DNS study of a pipe flow following a step increase in flow rate
Direct numerical simulation (DNS) is conducted to study the transient flow in a pipe following a near-step increase of flow rate from an initial turbulent flow. The results are compared with those of the transient flow in a channel reported in He and Seddighi (2013). It is shown that the flow again exhibits a laminar–turbulent transition, similar to that in a channel. The behaviours of the flow in a pipe and a channel are the same in the near-wall region, but there are significant differences in the centre of the flow. The correlation between the critical Reynolds number and free stream turbulence previously established for a channel flow has been shown to be applicable to the pipe flow. The responses of turbulent viscosity, vorticity Reynolds number, and budget terms are analysed. Some significant differences have been found to exist between the developments of the vorticity Reynolds number in the pipe and channel flows

Isosteviol Has Beneficial Effects on Palmitate-Induced α-Cell Dysfunction and Gene Expression
BACKGROUND: Long-term exposure to high levels of fatty acids impairs insulin secretion and exaggerates glucagon secretion. The aim of this study was to explore if the antihyperglycemic agent, Isosteviol (ISV), is able to counteract palmitate-induced α-cell dysfunction and to influence α-cell gene expression. METHODOLOGY/PRINCIPAL FINDINGS: Long-term incubation studies with clonal α-TC1-6 cells were performed in the presence of 0.5 mM palmitate with or without ISV. We investigated effects on glucagon secretion, glucagon content, cellular triglyceride (TG) content, cell proliferation, and expression of genes involved in controlling glucagon synthesis, fatty acid metabolism, and insulin signal transduction. Furthermore, we studied effects of ISV on palmitate-induced glucagon secretion from isolated mouse islets. Culturing α-cells for 72-h with 0.5 mM palmitate in the presence of 18 mM glucose resulted in a 56% (p<0.01) increase in glucagon secretion. Concomitantly, the TG content of α-cells increased by 78% (p<0.01) and cell proliferation decreased by 19% (p<0.05). At 18 mM glucose, ISV (10(-8) and 10(-6) M) reduced palmitate-stimulated glucagon release by 27% (p<0.05) and 27% (p<0.05), respectively. ISV (10(-6) M) also counteracted the palmitate-induced hypersecretion of glucagon in mouse islets. ISV (10(-6) M) reduced α-TC1-6 cell proliferation rate by 25% (p<0.05), but ISV (10(-8) and 10(-6) M) had no effect on TG content in the presence of palmitate. Palmitate (0.5 mM) increased Pcsk2 (p<0.001), Irs2 (p<0.001), Fasn (p<0.001), Srebf2 (p<0.001), Acaca (p<0.01), Pax6 (p<0.05) and Gcg mRNA expression (p<0.05). ISV significantly (p<0.05) up-regulated Insr, Irs1, Irs2, Pik3r1 and Akt1 gene expression in the presence of palmitate. CONCLUSIONS/SIGNIFICANCE: ISV counteracts α-cell hypersecretion and apparently contributes to changes in expression of key genes resulting from long-term exposure to palmitate. ISV apparently acts as a glucagonostatic drug with potential as a new anti-diabetic drug for the treatment of type 2 diabetes
Chemical features of Ganoderma polysaccharides with antioxidant, antitumor and antimicrobial activities
Review aricleGanoderma genus comprises one of the most commonly studied species worldwide, G.
lucidum. However, other Ganoderma species have been also reported as important
sources of bioactive compounds. Polysaccharides are important contributors to the
medicinal properties reported for Ganoderma species, as demonstrated by the numerous
publications, including reviews, on this matter. Yet, what are the chemical features of
Ganoderma polysaccharides that have bioactivity? In the present manuscript, the
chemical features of Ganoderma polysaccharides with reported antioxidant, antitumor
and antimicrobial activities (the most studied worldwide) are analyzed in detail. The
composition of sugars (homo- versus hetero-glucans and other polysaccharides), type of
glycosidic linkages, branching patterns, and linkage to proteins are discussed. Methods
for extraction, isolation and identification are evaluated and, finally, the bioactivity of
polysaccharidic extracts and purified compounds are discussed. The integration of data
allows deduction of structure-activity relationships and gives clues to the chemical
aspects involved in Ganoderma bioactivity
Mortality and life expectancy of Yokkaichi Asthma patients, Japan: Late effects of air pollution in 1960–70s
<p>Abstract</p> <p>Background</p> <p>The incidence of chronic obstructive pulmonary disease (COPD) and bronchial asthma began increasing in early 1960s in the population of Yokkaichi-city (Mie Prefecture, Japan). The cause of the disease was sulfur oxide air pollution, and it is known as Yokkaichi Asthma. The pollution markedly decreased by the end of 1970s; no new cases have been reported since 1988. This study aimed at examining the late effects of air pollution on the health of Yokkaichi Asthma patients.</p> <p>Methods</p> <p>Mortality rate and life expectancy of patients, registered between 1965 and 1988, were investigated from 1975 through 2000.</p> <p>Results</p> <p>Mortality rates for COPD and asthma in patients from Yokkaichi-city were significantly higher than in the whole population of Mie Prefecture. For all ages (except for males between 80 and 84 years in 1985), the life expectancy of both males and females were significantly reduced in patients from Yokkaichi-city as compared with the whole population of Mie Prefecture. The potential gains in life expectancy excluding the mortality for respiratory diseases including COPD and asthma were larger for all ages in patients from Yokkaichi-city.</p> <p>Conclusion</p> <p>Mortality and life expectancy were adversely affected in patients from Yokkaichi-city, despite the fact that the air pollution problem has been already solved.</p
