    MicroRNA in autoimmunity and autoimmune diseases

    MicroRNAs (miRNAs) are small conserved non-coding RNA molecules that post-transcriptionally regulate gene expression by targeting the 3′ untranslated region (UTR) of specific messenger RNAs (mRNAs) for degradation or translational repression. miRNA-mediated gene regulation is critical for normal cellular functions such as the cell cycle, differentiation, and apoptosis, and as much as one-third of human mRNAs may be miRNA targets. Emerging evidence has demonstrated that miRNAs play a vital role in the regulation of immunological functions and the prevention of autoimmunity. Here we review the many newly discovered roles of miRNA regulation in immune functions and in the development of autoimmunity and autoimmune disease. Specifically, we discuss the involvement of miRNA regulation in innate and adaptive immune responses, immune cell development, T regulatory cell stability and function, and differential miRNA expression in rheumatoid arthritis and systemic lupus erythematosus

    Presence of \u3cem\u3ePorphyromonas gingivalis\u3c/em\u3e in gingival squamous cell carcinoma

    Periodontal disease has been recently linked to a variety of systemic conditions such as diabetes, cardiovascular disease, preterm delivery, and oral cancer. The most common bacteria associated with periodontal disease, Porphyromonas gingivalis (P. gingivalis) has not yet been studied in the malignant gingival tissues. The objective of this study was to investigate the presence of P. gingivalis in specimens from squamous cell carcinoma patients. We have performed immunohistochemical staining to investigate the presence of P. gingivalis and Streptococcus gordonii (S. gordonii), a non invasive oral bacteria, in paraffin embedded samples of gingival squamous cell carcinoma (n=10) and normal gingiva (n=5). Staining for P. gingivalis revealed the presence of the bacteria in normal gingival tissues and gingival carcinoma, with higher levels (more than 33%,P\u3c0.05) detected in the carcinoma samples. The staining intensity was also significantly enhanced in the malignant tissue by 2 folds (P\u3c0.023) compared to specimens stained for the non‐invasive S. gordonii. P. gingivalis is abundantly present in malignant oral epithelium suggesting a potential association of the bacteria with gingival squamous cell carcinoma

    Effect of Metformin on miR-146a Expression

    Sjögren’s Syndrome (SjS) is an autoimmune disorder that affects secretory glands in the human body, restricting their function and causing extreme dryness in areas like the mouth and eyes. miR-146a is an anti-inflammatory microRNA that targets the NFκB activation pathway. Previous studies have shown that SjS patients have increased miR-146a expression, despite having high levels of inflammation. The objective of this study was to investigate whether metformin, a diabetes drug with a wide variety of effects and potential functions, reduces levels of miR-146a expression. Metformin is known to reduce inflammation by inhibiting the activation of NFκB. THP-1 human monocytes were treated with various concentrations of metformin ranging from 12.5uM to 200uM. The cells were treated for 24 hours before total RNA was isolated, and qRT-PCR was utilized to compare miR-146a expression in metformin-treated versus untreated cells. Our results showed a dose-dependent decrease of miR-146a expression in the presence of metformin. These results are reasonable since miR-146a expression is dependent on NFκB activation, and metformin is known to inhibit the activation of NFκB. Further studies will investigate metformin’s ability to suppress inflammation in varying conditions

    The impact of body composition on energy expenditure during walking and running in young adults

    The purpose of this study was to examine the impact of body composition on energy expenditure (EE) of 164 young adults during a 1-mile walk and a 1-mile run on a treadmill. Segmental bioimpedance was used to measure body composition variables. The EE in men (108.3 ± 17.6 kcal) was greater than (P \u3c 0.05) women (80.3 ± 10.6 kcal) during the 1-mile walk, and the difference increased in magnitude during the 1-mile run (144.9 ± 23.2 kcal vs. 105.1 ± 14.9 kcal, respectively). When EE was expressed per unit of body mass, men and women were similar. However, women had a higher EE per unit of fat-free mass (FFM). Regardless of gender, running 1-mile resulted in a greater EE than walking 1-mile. In addition, men expended more absolute calories than women due to a higher body mass. When EE was examined relative to FFM, women were found to be less economical than men, which was most likely due to carrying larger amounts of inactive adipose tissue

    Conservation Status and Habitat Use of the West Virginia Spring Salamander (Gyrinophilus Subterraneus) and Spring Salamander (G. Porphyriticus) in General Davis Cave, Greenbrier Co., West Virginia

    The West Virginia Spring Salamander (Gyrinophilus subterraneus) is one of four obligate cave-dwelling species of plethodontid salamanders found east of the Mississippi River in the United States. This species is endemic to a single cave system; General Davis Cave, in Greenbrier Co., West Virginia, where it is syntopic with the closely-related Spring Salamander (G. porphyriticus). Accordingly, the West Virginia Spring Salamander is a species of critical conservation concern. Because of it\u27s conservation status and lack of data regarding the ecology and life history, particularly about population trends, we present data on relative abundance of and habitat use by the West Virginia Spring Salamander during a 33-year period from 1975-2008. Specifically we address: (1) stability of the population during the last 33 years; (2) variation in habitat use by life stage and between species (Spring Salamanders and West Virginia Spring Salamanders); (3) plausibility of neoteny in the West Virginia Spring Salamander; and (4) the conservation status of the West Virginia Spring Salamander. We recorded 324 observations of Gyrinophilus salamanders, of which 192 were West Virginia Spring Salamanders, within the study area during 17 surveys. While both larval and metamorphosed West Virginia Spring Salamanders were encountered, only metamorphosed Spring Salamanders were observed. West Virginia Spring Salamander larvae were encountered in pools more often than in riffle habitat. Spring Salamanders were encountered more often in terrestrial habitats versus aquatic habitats. West Virginia Spring Salamanders reach relatively large size before metamorphosing, with some individuals becoming sexual mature as larvae. It remains unknown whether any of these individuals reproduce, however. Although the populations of both species appear to be stable over the past 33 years and not in immediate danger of extinction, the West Virginia Spring Salamander is still in critical conservation concern because of it\u27s extremely restricted distribution and current threats to the cave system it resides in

    Re-examination of the I-5 dust storm

    The infamous dust storm over the thanksgiving holiday of 1991 that led to loss of life from numerous automobile accidents on Interstate 5 (I-5) has been re-examined. Pauley et al. (1996) conducted an earlier investigation of this dust storm following the tenets of Danielsen's paradigm—a paradigm that links the tropopause fold phenomenon and a balanced thermally indirect circulation about the upper level jet stream. However, a cursory examination of mesoscale structures in the storm from the North American Regional Reanalysis (NARR) indicated evidence of a low-level unbalanced thermally direct circulation that demanded further investigation using a high-resolution Weather Research and Forecasting (WRF) model simulation. Principal results from the present study follow: (1) Although the model simulation showed evidence of a weak indirect circulation in the upper troposphere in support of the Danielsen's paradigm, the dynamic control of the storm stemmed from the lower tropospheric mesoscale response to geostrophic imbalance. (2) A lower tropospheric direct circulation led to mass/temperature adjustments that were confirmed by upper air observations at locations in proximity to the accident site, and (3) boundary layer deepening and destabilization due to these mesoscale processes pinpointed the timing and location of the dust storm. Although the present study does not underestimate the value of analyses that focus on the larger/synoptic scales of motion, it does bring to light the value of investigation that makes use of the mesoscale resources in order to clarify synoptic-mesoscale interactions
