    Behaviour of Escherichia coli O157:H7 during the manufacture and ripening of an Italian traditional raw goat milk cheese

    Formagelle di capra is a raw goat cheese produced from whole chilled goat milk; traditional technology involving unpasteurised milk and indigenous lactic starter cultures is employed for its production in Italy. The purpose of this study was to assess the behavior of Escherichia coli O157:H7 during the manufacturing and ripening of this raw goat milk cheese. Raw milk was experimentally inoculated with E. coli O157:H7 in a laboratory scale plant and the count was monitored during production and 30 days of ripening required for this cheese. Results showed that E. coli O157:H7 count increased to more than 1.5 Log cfu g-1 during cheese production and remained constant until the end of ripening. The evidence that E. coli O157:H7 is able to survive during the manufacturing and ripening process suggests that the 30-day ripening period alone is insufficient to eliminate levels of viable E. coli O157:H7 in Formaggelle di capra cheese and that the presence of low numbers of E. coli O157:H7 in milk destined for the production of raw goat milk cheeses could represent a potential source of infection for humans and a threat for consumer

    Spontaneous magnetization of aluminum nanowires deposited on the NaCl(100) surface

    We investigate electronic structures of Al quantum wires, both unsupported and supported on the (100) NaCl surface, using the density-functional theory. We confirm that unsupported nanowires, constrained to be linear, show magnetization when elongated beyond the equilibrium length. Allowing ions to relax, the wires deform to zig-zag structures with lower magnetization but no dimerization occurs. When an Al wire is deposited on the NaCl surface, a zig-zag geometry emerges again. The magnetization changes moderately from that for the corresponding unsupported wire. We analyse the findings using electron band structures and simple model wires.Comment: submitted to PHys. Rev.

    Modulation of Barrier Properties of Monolayer Films from Blends of Polyethylene with Ethylene-co-Norbornene

    Flexible monolayer films with modulated diffusional and optical properties were achieved from blends of a LDPE/LLDPE matrix and an ethylene/norbornene copolymer (COC). A significant decrease of O2 and CO2 permeability was observed for cast films with COC content from 5 to 20 wt %. SEM analysis showed that, despite the incompatibility of the blend components, good dispersion and distribution of the COC domains in the PE matrix were achieved; as a consequence, for COC content ranging from 5 to 10 wt % the film transparency results practically unchanged. The UV light transmission decreased on increasing the percentage of COC, which constitutes also a protection against UV radiations. A noticeable improvement was observed in the mechanical properties: adding only 5 wt % of COC, the elastic modulus becomes five times higher than that of the PE matrix. The rheological behaviour of the blends keeps practically unchanged with respect to the matrix, allowing to use the typical LDPE processing conditions. Sustainable inexpensive flexible films answering the packaging requirements of specific categories of food were achieved

    Behavior of listeria innocua during the manufacturing and pit-ripening of formaggio di fossa di sogliano pdo cheese

    Formaggio di Fossa di Sogliano is a traditional Italian Protected Designation of Origin (PDO) cheese ripened for a minimum of 5 months, with the feature of a ripening of at least 80 to at most 100 days in pits, digged into tuffaceous rocks according to medieval tradition of Italy. In this study, a challenge test using Listeria innocua as a surrogate of Listeria monocytogenes was performed, with the aim of increasing knowledge concerning the impact of the Fossa cheese process, and especially of the traditional ripening process of this PDO, on the behaviourof L. monocytogenes. Pasteurized milk was experimentally inoculated with 4.5 log CFU/mL cocktail by three L. innocua strains, and L. innocua and Mesophilic Lactic Acid Bacteria (LAB) counts as well as the evolution of temperatures, pH and aw values were monitored throughout the manufacturing and ripening processes. Throughout the ripening in maturation room a constant temperature of 8\ub0C was observed reaching a temperature between 10 and 15.5\ub0C during ripening into pit. In the final products data for LAB concentration, pH and aw values were roughly in accordance with literature, even if some differences were, probably due to variability of artisanal cheese productions. The numbers of L. innocua showed a slight decrease but remained stable until the end of ripening in maturation room, whereas a significant reduction of the microorganism was observed in the final product, at the end of the ripening into the pit. The findings give scientific evidence that the process of this PDO prevented the L. innocua growth, allowing us to speculate a similar behaviour of L. monocytogenes. Based on this study, the recommendation to extend as much as possible the ripening into pit (from 80 to 100 days) was provided to food business operators as a risk mitigation strategy to be implemented

    LDPE-based blends and films stabilized with nonreleasing polymeric antioxidants for safer food packaging

    Several novel random copolymers of ethylene and 1-olefin counits bearing a highly efficient phenolic antioxidant moiety placed at different distances from the polymerizable double bond were prepared in the presence of a metallocene catalyst. These copolymers were melt-blended with an antioxidant-free LDPE in an internal batch mixer to obtain innovative materials containing nonreleasing polymeric antioxidants suitable for safer food packaging applications. Blends and films, obtained by compression molding, were tested for their thermal and thermo-oxidative stability by thermogravimetric analysis both in dynamic and isothermal conditions. Films containing the macromolecular antioxidants showed a longer induction time before O2 uptake starts and, consequently, a higher degradation temperature than neat LDPE or LDPE containing a low molecular weight commercial additive. Aging tests demonstrated that the new polymeric antioxidants also exert a valid protection against photo-oxidation. Eventually, migration tests demonstrated the absence of any trace of products containing the antioxidant moiety when the films were kept in contact with a food simulant

    Assessment of the antibiotic resistance profile, genetic heterogeneity and biofilm production of methicillin-resistant staphylococcus aureus (MRSA) isolated from the italian swine production chain

    The main aim of the present study was to evaluate the level of antibiotic resistance, prevalence and virulence features of methicillin-resistant Staphylococcus aureus (MRSA) isolated from heavy swine at abattoir level and farming environments in Lombardy (Northern Italy). With this scope, 88 different heavy swine farms were surveyed, obtaining a total of n = 440 animal swabs and n = 150 environmental swabs. A total of n = 87 MRSA isolates were obtained, with an overall MRSA incidence of 17.50% (n = 77) among animal samples and a 6.67% (n = 10) among environmental. Molecular characterisation using multilocus sequence typing (MLST) plus spa-typing showed that sequence type ST398/t899 and ST398/t011 were the most commonly isolated genotypes, although other relevant sequence types such as ST1 or ST97 were also found. A lack of susceptibility to penicillins, tetracycline and ceftiofur was detected in >91.95, 85.05 and 48.28% of the isolates, respectively. Resistance to doxycycline (32.18%), enrofloxacin (27.59%) and gentamicin (25.29%) was also observed. Additionally, a remarkable level of antibiotic multiresistance (AMR) was observed representing a 77.01% (n = 67) of the obtained isolates. Genetic analysis revealed that 97.70% and 77.01% of the isolates harboured at least one antibiotic resistance or enterotoxin gene, respectively, pointing out a high isolate virulence potential. Lastly, 55.17% (n = 48) were able to produce measurable amounts of biofilm after 24 h. In spite of the current programmes for antibiotic reduction in intensively farming, a still on-going high level of AMR and virulence potential in MRSA was demonstrated, making this pathogen a serious risk in swine production chain, highlighting once more the need to develop efficient, pathogen-specific control strategies

    Standard and Light-Cycler PCR methods for animal DNA species detection in animal feedstuffs

    In this work four species-specific primers and probes were designed and evaluated for the detection and quantification of bovine, ovine, swine and chicken mitochondrial DNA in feeds. PCR primers were optimized using conventional and Real Time PCR, to detect short species-specific sequences amplifiable from heat treated material. Both methods confirmed the high specificity of the primers designed. Real time quantitative PCR assay allowed the detection of as few as 0.01 ng and 0.05 ng of ovine and bovine genomic DNA, respectively. The detection limit for swine and chicken genomic DNA was 0.5 ng. Sensitivity levels observed in DNA extracted from meat samples processed according to EU legislation were different compared to those in genomic DNAs previously described. They resulted in swine 5 fg of MBM DNA, in chicken 25 ng, in ovine and bovine 50 ng. We confirmed the efficiency and specificity of primers in RT-PCR to detect 0.5% of bovine, ovine, swine and chicken MBM in contaminated feedstuffs. (C) 2007 Elsevier Ltd. All rights reserved

    Novel Ethylene=Norbornene Copolymers as Nonreleasing Antioxidants for Food-Contact Polyolefinic Materials

    An efficient procedure for the copolymerization of ethylene (E) with a novel norbornenic comonomer (NArOH) bearing a stabilizing moiety analogous to commercial antioxidant 2,6-di-tert-butyl-4-methylphenol (BHT) is successfully developed. This study is aimed at: i) tuning the concentration of the stabilizing function along the polymer chain, and ii) preparing \u201cnonreleasing\u201d polymeric additives specifically destined to protect commercial low-density polyethylene (LDPE). Films obtained from blends of the novel E=NArOH copolymers with an antioxidant-free LDPE matrix are characterized by superior thermal, thermo-oxidative, and photostability when compared not only with neat LDPE films but also with films stabilized by the commercial BHT additive. Specific migration tests conducted in order to investigate the nonreleasing character of the novel macromolecular additives confirm the reduced risk of migration, from the films into food simulants, of unreacted comonomer or degradation products bearing the antioxidant moiety

    Identification of a major Listeria monocytogenes outbreak clone linked to soft cheese in Northern Italy - 2009-2011

    Background: Molecular subtyping and enhanced surveillance in Lombardy region identified a cluster of possibly related listeriosis cases from 2006 to 2010. This cluster grouped 31 isolates that belonged to serotype 1/2a and Sequence Type 38 (ST38) as defined by Multilocus Sequence Typing (MLST). Methods: Our study expanded the previous investigation to include cases from 2011 to 2014 and used Multi-Virulence- Locus Sequence Typing (MVLST) on all ST38 isolates to better understand their epidemiology and possibly identify a common source outbreak. Results: Out of 306 L. monocytogenes clinical isolates collected, 43 (14.1%) belonged to ST38 with cases occurring in nine out of twelve Lombardy provinces. The ST38 isolates were split by MVLST into two Virulence Types (VTs): VT80 (n = 12) and VT104 (n = 31). VT104 cases were concentrated between 2009 and 2011 in two provinces, Bergamo and Milan. An epidemiologic investigation was performed and in one case, a matching VT104 isolate was retrieved from a soft cheese sample from a patient's refrigerator. Conclusions: Our findings revealed a major listeriosis outbreak in Northern Italy linked to soft cheese in 20092011, which went undetected by local health authorities. Our study shows that integrating subtyping methods with conventional epidemiology can help identify the source of L. monocytogenes outbreak clones
