AKT activation controls cell survival in response to HDAC6 inhibition.
HDAC6 is emerging as an important therapeutic target for cancer. We investigated mechanisms responsible for survival of tumor cells treated with a HDAC6 inhibitor. Expression of the 20 000 genes examined did not change following HDAC6 treatment in vivo. We found that HDAC6 inhibition led to an increase of AKT activation (P-AKT) in vitro, and genetic knockdown of HDAC6 phenocopied drug-induced AKT activation. The activation of AKT was not observed in PTEN null cells; otherwise, PTEN/PIK3CA expression per se did not predict HDAC6 inhibitor sensitivity. Interestingly, HDAC6 inhibitor treatment led to inactivating phosphorylation of PTEN (P-PTEN Ser380), which likely led to the increased P-AKT in cells that express PTEN. Synergy was observed with phosphatidylinositol 3-kinases (PI3K) inhibitor treatment in vitro, accompanied by increased caspase 3/7 activity. Furthermore, combination of HDAC6 inhibitor with a PI3K inhibitor caused substantial tumor growth inhibition in vivo compared with either treatment alone, also detectable by Ki-67 immunostaining and (18)F-FLT positron emission tomography (PET). In aggregate AKT activation appears to be a key survival mechanism for HDAC6 inhibitor treatment. Our findings indicate that dual inhibition of HDAC6 and P-AKT may be necessary to substantially inhibit growth of solid tumors
Comprehensive surface magnetotransport study of SmB6
After the theoretical prediction that SmB6 is a topological Kondo insulator, there has been an explosion of studies on the SmB6 surface. However, there is not yet an agreement on even the most basic quantities such as the surface carrier density and mobility. In this paper, we carefully revisit Corbino disk magnetotransport studies to find those surface transport parameters. We first show that subsurface cracks exist in the SmB6 crystals, arising both from surface preparation and during the crystal growth. We provide evidence that these hidden subsurface cracks are additional conduction channels, and the large disagreement between earlier surface SmB6 studies may originate from previous interpretations not taking this extra conduction path into account. We provide an update of more reliable magnetotransport data than the previous one (S. Wolgast et al., Phys. Rev. B 92, 115110) and find that the orders-of-magnitude large disagreements in carrier density and mobility come from the surface preparation and the transport geometry rather than the intrinsic sample quality. From this magnetotransport study, we find an updated estimate of the carrier density and mobility of 2.71×1013 (1/cm2) and 104.5 (cm2/Vsec), respectively. We compare our results with other studies of the SmB6 surface. By this comparison, we provide insight into the disagreements and agreements of the previously reported angle-resolved photoemission spectroscopy, scanning tunneling microscopy, and magnetotorque quantum oscillations measurements
Efficacy of chloroquine, sulphadoxine-pyrimethamine and amodiaquine for treatment of uncomplicated Plasmodium falciparum malaria in Kajo Keji county, Sudan.
To provide advice on the rational use of antimalarial drugs, Médecins Sans Frontières conducted a randomized, an open label efficacy study in Kajo Keji, an area of high transmission of malaria in southern Sudan. The efficacy of chloroquine (CQ), sulphadoxine-pyrimethamine (SP) and amodiaquine (AQ) were measured in a 28-day in vivo study, with results corrected by PCR genotyping. Of 2010 children screened, 115 children aged 6-59 months with uncomplicated Plasmodium falciparum malaria were randomized into each group to receive a supervised course of treatment. Of these, 114, 103 and 111 were analysed in the CQ, SP and AQ groups, respectively. The overall parasitological failure rates at day 28 were 93.9% [95% confidence interval (CI) 87.3-97.3] for CQ, 69.9% (95% CI 60.0-78.3) for SP, and 25.2% (95% CI 17.7-34.5) for AQ. These results provide important missing data on antimalarial drug efficacy in southern Sudan. They indicate that none of the drugs could be used in monotherapy and suggest that even in combination with artemisinin, cure rates might not be efficacious enough. We recommend a combination of artemether and lumefantrine as first-line treatment for uncomplicated P. falciparum malaria cases in Kajo Keji county
The Web of Human Sexual Contacts
Many ``real-world'' networks are clearly defined while most ``social''
networks are to some extent subjective. Indeed, the accuracy of
empirically-determined social networks is a question of some concern because
individuals may have distinct perceptions of what constitutes a social link.
One unambiguous type of connection is sexual contact. Here we analyze data on
the sexual behavior of a random sample of individuals, and find that the
cumulative distributions of the number of sexual partners during the twelve
months prior to the survey decays as a power law with similar exponents for females and males. The scale-free nature of the web of human
sexual contacts suggests that strategic interventions aimed at preventing the
spread of sexually-transmitted diseases may be the most efficient approach.Comment: 7 pages with 2 eps figures. Latex file. For more details or for

Quantum oscillations in Kondo Insulator SmB
In Kondo insulator samarium hexaboride SmB, strong correlation and band
hybridization lead to an insulating gap and a diverging resistance at low
temperature. The resistance divergence ends at about 5 Kelvin, a behavior
recently demonstrated to arise from the surface conductance. However, questions
remain whether and where a topological surface state exists. Quantum
oscillations have not been observed to map the Fermi surface. We solve the
problem by resolving the Landau Level quantization and Fermi surface topology
using torque magnetometry. The observed Fermi surface suggests a two
dimensional surface state on the (101) plane. Furthermore, the tracking of the
Landau Levels in the infinite magnetic field limit points to -1/2, which
indicates a 2D Dirac electronic state
