Results Osler emphasised the Art of Detachment, the Virtue of Method, the Quality of Thoroughness and the Grace of Humility, which he considers the basic pillars on which a successful medical practice is achieved, Osler's words serve as a guiding light for physicians struggling with the demands of twenty first century medicine

    Rapid isolation of mycoviral double-stranded RNA from Botrytis cinerea and Saccharomyces cerevisiae

    <p>Abstract</p> <p>Background</p> <p>In most of the infected fungi, the mycoviruses are latent or cryptic, the infected fungus does not show disease symptoms, and it is phenotypically identical to a non-infected strain of the same species. Because of these properties, the initial stage in the search for fungi infected with mycoviruses is the detection of their viral genome, which in most of the described cases corresponds to double-stranded RNA (dsRNA). So to analyze a large number of fungal isolates it is necessary to have a simple and rapid method to detect dsRNA.</p> <p>Results</p> <p>A rapid method to isolate dsRNA from a virus-infected filamentous fungus, <it>Botrytis cinerea</it>, and from a killer strain of <it>Saccharomyces cerevisiae </it>using commercial minicolumns packed with CF11 cellulose was developed. In addition to being a rapid method, it allows to use small quantities of yeasts or mycelium as starting material, being obtained sufficient dsRNA quantity that can later be analyzed by agarose gel electrophoresis, treated with enzymes for its partial characterization, amplified by RT-PCR and cloned in appropriate vectors for further sequencing.</p> <p>Conclusions</p> <p>The method yields high quality dsRNA, free from DNA and ssRNA. The use of nucleases to degrade the DNA or the ssRNA is not required, and it can be used to isolate dsRNA from any type of fungi or any biological sample that contains dsRNA.</p

    Andy's Algorithms: new automated digital image analysis pipelines for FIJI.

    Quantification of cellular antigens and their interactions via antibody-based detection methods are widely used in scientific research. Accurate high-throughput quantitation of these assays using general image analysis software can be time consuming and challenging, particularly when attempted by users with limited image processing and analysis knowledge. To overcome this, we have designed Andy's Algorithms, a series of automated image analysis pipelines for FIJI, that permits rapid, accurate and reproducible batch-processing of 3,3'-diaminobenzidine (DAB) immunohistochemistry, proximity ligation assays (PLAs) and other common assays. Andy's Algorithms incorporates a step-by-step tutorial and optimization pipeline to make batch image analysis simple for the untrained user and adaptable across laboratories. Andy's algorithms provide a simpler, faster, standardized work flow compared to existing programs, while offering equivalent performance and additional features, in a free to use open-source application of FIJI.     Allele-Specific Impairment of GJB2 Expression by GJB6 Deletion del(GJB6-D13S1854)

    Mutations in the GJB2 gene, which encodes connexin 26, are a frequent cause of congenital non-syndromic sensorineural hearing loss. Two large deletions, del(GJB6-D13S1830) and del(GJB6-D13S1854), which truncate GJB6 (connexin 30), cause hearing loss in individuals homozygous, or compound heterozygous for these deletions or one such deletion and a mutation in GJB2. Recently, we have demonstrated that the del(GJB6-D13S1830) deletion contributes to hearing loss due to an allele-specific lack of GJB2 mRNA expression and not as a result of digenic inheritance, as was postulated earlier. In the current study we investigated the smaller del(GJB6-D13S1854) deletion, which disrupts the expression of GJB2 at the transcriptional level in a manner similar to the more common del(GJB6-D13S1830) deletion. Interestingly, in the presence of this deletion, GJB2 expression remains minimally but reproducibly present. The relative allele-specific expression of GJB2 was assessed by reverse-transcriptase PCR and restriction digestions in three probands who were compound heterozygous for a GJB2 mutation and del(GJB6-D13S1854). Each individual carried a different sequence variant in GJB2. All three individuals expressed the mutated GJB2 allele in trans with del(GJB6-D13S1854), but expression of the GJB2 allele in cis with the deletion was almost absent. Our study clearly corroborates the hypothesis that the del(GJB6-D13S1854), similar to the larger and more common del(GJB6-D13S1830), removes (a) putative cis-regulatory element(s) upstream of GJB6 and narrows down the region of location

    ECOLOGICAL SERVICES OF AGROFORESTRY LANDSCAPES IN SELECTED WATERSHED AREAS IN THE PHILIPPINES AND INDONESIA

    This article argues that the practice of agroforestry provides ecological contributions to the smallholder farmers cultivating in the watershed areas. Specifically, this farming system provides contribution to carbon sequestration potential of the woody perennials and the biodiversity conservation of the other components of the system.   This argument is based on the research conducted in Molawin-Dampalit Sub-Watershed, Mt. Makiling Forest Reserve in the Philippines and Way Betung Watershed in Indonesia.  The research involved an interview session of 106 and 261 smallholder farmers and an assessment of 27 and 14 agroforesty plots for carbon stock assessment and biodiversity assessment, respectively. Results indicated that the total carbon found among the crop components was 52.32 MgC/ha in Molawin-Dampalit Sub-Watershed­ and 244.26 MgC/ha in Way Betung Watershed­, which suggested the high carbon sequestration potential of the woody perennials and understory crops in an agroforestry system.  The farm lots being cultivated by the smallholder farmers were found to contribute to biodiversity conservation having a moderate biodiversity index of 2.59 and 2.53, respectively.  With these findings, promotion of desired agroforestry systems in suitable portions of the watershed areas should be intensified and heightened to contribute to ecological balance across the landscape.  Agroforestry should always be an integral part of all initiatives toward ecological restoration with the cultivators/smallholder farmers as potential partners.  The agroforestry system should consider all the technical and socioeconomic considerations toward having diverse components and ensure food security among the smallholder farmers throughout the year

    Influence of system dynamics in brake blending strategies for electric vehicles

    Regenerative and friction braking blending strategies need to consider both system dynamics in order to optimize their performance. Usually, the priority in electric vehicles is battery regeneration through electric braking instead of friction braking. This work studies the dynamics of both systems and proposes an optimized brake-blending strategy. The goal is to maximize regeneration without affecting safety. Both dynamics are studied separately with commercial systems: electric drivetrain and friction brake-by-wire. The proposed strategy takes into account temporary response as well as the physical limitations of the systems. Therefore, this strategy limits the influence of the slowest system, in our case, the electric one, during the braking process while maximizing battery regeneration.Universidad de MĂĄlaga.     Analysis of symmetries in models of multi-strain infections

    In mathematical studies of the dynamics of multi-strain diseases caused by antigenically diverse pathogens, there is a substantial interest in analytical insights. Using the example of a generic model of multi-strain diseases with cross-immunity between strains, we show that a significant understanding of the stability of steady states and possible dynamical behaviours can be achieved when the symmetry of interactions between strains is taken into account. Techniques of equivariant bifurcation theory allow one to identify the type of possible symmetry-breaking Hopf bifurcation, as well as to classify different periodic solutions in terms of their spatial and temporal symmetries. The approach is also illustrated on other models of multi-strain diseases, where the same methodology provides a systematic understanding of bifurcation scenarios and periodic behaviours. The results of the analysis are quite generic, and have wider implications for understanding the dynamics of a large class of models of multi-strain diseases
