    THE TRAINING OF NURSES TO WORK IN PRIMARY CARE FOR THE PREVENTION OF BREAST CANCER

    Objetivo: é identificar como deve ser a prevenção do câncer de mama na atenção básica e descrever a formação profissional do enfermeiro para atuar na atenção básica prevenindo a neoplasia mamária. Métodos: estudo exploratório, desenvolvido através de uma revisão sistemática de literatura na BVS (SCIELO, LILACS e BDENF), com abordagem qualitativa. Realizou-se a pré-leitura, leitura seletiva, interpretativa e finalizou-se com a análise temática. Destaca-se que foram selecionados 9 (nove) estudos. Resultados: as categorias que emergiram foram: Prevenção do câncer de mama na rede básica de saúde e Formação Profissional de enfermagem com o foco na prevenção. Conclusão: conclui-se que a prevenção é a forma mais eficaz e barata de diminuir a incidência da doença e que a formação profissional deve ser valorizada na graduação tanto em conteúdos teóricos quanto práticos, para que o enfermeiro tenha uma visão integral do cuidado e saia do foco que ainda continua presente que é a doença

    Rat skin physiology is modified by age

    Animal models have been useful in the study of specific mechanisms affecting human skin. Such is the case with ageing and the micromechanical changes determining wrinkles in UV- irradiated mice. These models enabled us to understand that ageing encompasses many peculiar mechanical responses which cannot be explained by homogeneous deformation of the skin. Moreover, a major aspect to consider is that ageing processes differ among species and direct inferences are rarely possible. This project aimed to compare age-induced changes in skin properties by studying two Wistar rat groups of different ages – young adult rats (n=7, 20–24 week-old) and old adult rats (n=5, 48–72 week-old). Non-invasive measuring approaches involving transepidermal water loss (TEWL), superficial hydration (MoistureMeter), and biomechanics (Reviscometer, and Cutometer by MPA80) were taken in the animal dorsum (neck, bottom) over a period of five days, allowing statistical comparisons between groups. Results revealed no significant differences for TEWL, firmness or viscoelasticity between the two groups. However, a significant hydration decrease, of nearly 40%, was noted in the old adult rats group. These results, although preliminary, if translatable to humans could be useful to address age-related changes in processes such as healing, or patch dressing efficacy/safety where hydration is a critical determinant. Their potential use for the prediction of efficacy and or safety of topically applied products for the elderly as compared to young adults may warrant further investigation.

    The polymyxin B-induced transcriptomic response of a clinical, multidrug-resistant Klebsiella pneumoniae involves multiple regulatory elements and intracellular targets

    Background: The emergence of multidrug-resistant Klebsiella pneumoniae is a major public health concern. Many K. pneumoniae infections can only be treated when resorting to last-line drugs such as polymyxin B (PB). However, resistance to this antibiotic is also observed, although insufficient information is described on its mode of action as well as the mechanisms used by resistant bacteria to evade its effects. We aimed to study PB resistance and the influence of abiotic stresses in a clinical K. pneumoniae strain using whole transcriptome profiling. Results: We sequenced 12 cDNA libraries of K. pneumoniae Kp13 bacteria, from two biological replicates of the original strain Kp13 (Kp13) and five derivative strains: induced high-level PB resistance in acidic pH (Kp13(pH)), magnesium deprivation (Kp13(Mg)), high concentrations of calcium (Kp13(Ca)) and iron (Kp13(Fe)), and a control condition with PB (Kp13(PolB)). Our results show the involvement of multiple regulatory loci that differentially respond to each condition as well as a shared gene expression response elicited by PB treatment, and indicate the participation of two-regulatory components such as ArcA-ArcB, which could be involved in re-routing the K. pneumoniae metabolism following PB treatment. Modules of co-expressed genes could be determined, which correlated to growth in acid stress and PB exposure. We hypothesize that polymyxin B induces metabolic shifts in K. pneumoniae that could relate to surviving against the action of this antibiotic. Conclusions: We obtained whole transcriptome data for K. pneumoniae under different environmental conditions and PB treatment.     High Prevalence of Multidrug-Resistant Klebsiella pneumoniae Harboring Several Virulence and β-Lactamase Encoding Genes in a Brazilian Intensive Care Unit

    Klebsiella pneumoniae is an important opportunistic pathogen that commonly causes nosocomial infections and contributes to substantial morbidity and mortality. We sought to investigate the antibiotic resistance profile, pathogenic potential and the clonal relationships between K. pneumoniae (n = 25) isolated from patients and sources at a tertiary care hospital’s intensive care units (ICUs) in the northern region of Brazil. Most of K. pneumoniae isolates (n = 21, 84%) were classified as multidrug resistant (MDR) with high-level resistance to β-lactams, aminoglycosides, quinolones, tigecycline, and colistin. All the 25 isolates presented extended-spectrum beta-lactamase-producing (ESBL), including carbapenemase producers, and carried the blaKPC (100%), blaTEM (100%), blaSHV variants (n = 24, 96%), blaOXA-1 group (n = 21, 84%) and blaCTX-M-1 group (n = 18, 72%) genes. The K2 serotype was found in 4% (n = 1) of the isolates, and the K1 was not detected. The virulence-associated genes found among the 25 isolates were mrkD (n = 24, 96%), fimH-1 (n = 22, 88%), entB (100%), iutA (n = 10, 40%), ybtS (n = 15, 60%). The genes related with efflux pumps and outer membrane porins found were AcrAB (100%), tolC (n = 24, 96%), mdtK (n = 22, 88%), OmpK35 (n = 15, 60%), and OmpK36 (n = 7, 28%). ERIC-PCR was employed to determine the clonal relationship between the different isolated strains. The obtained ERIC-PCR patterns revealed that the similarity between isolates was above 70%. To determine the sequence types (STs) a multilocus sequence typing (MLST) assay was used. The results indicated the presence of high-risk international clones among the isolates. In our study, the wide variety of MDR K. pneumoniae harboring β-lactams and virulence genes strongly suggest a necessity for the implementation of effective strategies to prevent and control the spread of antibiotic resistant infections

    Biological evaluation of crude and degummed oil from Moringa oleifera seeds.

    The aim of this work was to evaluate the oil extracted from Moringa oleifera (fam. Moringaceae) seeds from the nutritional standpoint. Nutritional evaluation of crude or degummed moringa oil or soybean oil (as a control) involved the determination of the Food Efficiency (FE) in male Fisher rats and the fatty acid composition of the moringa oil. Hepatic and renal functions were assessed by measuring serum transaminases activity and urea and creatinine concentrations, respectively. Serum cholesterol and triglycerides, alkaline phosphatase, total proteins and albumine were also measured. Results showed that FE was slightly increased by the crude moringa oil while no differences were found between the soybean and degummed moringa oil regarding this parameter. Renal or hepatic injures as well as major alterations in serum proteins were not induced by the tested oils. These results suggested that degummed M. oleifera oil possessed adequate biological quality as compared to the crude oil

    Genomics and epidemiology for gastric adenocarcinomas (GE4GAC): a Brazilian initiative to study gastric cancer

    Abstract Gastric cancer (GC) is the fifth most common type of cancer worldwide with high incidences in Asia, Central, and South American countries. This patchy distribution means that GC studies are neglected by large research centers from developed countries. The need for further understanding of this complex disease, including the local importance of epidemiological factors and the rich ancestral admixture found in Brazil, stimulated the implementation of the GE4GAC project. GE4GAC aims to embrace epidemiological, clinical, molecular and microbiological data from Brazilian controls and patients with malignant and pre-malignant gastric disease. In this letter, we summarize the main goals of the project, including subject and sample accrual and current findings
