Nonlinear ion-acoustic structures in a nonextensive electron–positron–ion–dust plasma: Modulational instability and rogue waves
The nonlinear propagation of planar and nonplanar (cylindrical and spherical) ion-acoustic waves in an unmagnetized electron–positron–ion–dust plasma with two-electron temperature distributions is investigated in the context of the nonextensive statistics. Using the reductive perturbation method, a modified nonlinear Schrödinger equation is derived for the potential wave amplitude. The effects of plasma parameters on the modulational instability of ion-acoustic waves are discussed in detail for planar as well as for cylindrical and spherical geometries. In addition, for the planar case, we analyze how the plasma parameters influence the nonlinear structures of the first- and second-order ion-acoustic rogue waves within the modulational instability region. The present results may be helpful in providing a good fit between the theoretical analysis and real applications in future spatial observations and laboratory plasma experiments
Exposure to excess insulin (glargine) induces type 2 diabetes mellitus in mice fed on a chow diet
We have previously shown that insulin plays an important role in the nutrient-induced insulin resistance. In this study, we tested the hypothesis that chronic exposure to excess long-acting insulin (glargine) can cause typical type 2 diabetes mellitus (T2DM) in normal mice fed on a chow diet. C57BL/6 mice were treated with glargine once a day for 8 weeks, followed by evaluations of food intake, body weight, blood levels of glucose, insulin, lipids, and cytokines, insulin signaling, histology of pancreas, ectopic fat accumulation, oxidative stress level, and cholesterol content in mitochondria in tissues. Cholesterol content in mitochondria and its association with oxidative stress in cultured hepatocytes and β-cells were also examined. Results show that chronic exposure to glargine caused insulin resistance, hyperinsulinemia, and relative insulin deficiency (T2DM). Treatment with excess glargine led to loss of pancreatic islets, ectopic fat accumulation in liver, oxidative stress in liver and pancreas, and increased cholesterol content in mitochondria of liver and pancreas. Prolonged exposure of cultured primary hepatocytes and HIT-TI5 β-cells to insulin induced oxidative stress in a cholesterol synthesis-dependent manner. Together, our results show that chronic exposure to excess insulin can induce typical T2DM in normal mice fed on a chow diet
Gene expression changes induced by the tumorigenic pyrrolizidine alkaloid riddelliine in liver of Big Blue rats

Abstract

Background

Pyrrolizidine alkaloids (PAs) are probably the most common plant constituents that poison livestock, wildlife, and humans worldwide. Riddelliine is isolated from plants grown in the western United States and is a prototype of genotoxic PAs. Riddelliine was used to investigate the genotoxic effects of PAs via analysis of gene expression in the target tissue of rats in this study. Previously we observed that the mutant frequency in the liver of rats gavaged with riddelliine was 3-fold higher than that in the control group. Molecular analysis of the mutants indicated that there was a statistically significant difference between the mutational spectra from riddelliine-treated and control rats.

Results

Riddelliine-induced gene expression profiles in livers of Big Blue transgenic rats were determined. The female rats were gavaged with riddelliine at a dose of 1 mg/kg body weight 5 days a week for 12 weeks. Rat whole genome microarray was used to perform genome-wide gene expression studies. When a cutoff value of a two-fold change and a <it>P</it>-value less than 0.01 were used as gene selection criteria, 919 genes were identified as differentially expressed in riddelliine-treated rats compared to the control animals. By analysis with the Ingenuity Pathway Analysis Network, we found that these significantly changed genes were mainly involved in cancer, cell death, tissue development, cellular movement, tissue morphology, cell-to-cell signaling and interaction, and cellular growth and proliferation. We further analyzed the genes involved in metabolism, injury of endothelial cells, liver abnormalities, and cancer development in detail.

Conclusion

The alterations in gene expression were directly related to the pathological outcomes reported previously. These results provided further insight into the mechanisms involved in toxicity and carcinogenesis after exposure to riddelliine, and permitted us to investigate the interaction of gene products inside the signaling networks.</p
A Hydrodynamic and Transport Model for Ultrarelativistic Heavy Ion Collisions at RHIC and LHC Energies
Combining the hydrodynamic model (Hydro code) and the transport model (PACIAE
model), we present the Hydro-PACIAE hybrid model. We use the Hydro-PACIAE
hybrid model to calculate Au+Au collisions at =130 GeV and Pb+Pb
collisions at =2.76 TeV. The obtained pseudo-rapidity and
transverse momentum distributions well reproduce the experimental data. This
shows that the Hydro-PACIAE hybrid model is useful to describe the
ultrarelativistic Heavy Ion Collisions at RHIC and LHC Energies. We used the
hybrid model to calculate the elliptical flow for the Pb+Pb collisions at
=2.76 TeV. The values are bigger than the experimental
data at high , and will become bigger when the Hydro evolution time is
longer. The results indicate that the selection of the transition hypersurface
has significant effect on the observable quantities.Comment: 8 pages, 8 figure

The Reproducibility of Lists of Differentially Expressed Genes in Microarray Studies
Reproducibility is a fundamental requirement in scientific experiments and clinical contexts. Recent publications raise concerns about the reliability of microarray technology because of the apparent lack of agreement between lists of differentially expressed genes (DEGs). In this study we demonstrate that (1) such discordance may stem from ranking and selecting DEGs solely by statistical significance (P) derived from widely used simple t-tests; (2) when fold change (FC) is used as the ranking criterion, the lists become much more reproducible, especially when fewer genes are selected; and (3) the instability of short DEG lists based on P cutoffs is an expected mathematical consequence of the high variability of the t-values. We recommend the use of FC ranking plus a non-stringent P cutoff as a baseline practice in order to generate more reproducible DEG lists. The FC criterion enhances reproducibility while the P criterion balances sensitivity and specificity
Fractal Reconnection in Solar and Stellar Environments
Recent space based observations of the Sun revealed that magnetic
reconnection is ubiquitous in the solar atmosphere, ranging from small scale
reconnection (observed as nanoflares) to large scale one (observed as long
duration flares or giant arcades). Often the magnetic reconnection events are
associated with mass ejections or jets, which seem to be closely related to
multiple plasmoid ejections from fractal current sheet. The bursty radio and
hard X-ray emissions from flares also suggest the fractal reconnection and
associated particle acceleration. We shall discuss recent observations and
theories related to the plasmoid-induced-reconnection and the fractal
reconnection in solar flares, and their implication to reconnection physics and
particle acceleration. Recent findings of many superflares on solar type stars
that has extended the applicability of the fractal reconnection model of solar
flares to much a wider parameter space suitable for stellar flares are also
discussed.Comment: Invited chapter to appear in "Magnetic Reconnection: Concepts and
