Virtual depth by active background suppression: Revisiting the cosmic muon induced background of GERDA Phase II
In-situ production of long-lived isotopes by cosmic muon interactions may
generate a non-negligible background for deep underground rare event searches.
Previous Monte Carlo studies for the GERDA experiment at LNGS identified the
delayed decays of Ge and its metastable state Ge as dominant
cosmogenic background in the search for neutrinoless double beta decay of
Ge. This might limit the sensitivity of next generation experiments
aiming for increased Ge mass at background-free conditions and thereby
define a minimum depth requirement. A re-evaluation of the Ge
background for the GERDA experiment has been carried out by a set of Monte
Carlo simulations. The obtained Ge production rate is (0.210.01)
nuclei/(kgyr). After application of state-of-the-art active background
suppression techniques and simple delayed coincidence cuts this corresponds to
a background contribution of (2.70.3)
cts/(keVkgyr). The suppression achieved by this strategy equals
an effective muon flux reduction of more than one order of magnitude. This
virtual depth increase opens the way for next generation rare event searches.Comment: 9 pages, 5 figure
LArGe: Background suppression using liquid argon (LAr) scintillation for 0 decay search with enriched germanium (Ge) detectors
Measurements with a bare p-type high purity germanium diode (HPGe) submerged
in a 19 kg liquid argon (LAr) scintillation detector at MPIK Heidelberg are
reported. The liquid argon--germanium system (LArGe) is operated as a 4
anti-Compton spectrometer to suppress backgrounds in the HPGe. This R&D is
carried out in the framework of the GERDA experiment which searches for
0 decays with HPGe detectors enriched in Ge. The goal of
this work is to develop a novel method to discriminate backgrounds in
0 search which would ultimately allow to investigate the
effective neutrino mass free of background events down to the inverse mass

SelbstÀndigkeitserklÀrungDie Verdachtsdiagnose Lyme-Borreliose wird beim Pferd hÀufig durch den
Nachweis von Antikörpern gegen Borrelia (B.) burgdorferi sensu lato (s.l.) im
Zusammenhang mit der in der Literatur beschriebenen Symptomatik gestellt. Ein
direkter Nachweis von Borrelien aus Probenmaterial gestaltet sich durch die
fehlende Standardisierung der Tests sehr schwierig. Die vorliegende Arbeit
versuchte zu klÀren, ob die derzeitige Praxis des indirekten Erregernachweises
fĂŒr die Diagnosestellung Lyme-Borreliose (LB) beim Pferd geeignet ist und
welche Rolle dem direkten Erregernachweis in der Routinediagnostik der LB
ufĂ€llt. In der Klinik fĂŒr Pferdekrankheiten, Allgemeine Chirurgie und
Radiologie der Freien UniversitĂ€t Berlin wurden ĂŒber einen Zeitraum von 10
Monaten 337 Patienten klinisch und labordiagnostisch auf LB untersucht. Die
Einteilung der Probanden in Gruppe A (Pferde ohne auffÀllige Symptomatik im
Sinne einer LB, n=195) und Gruppe B (Pferde mit LB-Ă€hnlichen Symptomen, n=142)
erfolgte im Anschluss an die vollstÀndige klinische Untersuchung. Mit Hilfe
von 2 kommerziell erhÀltlichen ELISA (A und B), einen im BgVV entwickelten
ELISA (C) sowie einem IFT wurden alle Tiere (n=337) auf Antikörper (IgG) gegen
B. burgdorferi s.l. getestet. ZusÀtzlich erfolgte die Gewinnung von
Probenmaterial (Haut, Liquor, Synovia, Vollblut) von LB-verdÀchtigen Pferden,
das fĂŒr den direkten Erregernachweis (kulturelle AnzĂŒchtung, PCR) zur
VerfĂŒgung stand. Zur ĂberprĂŒfung der Reproduzierbarkeit der ELISA wurden im
Western Blot (Labor ALOMED) hochpositive Seren der ELISA A, B, und C (je n=5),
in allen Tests negative Seren (n=3) sowie die beiden Positivkontrollen der
kommerziellen ELISA (A und B) untersucht. Die Positivkontrolle des ELISA C
entsprach einer der hochpositiven Serumproben der ELISA. Mit Hilfe eines
weiteren ELISA (LGL) erfolgte die Detektion von IgA-, IgM- und IgG-AK in
Liquorproben (n=15) sowie von IgM- und IgG-AK in den dazugehörigen Seren
(n=15). Die kulturelle AnzĂŒchtung und die PCR wurden fĂŒr den direkten
Erregernachweis verwendet. Die Kultivierung von Haut- (n=16), Liquor- (n=15)
und Synoviaproben (n=26) erfolgte in nativen sowie mit verschiedenen
Hemmstoffen versetzten BSK- und MKP-Medien bei 33°C ĂŒber 3 Monate. Die
Kulturen wurden 1x wöchentlich subkultiviert und auf Borrelien mittels
Dunkelfeldmikroskopie untersucht. In Haut- (n=14), Liquor- (n=15), Synovia-
(n=26) und Vollblutproben (n=19) erfolgte der Nachweis von B. burgdorferi
s.l.-DNA mit Hilfe der OspA-spezifischen nested-PCR. Zur ĂberprĂŒfung der
SpezifitÀt der positiven PCR-Ergebnisse wurde vom Labor ALOMED eine PCR mit
Hilfe des LightCyclers durchgefĂŒhrt. Die Ergebnisse der serologischen Tests
lassen eine hohe Diskrepanz innerhalb der Seroreagenten erkennen. So
detektierten der ELISA A 61,1%, der ELISA B 9,8% und der ELISA C 3,3% der
Pferde als seropositiv (n=337). Mit dem IFT (Grenztiter 1:128) konnten 33,2%
der Seren positiv bewertet werden. Eine Ăbereinstimmung der serologischen
Testergebnisse lag bei 74 Tieren (22,0%) vor. Davon zeigten 68 Pferde (20,2%)
ein negatives und 6 (1,8%) ein positives Resultat. Ein Unterschied der
SeropositivitÀt zwischen klinisch unauffÀlligen (Gruppe A) und auffÀlligen
Tieren (Gruppe B) war nicht zu erkennen. DarĂŒber hinaus konnte kein
Zusammenhang zwischen Antikörpertitern und verdÀchtigen LB-Symptomen
beobachtet werden. Im Western Blot wurden alle positiven (n=15) und negativen
(n=3) Seren bestÀtigt, die Positivkontrolle der zwei kommerziell erhÀltlichen
ELISA A und B jedoch nicht. Der LGLELISA wies bei den unterschiedlichen
SerumverdĂŒnnungen: (1:50 = VerdĂŒnnung ELISA A) 53,3%, (1:200 = VerdĂŒnnung
ELISA C) 26,7% und (1:400 = VerdĂŒnnung ELISA B) 6,7% seropositive Reagenten
auf. Eine qualitative Ăbereinstimmung lag mit dem ELISA A vor. Die kulturelle
AnzĂŒchtung von B. burgdorferi s.l. aus dem Probenmaterial (n=57) verlief in
allen FĂ€llen negativ. In einer Hautprobe wurden unbewegliche,
spirochÀtenÀhnliche Gebilde beobachtet, bei denen es sich möglicherweise um
Flagellenzöpfe der Begleitkeime handelt. Der spezifische DNA-Nachweis mit
Hilfe der nested-PCR gelang in drei Haut- (3/16), zwei Liquor- (2/15), sechs
Synovia- (6/26) und einer Vollblutprobe (1/19),. Die Realtime-PCR konnte keine
B. burgdorferi s.l.-DNA in diesen Proben nachweisen. Eine Ăbereinstimmung
zwischen dem Nachweis von spezifischer Borrelien-DNA und SeropositivitÀt wurde
nicht registriert. Diese widersprĂŒchlichen Resultate implizieren die
Dringlichkeit der Evaluierung direkter und indirekter Nachweisverfahren von B.
burgdorferi s.l. in der Labordiagnostik. Ohne standardisierte Methodik
(Antigenbeschichtung, SerumverdĂŒnnung, Positivkontrolle) der
serodiagnostischen Tests sind die erforderlichen AnsprĂŒche an die SensitivitĂ€t
und SpezifitĂ€t nicht erfĂŒllt. Nur unter der Voraussetzung einer zuverlĂ€ssigen
Methode sind serologische Verlaufsuntersuchungen sinnvoll, die bei
charakteristischer Titerdynamik und Vorliegen klinischer Zeichen auf die
Verdachtsdiagnose LB hinweisen. Aufgrund der fehlenden Standardisierung der
PCR und der hohen AnsprĂŒche des Erregers an Wachstum und Milieu bei der
kulturellen AnzĂŒchtung sind direkte Nachweisverfahren fĂŒr die
Routinediagnostik derzeit nicht geeignet.The clinical diagnosis of Lyme disease (LD) in horses is often established by
detection of Borrelia (B.) burgdorferi sensu lato (s.l.) antibodies in
conjunction with clinical symptoms as specified in literature. Because of the
lacking standardization of the tests it turned out that the detection of
Borrelia in the specimens was very difficult.The aim of the study was to
evaluate the common practice of serological testing for the establishment of
the diagnosis LD in horses. 337 patients referred to the Clinic for Horses,
Surgery and Radiology of the Freie UniversitÀt Berlin were examined clinically
for a period of ten month and the collected samples were tested further in the
laboratory of the BgVV Berlin. The probands were classified in group A (horses
independent of clinical symptoms, n=195) and group B (horses with clinical
symptoms, n=142) after a full clinical examination. In the study serum samples
of horses (n=337) were tested using two commercial ELISA testkits (A and B),
an ELISA (C) and IFT homemade at the BgVV for B. burgdorferi s.l. antibodies
(immunglobulin G). In addition, specimens of skin, liquor, synovial fluid and
blood of horses with clinical symptoms were analysed by PCR and cultivation
(n=76). In order to determine the sensitivity of the serological assays, both
positive controls of the commercial ELISA testkits (A and B), 5 highly
positive sera of each ELISA (A, B and C) and 3 sera tested negative in all of
them were examined with Western blot (ALOMED, n=20). The positive control of
the ELISA C consisted of a sample that was high positive in all serological
tests. In a further study, 15 liquor samples were tested for B. burgdorferi
s.l. antibodies (immunglobulin A, M and G) and the corresponding serum samples
(n=15) for immunglobulin M and G using another ELISA (LGL). In our
investigations we examined Lyme disease in horses using direct methods for
detection of B. burgdorferi s.l.: culture and Polymerase Chain Reaction (PCR).
For the culture of B. burgdorferi skin (n=16), liquor (n=15) and synovial
fluid (n=26) were inoculated into BSK- and MKP-medium native and with
different inhibitoring substances. Cultures were incubated for 3 months at
33°C, subcultivated and examinated weekly by dark-field microscopy. Skin
(n=16), liquor (n=15), synovial fluid (n=26) and blood (n=19) were tested for
the presence of B. burgdorferi s.l. specific DNA by OspA-specific nested PCR.
In order to determine the specivity of the nested PCR the positive PCR-results
were examined with Realtime PCR (ALOMED). The results of the serologic testing
revealed a high discrepancy within the tested probands. Following proportions
of positive antibody titers were found (n=337): ELISA A: 61,1%, B: 9,8%, C:
3,3%. In the IFT 33,2% of samples were positive (cut off 1:128). Compatibility
of results of serological tests existed in 22,0% of samples (74/337), only 1,8
% of samples were positive in all tests (6/337). There was no significant
difference in serological results between horses with (group B) and without
(group A) clinical symptoms of LD. A correlation between symptoms and
seropositivity of serum samples could not be proved. In the Western blot,
positive (n=15) and negative in all ELISA (n=3) sera except the positive
controls of the commercial ELISA testkits (A and B) were confirmed. The LGL-
ELISA detected incidences of seropositive samples (depending on dilution of
serum) as follows: 53,3% (1:50), 26,7% (1:200) and 6,7% (1:400). These results
are in correspondence with ELISA A. The cultivation of B. burgdorferi was
negative in all cases (n=57). In one of them we could find immobile
spirochete-like forms. They are supposed to be giant whips , flagellae of the
contaminant flora, which are not involved in the development of Lyme disease.
In the OspA-specific nested PCR 3 skin (3/16), 2 liquor (2/15), 6 synovial
fluid (6/26) and 1 blood (1/19) samples were positive for B. burgdorferi
s.l.-DNA. In the Realtime PCR none of these 12 positive samples were positive
for B. burgdorferi s.l.-DNA (0/12). A correlation between seropositivity of
serum samples and the presence of specific DNA could not be proven. These
contradictory results revealed the necessity for evaluation of current direct
and serological laboratory testing for B. burgdorferi s.l. Without
standardised methods (concerning antigencoating, serumdilution, positive
control) for the serodiagnostical tests the demands regarding specifity and
sensitivity are not met. Reliable laboratory tools are a prerequisite under
which a characteristic titerdynamic in conjunction with clinical signs can
leed to the presumptive diagnosis LD. Due to the lack of standardization of
PCR and the complex demands on cultivation direct detection of B. burgdorferi
s.l. is not a suitable method for routine diagnostic at the moment
Solar and Reactor Neutrinos: Upcoming Experiments and Future Projects
Sub-MeV solar neutrino experiments and long-baseline reactor oscillation
experiments toe the cutting edge of neutrino research. The upcoming experiments
KamLAND and BOREXINO, currently in their startup and final construction phase
respectively, will provide essential information on neutrino properties as well
as on solar physics. Future projects, at present under development, will
measure the primary solar neutrino fluxes via electron scattering and neutrino
capture in real time. High precision data for lepton mixing as well as for
stellar evolution theory will become available in the future. This paper aims
to give an overview of the upcoming experiments and of the projects under
