    The most important thing for the company to determines financial performance in the matter of financial report is to acknowledge the outcome of financial performance in order to compete in the economic development to maintain study performance of the company. In order to evaluate the performance, the calculation using profitability ratio and achieves Gross Profit Margin Net Profit Margin every year advance, because every year income experience increase. The analysis score over Return on Asset/ Return on Investment instability, because to the increase in profit in the third year is not to large compared with the previous year. For Return on Equity decreased, because income earned each year not by the equity every year, because despite rising profit, rising equity is greater than the profit. On the Profit Margin and Economical Rentability has increased, as earnings before interest and taxes in the financial statements in the financial statements on income (loss) from operations increased, and that the company\u27s revenue underwriting income indicates that the increase is happening and therefore contributes to the increase in Profit Margin and Economical Rentability every year. And the outcome of above analysis can be sean on PT. Jasa Raharja (Persero) financial performance can be said well achieved, because there are fluctuation in every year. Dispite on the RoA and RoE having a bit of downside, and the company has to increase RoA and RoE in order for having eficienly increasment. However the overall company financial performance having a well forward achievement every year with the increasment of company profit

    Forest as Stronghold of Local Ecological Practice: Currently Used Wild Food Plants in Polesia, Northern Ukraine

    Local ecological practice (LEP, e.g., the everyday practice of collecting and using plants, including wild food plants) is shaped by nature (available local resources) and culture (local perceptions and knowledge on their usability), including a multitude of factors, among which language and geographical or cultural separation have been found to play crucial roles in affecting biocultural diversity. Also, proximity to the forest has been shown to increase the use of plants. We conducted ethnobotanical fieldwork within eastern and western regions of Ukrainian Polesia, during which we interviewed 118 people. Through semi-structured interviews, we recorded the distribution of the current uses of 70 wild food taxa. The analysis of use records revealed homogeneous distribution of use despite the geographical distance and different spoken dialects; however, we were able to single out the highly sylvan region of eastern Polesia as the area with highest biocultural diversity for the use of wild food plants. The results suggest that in the context of the overall homogenization of local ecological knowledge, the continued existence of unintended contact with nature through living and working in the forest may be the primary factor maintaining the broader LEP in the sylvan area of eastern Polesia. Місцева екологічна практика (MЕП, що включає повсякденну практику збору та використання дикорослих рослин у їжу) формується природою (місцевими ресурсами) та культурою (місцеві знання про їх використання), що включає безліч чинників, серед яких мова та географічне або культурне відокремлення відіграють вирішальну роль у впливі на біокультурне різноманіття. Також показано, що близькість до лісу збільшує використання рослин. Ми проводили етноботанічні польові роботи у двох областях Українського Полісся, в ході яких ми провели інтерв’ю з 118 людьми. За допомогою напівструктурованих інтерв’ю ми задокументували поточне використання 70 таксонів дикорослих рослин у їжу. Аналіз записів про вживання свідчить про однорідний розподіл використання, незважаючи на географічне розташування та різні розмовні діалекти; однак, ми змогли виділити лісистий регіон Східного Полісся як територію з найвищим біокультурним різноманіттям використання дикорослих рослин уїжу. Результати свідчать, що в контексті загальної гомогенізації місцевих екологічних знань, продовження існування контакту з природою живучи та працюючи в лісі може бути основним чинником, який підтримує ширшу MЕПу в лісовій зоні Східного Полісся

    Liver Is Able to Activate Naïve CD8+ T Cells with Dysfunctional Anti-Viral Activity in the Murine System

    The liver possesses distinct tolerogenic properties because of continuous exposure to bacterial constituents and nonpathogenic food antigen. The central immune mediators required for the generation of effective immune responses in the liver environment have not been fully elucidated. In this report, we demonstrate that the liver can indeed support effector CD8+ T cells during adenovirus infection when the T cells are primed in secondary lymphoid tissues. In contrast, when viral antigen is delivered predominantly to the liver via intravenous (IV) adenovirus infection, intrahepatic CD8+ T cells are significantly impaired in their ability to produce inflammatory cytokines and lyse target cells. Additionally, intrahepatic CD8+ T cells generated during IV adenovirus infection express elevated levels of PD-1. Notably, lower doses of adenovirus infection do not rescue the impaired effector function of intrahepatic CD8+ T cell responses. Instead, intrahepatic antigen recognition limits the generation of potent anti-viral responses at both priming and effector stages of the CD8+ T cell response and accounts for the dysfunctional CD8+ T cell response observed during IV adenovirus infection. These results also implicate that manipulation of antigen delivery will facilitate the design of improved vaccination strategies to persistent viral infection

    Of the importance of a leaf: the ethnobotany of sarma in Turkey and the Balkans

    BACKGROUND: Sarma - cooked leaves rolled around a filling made from rice and/or minced meat, possibly vegetables and seasoning plants - represents one of the most widespread feasting dishes of the Middle Eastern and South-Eastern European cuisines. Although cabbage and grape vine sarma is well-known worldwide, the use of alternative plant leaves remains largely unexplored. The aim of this research was to document all of the botanical taxa whose leaves are used for preparing sarma in the folk cuisines of Turkey and the Balkans. Methods: Field studies were conducted during broader ethnobotanical surveys, as well as during ad-hoc investigations between the years 2011 and 2014 that included diverse rural communities in Croatia, Bosnia and Herzegovina, Serbia, Kosovo, Albania, Macedonia, Bulgaria, Romania, and Turkey. Primary ethnobotanical and folkloric literatures in each country were also considered. Results: Eighty-seven botanical taxa, mainly wild, belonging to 50 genera and 27 families, were found to represent the bio-cultural heritage of sarma in Turkey and the Balkans. The greatest plant biodiversity in sarma was found in Turkey and, to less extent, in Bulgaria and Romania. The most commonly used leaves for preparing sarma were those of cabbage (both fresh and lacto-fermented), grape vine, beet, dock, sorrel, horseradish, lime tree, bean, and spinach. In a few cases, the leaves of endemic species (Centaurea haradjianii, Rumex gracilescens, and R. olympicus in Turkey) were recorded. Other uncommon sarma preparations were based on lightly toxic taxa, such as potato leaves in NE Albania, leaves of Arum, Convolvulus, and Smilax species in Turkey, of Phytolacca americana in Macedonia, and of Tussilago farfara in diverse countries. Moreover, the use of leaves of the introduced species Reynoutria japonica in Romania, Colocasia esculenta in Turkey, and Phytolacca americana in Macedonia shows the dynamic nature of folk cuisines. Conclusion: The rich ethnobotanical diversity of sarma confirms the urgent need to record folk culinary plant knowledge. The results presented here can be implemented into initiatives aimed at re-evaluating folk cuisines and niche food markets based on local neglected ingredients, and possibly also to foster trajectories of the avant-garde cuisines inspired by ethnobotanical knowledge

    Synthetic Nanoparticles for Vaccines and Immunotherapy

    The immune system plays a critical role in our health. No other component of human physiology plays a decisive role in as diverse an array of maladies, from deadly diseases with which we are all familiar to equally terrible esoteric conditions: HIV, malaria, pneumococcal and influenza infections; cancer; atherosclerosis; autoimmune diseases such as lupus, diabetes, and multiple sclerosis. The importance of understanding the function of the immune system and learning how to modulate immunity to protect against or treat disease thus cannot be overstated. Fortunately, we are entering an exciting era where the science of immunology is defining pathways for the rational manipulation of the immune system at the cellular and molecular level, and this understanding is leading to dramatic advances in the clinic that are transforming the future of medicine.1,2 These initial advances are being made primarily through biologic drugs– recombinant proteins (especially antibodies) or patient-derived cell therapies– but exciting data from preclinical studies suggest that a marriage of approaches based in biotechnology with the materials science and chemistry of nanomaterials, especially nanoparticles, could enable more effective and safer immune engineering strategies.     Class-B CpG-ODN Formulated With a Nanostructure Induces Type I Interferons-Dependent and CD4+ T Cell-Independent CD8+ T-Cell Response Against Unconjugated Protein Antigen

    There is a need for new vaccine adjuvant strategies that offer both vigorous antibody and T-cell mediated protection to combat difficult intracellular pathogens and cancer. To this aim, we formulated class-B synthetic oligodeoxynucleotide containing unmethylated cytosine-guanine motifs (CpG-ODN) with a nanostructure (Coa-ASC16 or coagel) formed by self-assembly of 6-0-ascorbyl palmitate ester. Our previous results demonstrated that mice immunized with ovalbumin (OVA) and CpG-ODN formulated with Coa-ASC16 (OVA/CpG-ODN/Coa-ASC16) elicited strong antibodies (IgG1 and IgG2a) and Th1/Th17 cellular responses without toxic systemic effects. These responses were superior to those induced by a solution of OVA with CpG-ODN or OVA/CpG-ODN formulated with aluminum salts. In this study, we investigated the capacity of this adjuvant strategy (CpG-ODN/Coa-ASC16) to elicit CD8+ T-cell response and some of the underlying cellular and molecular mechanisms involved in adaptive response. We also analyzed whether this adjuvant strategy allows a switch from an immunization scheme of three-doses to one of single-dose. Our results demonstrated that vaccination with OVA/CpG-ODN/Coa-ASC16 elicited an antigen-specific long-lasting humoral response and importantly-high quality CD8+ T-cell immunity with a single-dose immunization. Moreover, Coa-ASC16 promoted co-uptake of OVA and CpG-ODN by dendritic cells. The CD8+ T-cell response induced by OVA/CpG-ODN/Coa-ASC16 was dependent of type I interferons and independent of CD4+ T-cells, and showed polyfunctionality and efficiency against an intracellular pathogen. Furthermore, the cellular and humoral responses elicited by the nanostructured formulation were IL-6-independent. This system provides a simple and inexpensive adjuvant strategy with great potential for future rationally designed vaccines

    Influence of Magnafloc 919 flocculant dosage on centrifugal filtration of post-decantation dump suspension

    Niniejszy artykuł jest kontynuacją cyklu prac Palicy i współpracowników dotyczących separacji fazowej zawiesin zrzutowych po wirówkach DECANTER, na których odwadnia się flotokoncentraty węgli kamiennych [1÷12]. Wiele prac w tej problematyce począwszy od lat 70-tych prowadził Piecuch i współpracownicy.In the paper experimental results dealing with centrifugal filtration of postdecantation dump suspension after the DECANTER centrifuge and obtained in a coal mine of "Jastrzębska Spółka Węglowa". Magnaflog-919 flocculant, used by operating service, was added to suspension. The suspension is characterized by very fine grainsize increasing during subsequent cycles, what leads, in consequence, to its replacement. This is connected not only with water and deposition costs but also with the losses of solid phase which is about 81% coal. In the mentioned coal mine 30 g of dry Magnaflog-919 flocculant per 1 Mg of dry product is added to flotoconcentrate. Floccu lant is dosed in a form of 0.1% gel. Therefore it appears a question if during dewatering of dump suspension containing 7÷8% of solid phase (using e.g. centrifugal or pressure filtration) flocculant should be again added, and if an answer is positive - in what amount? As a measure of dewatering efficiency, the final moisture content in cake obtained during centrifugal filtration for defined parameters of the laboratory centrifuge (adapted to a filtration centrifuge) was assumed. The test were carried out in the range of multiplication coefficient ? = a/g = 251.3÷1786.8 and centrifugal time ? = 280÷1800 s for dump suspension containing a base flocculant dose of 30 g/1 Mg, and doses of 15 and 60 g/1 Mg. Each experimental run consists of 6 values of ? and 7 of ? (altogether 42x4 points). For each experimental run tri-parameter correlations in a form of w = f(a/g, ?) = exp[A(a/g)b ?c] + D with regression coefficient R2>0.983 were elaborated. It should be noted that however the correlations quite well describe the process they have no theoretical bases. It was stated that the Magnaflog-919 flocculant dosage in the range 15÷60 g/1 Mg of dry product, for the same a/g and ?, produced an increase of moisture content in cake after centrifugation in comparison with values obtained for dump suspension. This, rather unexpected effect, can be explained by disruption of large flocs during decantation in DECANTER centrifuges and deposition of filtrate in smaller fragments or newly created flocs, for which centrifugal forces were not sufficiently high to remove it from cake. It was proved by higher values of moisture content in cake after centrifugal filtration in comparison with pressure filtration with cake compression. It was also stated the a good usability of filter cloth PT 912 for filtration, while an additive of flocculant accelerated sedimentation. Taking these into account, it seems that dewatering of dump suspension should be carried on in two stages. In the first stage suspension (with 30 g of flocculant per 1 Mg) should be directed to a settling tank, clarified liquid from this apparatus should be used in recirculation. In the second stage post-sedimentation slurry should be directed to a mixing process unit with flotoconcentrate, situated before DECANTER centrifuges
