    Galactic Plane Hα\alpha Surveys: IPHAS & VPHAS+

    The optical Galactic Plane Hα\alpha surveys IPHAS and VPHAS+ are dramatically improving our understanding of Galactic stellar populations and stellar evolution by providing large samples of stars in short lived, but important, evolutionary phases, and high quality homogeneous photometry and images over the entire Galactic Plane.     First results of an Hα based search of classical Be stars in the Perseus Arm and beyond

    We investigate a region of the Galactic plane, between 120° ≤ l ≤ 140° and-1° ≤ b≤+4°, and uncover a population of moderately reddened (E(B-V) ~ 1) classical Be stars within and beyond the Perseus and Outer Arms. 370 candidate emission-line stars (13≲r≲16) selected from the Isaac Newton Telescope Photometric Ha Survey of the Northern Galactic plane have been followed up spectroscopically. A subset of these, 67 stars with properties consistent with those of classical Be stars, have been observed at sufficient spectral resolution (δλ ≈ 2-4 Å) at blue wavelengths to narrow down their spectral types. We determine these to a precision estimated to be ±1 subtype and then we measure reddenings via spectral energy distribution fitting with reference to appropriate model atmospheres. Corrections for contribution to colour excess from circumstellar discs are made using an established scaling to Ha emission equivalent width. Spectroscopic parallaxes are obtained after luminosity class has been constrained via estimates of distances to neighbouring A/F stars with similar reddenings. Overwhelmingly, the stars in the sample are confirmed as luminous classical Be stars at heliocentric distances ranging from 2 kpc up to ~12 kpc. However, the errors are presently too large to enable the cumulative distribution function with respect to distance to distinguish between models placing the stars exclusively in spiral arms, or in a smooth exponentially declining distribution.Peer reviewe

    Bat and pig IFN-induced transmembrane protein 3 restrict cell entry by influenza virus and lyssaviruses

    IFN-induced transmembrane protein 3 (IFITM3) is a restriction factor that blocks cytosolic entry of numerous viruses that utilize acidic endosomal entry pathways. In humans and mice, IFITM3 limits influenza-induced morbidity and mortality. Although many IFITM3-sensitive viruses are zoonotic, whether IFITMs function as antiviral restriction factors in mammalian species other than humans and mice is unknown. Here, IFITM3 orthologues in the microbat (Myotis myotis) and pig (Sus scrofa domesticus) were identified using rapid amplification of cDNA ends. Amino acid residues known to be important for IFITM3 function were conserved in the pig and microbat orthologues. Ectopically expressed pig and microbat IFITM3 co-localized with transferrin (early endosomes) and CD63 (late endosomes/multivesicular bodies). Pig and microbat IFITM3 restricted cell entry mediated by multiple influenza haemagglutinin subtypes and lyssavirus glycoproteins. Expression of pig or microbat IFITM3 in A549 cells reduced influenza virus yields and nucleoprotein expression. Conversely, small interfering RNA knockdown of IFITM3 in pig NPTr cells and primary microbat cells enhanced virus replication, demonstrating that these genes are functional in their species of origin at endogenous levels. In summary, we showed that IFITMs function as potent broad-spectrum antiviral effectors in two mammals – pigs and bats – identified as major reservoirs for emerging viruses

    Sperm competition shapes gene expression and sequence evolution in the ocellated wrasse.

    Gene expression differences between males and females often underlie sexually dimorphic phenotypes, and the expression levels of genes that are differentially expressed between the sexes are thought to respond to sexual selection. Most studies on the transcriptomic response to sexual selection treat sexual selection as a single force, but postmating sexual selection in particular is expected to specifically target gonadal tissue. The three male morphs of the ocellated wrasse (Symphodus ocellatus) make it possible to test the role of postmating sexual selection in shaping the gonadal transcriptome. Nesting males hold territories and have the highest reproductive success, yet we detected feminization of their gonadal gene expression compared to satellite males. Satellite males are less brightly coloured and experience more intense sperm competition than nesting males. In line with postmating sexual selection affecting gonadal gene expression, we detected a more masculinized expression profile in satellites. Sneakers are the lowest quality males and showed both de-masculinization and de-feminization of gene expression. We also detected higher rates of gene sequence evolution of male-biased genes compared to unbiased genes, which could at least in part be explained by positive selection. Together, these results reveal the potential for postmating sexual selection to drive higher rates of gene sequence evolution and shape the gonadal transcriptome profile

    Productivity in temperate tree crops such as apple has been lifted several-fold by research focusing on optimising a combination of canopy components including light relations, vigour control, tree architecture and crop load. This paper outlines the research behind the Small Tree-High Productivity Initiative (STHPI), which is focused on improving productivity of mango, avocado and macadamia. Preliminary results from work we are undertaking for each of the above canopy components in mango will be outlined. A rootstock screening trial to identify vigour-managing, high-productivity rootstocks is being undertaken, and we present a comparison of the best-performing low-medium vigour rootstock varieties compared with control 'Kensington Pride' (KP) rootstock at 6 months old. Comparisons between 'Keitt', 'NMBP 1243' and 'Calypso' scion cultivars with regard to tree diameter, height and canopy growth at different orchard densities and training systems will also be presented. Preliminary results from an orchard light-relations study indicate that mango yields continued to increase with light interception up to 50%, and reached a maximum of 20-30 t ha at 68% light interception in KP trees approximately 25 years old. In a crop load trial, inflorescence thinning in a 'Calypso' orchard did not significantly reduce yields when up to 90% of inflorescences were removed, but did when 95% of inflorescences were removed, as trees were unable to compensate by adjusting fruit set, size and yield. Inflorescence thinning beyond 80% increased the number of fruit set per panicle, and thinning up to and including 90% of inflorescences increased fruit weight from 340 g to >400 g per fruit. This project is still in its initial stages; however, early indications suggest there may be opportunities to improve early orchard yields through optimising light interception in an orchard's life, potentially through the use of higher densities, and that rootstocks and tree training methods, once identified, may help in the management of vigour. It is also hoped to obtain a better understanding of how crop load influences the balance between vegetative growth, flowering, fruiting, alternate bearing and fruit quality

    Mango germplasm screening for the identification of sources of tolerance to anthracnose

    Colletotrichum species are one of the most common causes of postharvest fruit rot in mango in Australia, particularly in the tropical region of north Queensland, and can result in significant losses if not managed. The research aims were to identify sources of anthracnose tolerance and to determine if host material other than fruit could improve or fast track the screening process and result in improved breeding efficiency. Access to the Australian National Mango Genebank (ANMG) collection enabled fruit screening of more than 100 Mangifera indica cultivars or Mangifera species for tolerance to anthracnose by artificial inoculation with Colletotrichum asianum over a period of 14 years. Mean lesion diameters were compared with those on a known susceptible M. indica cultivar Kensington Pride (KP) and a tolerant M. laurina cultivar Lombok. Inoculation of leaf discs and entire leaves was evaluated in the laboratory and the field as alternative assays for tolerance to anthracnose and was assessed by presence/absence of disease. Screening of fruit has shown that anthracnose tolerance within the mango germplasm is highly variable and needs to be assessed over multiple years. None of the alternative laboratory bioassays provided consistent or reliable data. The in-field artificial inoculation of immature leaf flush was successful but was not deemed suitable for adoption due to practical restraints. While resistance to anthracnose in fruit has not yet been identified, some cultivars and Mangifera spp. showed promise for inclusion as parents in future breeding programs

    Site-Specific Iron Substitution in STA-28, a Large Pore Aluminophosphate Zeotype Prepared by Using 1, 10-Phenanthrolines as Framework-Bound Templates

    An AlPO4 zeotype has been prepared using the aromatic diamine 1, 10-phenanthroline and some of its methylated analogues as templates. In each case the two template N atoms bind to a specific framework Al site to expand its coordination to the unusual octahedral AlO4N2 environment. Furthermore, using this framework-bound template, Fe atoms can be included selectively at this site in the framework by direct synthesis, as confirmed by annular dark field scanning transmission electron microscopy and Rietveld refinement. Calcination removes the organic molecules to give large pore framework solids, with BET surface areas up to 540 m2 g-1 and two perpendicular sets of channels that intersect to give pore space connected by 12-ring openings along all crystallographic directions
