open-source Remotely Operated Vehicle (ROV) systems has provided almost unrestricted access for researchers looking to monitor the marine environment in ever greater resolution. Sampling microbial communities from the marine environment, however, still usually relies on Niskin-bottle sampling (ROV or Conductivity-Temperature-Depth sampler (CTD) based), a method which introduces an inaccuracy and variability that is incompatible with metatranscriptomic analysis, for example. Here, we describe a versatile, easily-replicated platform which achieves in situ mRNA preservation, via the addition of RNAlater to filtered microbial cells, to enhance ROV or CTD functionality

    Copper signaling axis as a target for prostate cancer therapeutics.

    Previously published reports indicate that serum copper levels are elevated in patients with prostate cancer and that increased copper uptake can be used as a means to image prostate tumors. It is unclear, however, to what extent copper is required for prostate cancer cell function as we observed only modest effects of chelation strategies on the growth of these cells in vitro. With the goal of exploiting prostate cancer cell proclivity for copper uptake, we developed a "conditional lethal" screen to identify compounds whose cytotoxic actions were manifested in a copper-dependent manner. Emerging from this screen was a series of dithiocarbamates, which, when complexed with copper, induced reactive oxygen species-dependent apoptosis of malignant, but not normal, prostate cells. One of the dithiocarbamates identified, disulfiram (DSF), is an FDA-approved drug that has previously yielded disappointing results in clinical trials in patients with recurrent prostate cancer. Similarly, in our studies, DSF alone had a minimal effect on the growth of prostate cancer tumors when propagated as xenografts. However, when DSF was coadministered with copper, a very dramatic inhibition of tumor growth in models of hormone-sensitive and of castrate-resistant disease was observed. Furthermore, we determined that prostate cancer cells express high levels of CTR1, the primary copper transporter, and additional chaperones that are required to maintain intracellular copper homeostasis. The expression levels of most of these proteins are increased further upon treatment of androgen receptor (AR)-positive prostate cancer cell lines with androgens. Not surprisingly, robust CTR1-dependent uptake of copper into prostate cancer cells was observed, an activity that was accentuated by activation of AR. Given these data linking AR to intracellular copper uptake, we believe that dithiocarbamate/copper complexes are likely to be effective for the treatment of patients with prostate cancer whose disease is resistant to classical androgen ablation therapies

    Skills of different mesoscale models over Indian region during monsoon season: Forecast errors

    Performance of four mesoscale models namely, the MM5, ETA, RSM and WRF, run at NCMRWF for short range weather forecasting has been examined during monsoon-2006. Evaluation is carried out based upon comparisons between observations and day-1 and day-3 forecasts of wind, temperature, speci.c humidity, geopotential height, rainfall, systematic errors, root mean square errors and specific events like the monsoon depressions. It is very difficult to address the question of which model performs best over the Indian region? An honest answer is 'none'. Perhaps an ensemble approach would be the best. However, if we must make a final verdict, it can be stated that in general, (i) the WRF is able to produce best All India rainfall prediction compared to observations in the day-1 forecast and, the MM5 is able to produce best All India rainfall forecasts in day-3, but ETA and RSM are able to depict the best distribution of rainfall maxima along the west coast of India, (ii) the MM5 is able to produce least RMSE of wind and geopotential fields at most of the time, and (iii) the RSM is able to produce least errors in the day-1 forecasts of the tracks, while the ETA model produces least errors in the day-3 forecasts

    A Catalog of Reference Genomes from the Human Microbiome

    The human microbiome refers to the community of microorganisms including prokaryotes, viruses and microbial eukaryotes that populate the human body. The National Institutes of Health launched an initiative that focuses describing the diversity of microbial species associated with health and disease. The first phase of this initiative includes the sequencing of hundreds of microbial reference genomes, coupled to metagenomic sequencing from multiple body sites. Here we present results from an initial reference genome sequencing of 178 microbial genomes. From 547,968 predicted polypeptides that correspond to the gene complement of these strains “novel” polypeptides that had both unmasked sequence length > 100 amino acids and no BLASTP match to any non-reference entry in the nr subset were defined. This analysis resulted in a set of 30,867 polypeptides, of which 29,987 (~97%) were unique. In addition, this set of microbial genomes allows for ~ 40% of random sequences from the microbiome of the gastrointestinal tract to be associated with organisms based on the match criteria used. Insights into pan-genome analysis suggest that we are still far from saturating microbial species genetic datasets.     Influence of Matrix Polarity on the Properties of Ethylene Vinyl Acetate–Carbon Nanofiller Nanocomposites

    A series of ethylene vinyl acetate (EVA) nanocomposites using four kinds of EVA with 40, 50, 60, and 70 wt% vinyl acetate (VA) contents and three different carbon-based nanofillers—expanded graphite (EG), multi-walled carbon nanotube (MWCNT), and carbon nanofiber (CNF) have been prepared via solution blending. The influence of the matrix polarity and the nature of nanofillers on the morphology and properties of EVA nanocomposites have been investigated. It is observed that the sample with lowest vinyl acetate content exhibits highest mechanical properties. However, the enhancement in mechanical properties with the incorporation of various nanofillers is the highest for EVA with high VA content. This trend has been followed in both dynamic mechanical properties and thermal conductivity of the nanocomposites. EVA copolymer undergoes a transition from partial to complete amorphousness between 40 and 50 wt% VA content, and this changes the dispersion of the nanofillers. The high VA-containing polymers show more affinity toward fillers due to the large free volume available and allow easy dispersion of nanofillers in the amorphous rubbery phase, as confirmed from the morphological studies. The thermal stability of the nanocomposites is also influenced by the type of nanofiller

    Collaborative Gaze Channelling for Improved Cooperation During Robotic Assisted Surgery

    The use of multiple robots for performing complex tasks is becoming a common practice for many robot applications. When different operators are involved, effective cooperation with anticipated manoeuvres is important for seamless, synergistic control of all the end-effectors. In this paper, the concept of Collaborative Gaze Channelling (CGC) is presented for improved control of surgical robots for a shared task. Through eye tracking, the fixations of each operator are monitored and presented in a shared surgical workspace. CGC permits remote or physically separated collaborators to share their intention by visualising the eye gaze of their counterparts, and thus recovers, to a certain extent, the information of mutual intent that we rely upon in a vis-à-vis working setting. In this study, the efficiency of surgical manipulation with and without CGC for controlling a pair of bimanual surgical robots is evaluated by analysing the level of coordination of two independent operators. Fitts' law is used to compare the quality of movement with or without CGC. A total of 40 subjects have been recruited for this study and the results show that the proposed CGC framework exhibits significant improvement (p<0.05) on all the motion indices used for quality assessment. This study demonstrates that visual guidance is an implicit yet effective way of communication during collaborative tasks for robotic surgery. Detailed experimental validation results demonstrate the potential clinical value of the proposed CGC framework. © 2012 Biomedical Engineering Society.link_to_subscribed_fulltex
