    Dioctadecyldimethylammonium:monoolein nanocarriers for efficient in vitro gene silencing

    This study describes a novel liposomal formulation for siRNA delivery, based on the mixture of the neutral lipid monoolein (MO) and cationic lipids of the dioctadecyldimethylammonium (DODA) family. The cationic lipids dioctadecyldimethylammonium bromide (DODAB) and chloride (DODAC) were compared in order to identify which one will most efficiently induce gene silencing. MO has a fluidizing effect on DODAC and DODAB liposomes, although it was more homogeneously distributed in DODAC bilayers. All MO-based liposomal formulations were able to efficiently encapsulate siRNA. Stable lipoplexes of small size (100-160 nm) with a positive surface charge (>+45 mV) were formed. A more uniform MO incorporation in DODAC:MO may explain an increase of the fusogenic potential of these liposomes. The siRNA-lipoplexes were readily internalized by human nonsmall cell lung carcinoma (H1299) cells, in an energy dependent process. DODAB:MO nanocarriers showed a higher internalization efficiency in comparison to DODAC:MO lipoplexes, and were also more efficient in promoting gene silencing. MO had a similar gene silencing ability as the commonly used helper lipid 1,2-dioleyl-3-phosphatidylethanolamine (DOPE), but with much lower cytotoxicity.     Production of Composite Steel Strips on Continuous Casting and Deformation Assembly

    Abstract: This article describes the resource-saving assembly of continuous casting and deformation for production of three-layer bimetallic strips based on dissimilar steels and alloys. The results describe the theoretical analysis of the metal’s stress and strain state in the deformation zone upon bimetal strip production by sequential combining the layer thickness in the form of steel grades 09G2S–13KhFA–09G2S. Metallographic studies of bimetal structure and hardness along the distance of cyclic deformation zone in various sheet thickness areas are described. © 2019, Allerton Press, Inc

    Influence of long-term antibiotic therapy on gut microbiome composition and metabolic profile in pulmonary tuberculosis

    The use of long-term multicomponent antibiotic therapy is the most effective way to treat tuberculosis (TB). However, little is known about the effect of this chemotherapy on the human intestinal microflora. The purpose of this study was to analyze an effect of long-term antibiotic therapy on gut microbiome composition and metabolic profile in TB patients. We used deep sequencing of fecal samples from 23 treatment-naive TB patients to reconstruct the metabolic capacity and strain/species-level abundance in the gut microbiome. Two fecal samples were obtained from each patient: before and after treatment. We showed that TB treatment regimen does not disrupt the overall diversity of the gut microbiome but does have an impact on gut bacterial microbiome composition and metabolic profile. While taking first-line anti-tuberculosis drugs (isoniazid, rifampicin, ethambutol, pyrazinamide), TB patients showed an apparent increase in Actinobacteria abundance. Pairwise comparison of metagenomic data revealed 28 differentially represented bacterial taxa, of which three species Bacteroides cellulosilyticus, Enterocloster aldensis, Clostridium spiroforme were strongly enriched in TB patients post-chemotherapy, whereas 25 species were enriched in TB patients before treatment (Bifidobacterium catenulatum, Enterococcus faecium, Bacteroides salyersiae, Bacteroides xylanisolvens, Bacteroides eggerthii, Lachnospira eligens, Akkermansia muciniphila, Ruminococcus lactaris, etc.) (p 0.05). The metabolic profile of the gut microbiome was characterized by increased metabolic processes aimed at the growth and division of microbial cells. Iron is the main limiting factor for growth and reproduction. In addition, it is important to note the prevalence of glycolysis and lactate fermentation as the major means for energy production by intestinal microbiota

    The assessment of lacrimal film condition in patients with dry eye syndrome during therapy

    To evaluate the efficiency of modern imaging methods of meibomian gland ducts and determine the tear film break-up time (TFBUT) during therapy in patients with the secondary dry eye syndrome (SDES) of allergic etiology.Material and methods. 66 patients aged 38.0 ± 1.5 with SDES after allergic conjunctivitis were examined. The patients were divided into 2 groups depending on the therapy administered. The main group (n = 33) received Oftalron as tear replacement therapy for 20 days, 2 drops, 2 times a day. The patients of the comparison group (n = 33) used Super Optic HydroBalance for 20 days, 2 drops 2 times a day. The control group consisted of 25 healthy subjects without eye pathology. All subjects underwent Schirmer’s tests I and II, as well as TFBUT and meibography on an automatic refractokeratometer HRK-9000A (Huvitz, Korea).Results. In both SDES groups, a significant decrease of the OSDI (Ocular Surface Disease Index) in the treatment process was observed (p ≤ 0.05), which corresponded to low intensity of SDES symptoms. In both groups morphofunctional changes of meibomian glands of 1 and 2 stages prevailed. The most significant dynamics was observed in TFBUT results, they were significantly lower in comparison with the similar indexes in the patients of the control group.Conclusion. Specific modes of autorefractokeratometer HRK-9000A allow receiving objective information about the condition of the tear film and high-quality imaging of meibomian glands, which allow expanding possibilities of complex diagnostics of various forms of DES
