    Tracking Optical and Electronic Behaviour of Quantum Contacts in Sub-Nanometre Plasmonic Cavities.

    Plasmonic interactions between two metallic tips are dynamically studied in a supercontinuum dark-field microscope and the transition between coupled and charge-transfer plasmons is directly observed in the sub-nm regime. Simultaneous measurement of the dc current, applied force, and optical scattering as the tips come together is used to determine the effects of conductive pathways within the plasmonic nano-gap. Critical conductances are experimentally identified for the first time, determining the points at which quantum tunnelling and conductive charge transport begin to influence plasmon coupling. These results advance our understanding of the relationship between conduction and plasmonics, and the fundamental quantum mechanical behaviours of plasmonic coupling.The authors would like to acknowledge Nanotools GmbH for their contributions and support to this project.     Bars from the Inside Out: An HST Study of their Dusty Circumnuclear Regions

    The results of bar-driven mass inflow are directly observable in high-resolution HST observations of their circumnuclear regions. These observations reveal a wealth of structures dominated by dust lanes, often with a spiral-like morphology, and recent star formation. Recent work has shown that some of these structures are correlated with the presence or absence of a bar. I extend this work with an investigation of circumnuclear morphology as a function of bar strength for a sample of 48 galaxies with both measured bar strengths and ``structure maps'' computed from HST images. The structure maps for these galaxies, which have projected spatial resolutions of 2 - 15 pc, show that the fraction of galaxies with grand-design (GD) circumnuclear dust spirals increases significantly with bar strength, while tightly wound dust spirals are only present in the most axisymmetric galaxies. GD structure is only found at the centers of galaxies classified as SB(s) or SB(rs) and not SB(r). SB(s) galaxies on average have stronger bars than SB(r) galaxies. There is also a modest increase in the fraction of loosely wound dust spirals at later morphological types, which may reflect an increase in the fraction of galaxies with circumnuclear, gaseous disks.     Potent induction of antibody-secreting B-cells by human dermal-derived CD14+ dendritic cells triggered by dual toll-like receptor ligation

    Targeting CD14(+) dermal-derived dendritic cells (DDCs) is a rational approach for vaccination strategies aimed at improving humoral immune responses, because of their natural ability to stimulate naïve B-cells. Here, we show that CD14(+) DDCs express mRNA for TLRs 1–9, but respond differentially to single or paired TLR ligands. Compared to single ligands, some combinations were particularly effective at activating CD14(+) DDCs, as shown by enhanced expression of B-cell stimulatory cytokines (IL-6, IL-10 and TNF-α) and more pronounced phenotypic maturation. These combinations were Resiquimod (R-848) plus Polyinosinic:polycytidylic acid (Poly(I:C)); R-848 plus LPS; Pam3CSK4 plus Poly(I:C); LPS plus Poly(I:C). We also found that selected TLR ligand pairs (R-848 plus either LPS or Poly(I:C)) were superior to individual agents at boosting the inherent capacity of CD14(+) DDCs to induce naïve B-cells to proliferate and differentiate into CD27(+) CD38(+) B-cells that secrete high levels of IgG and IgA. When treated with the same TLR ligand combinations, CD14(+) DDCs also promoted the differentiation of Th1 (IFN-γ-secreting) CD4(+) T-cells, but not of Th2 or Th17 CD4(+) T-cells. These observations may help to identify adjuvant strategies aimed at inducing protective immune responses to various pathogens, including but not limited to HIV-1

    Understanding the plasmonics of nanostructured atomic force microscopy tips

    Structured metallic tips are increasingly important for optical spectroscopies such as tip-enhanced Raman spectroscopy, with plasmonic resonances frequently cited as a mechanism for electric field enhancement. We probe the local optical response of sharp and spherical-tipped atomic force microscopy (AFM) tips using a scanning hyperspectral imaging technique to identify the plasmonic behaviour. Localised surface plasmon resonances which radiatively couple with far-field light are found only for spherical AFM tips, with little response for sharp AFM tips, in agreement with numerical simulations of the near-field response. The precise tip geometry is thus crucial for plasmon-enhanced spectroscopies, and the typical sharp cones are not preferred.The authors thank EPSRC Grant Nos.     Generation of a stable, soluble mimic of the HIV-1 envelope glycoprotein (Env) trimer on the virion surface has been considered an important first step for developing a successful HIV-1 vaccine. Recently, a soluble native-like Env trimer (BG505 SOSIP.664) has been described. This protein has facilitated major advances in the HIV-1 vaccine field, since it was the first Env immunogen that induced consistent neutralizing antibodies against a neutralization-resistant (tier 2) virus. Moreover, BG505 SOSIP.664 enabled elucidation of the atomic resolution structure of the Env trimer and facilitated the isolation and characterization of new broadly neutralizing antibodies against HIV-1. Here, we designed and characterized the BG505 SOSIP.664 trimer fused to fluorescent superfolder GFP (sfGFP), a GFP variant that allows efficient folding (BG505 SOSIP.664-sfGFP). Despite the presence of the sfGFP, the Env protein largely retained its morphology, antigenicity, glycan composition, and thermostability. In addition, we show that BG505 SOSIP.664-sfGFP can be used for fluorescence-based assays, such as flow cytometry.</p

    Vigorous star formation hidden by dust in a galaxy at z=1.4z=1.4

    Near-infrared surveys have revealed a substantial population of enigmatic faint galaxies with extremely red optical-to-near-infrared colours and with a sky surface density comparable to that of faint quasars. There are two scenarios for these extreme colours: (i) these distant galaxies have formed virtually all their stars at very high redshifts and, due to the absence of recently formed stars, the colours are extremely red and (ii) these distant galaxies contain large amounts of dust, severely reddening the rest-frame UV--optical spectrum. HR10 (z=1.44z = 1.44) is considered the archetype of the extremely red galaxies. Here we report the detection of the continuum emission from HR10 at 850ÎĽ\mum and at 1250ÎĽ\mum, demonstrating that HR10 is a very dusty galaxy undergoing a major episode of star formation. Our result provides a clear example of a high-redshift galaxy where the star formation rate inferred from the ultraviolet luminosity would be underestimated by a factor up to 1000, and shows that great caution should be used to infer the global star formation history of the Universe from optical observations only.Comment: 12 pages, 1 figure, Nature, in press (30 April 1998

    HIV vaccine: it may take two to tango, but no party time yet

    A press conference on Thursday September 24 in Bangkok, Thailand, released data that an experimental vaccine provided mild protection against HIV-1 infection. This is the first positive signal of any degree of vaccine efficacy in humans, more than a quarter-century after scientists discovered the virus that causes AIDS. The research was conducted by a team including Thai researchers, the U.S. Army and the U.S. National Institutes of Health. The RV144 Phase III clinical trial, which began in 2003, had been disparaged by many critics as a waste of time and money because each of the two components had been shown to produce no benefit as individual vaccines and because the scientific rationales behind the immunogens were just wrong. It was nevertheless speculated that using them together in the prime-boost scenario could be more effective, with the aim to induce heightened CD4+ cellular immune responses against the viral Envelope protein. This optimism seems to have been validated. In fact, this would not be the first time that the discovery of an effective vaccine relied as much on serendipity as opposed to scientific rationale. On the other hand, many questions remain about the RV144 trial, and these issues will be addressed in this editorial
