    Environment-sensitive mass changes influence breeding frequency in a capital breeding marine top predator

The trade‐off between survival and reproduction in resource‐limited iteroparous animals can result in some individuals missing some breeding opportunities. In practice, even with the best observation regimes, deciding whether ‘missed’ years represent real pauses in breeding or failures to detect breeding can be difficult, posing problems for the estimation of individual reproductive output and overall population fecundity. 2. We corrected fecundity estimates by determining whether breeding had occurred in skipped years, using long‐term capture–recapture observation datasets with parallel longitudinal mass measurements, based on informative underlying relationships between individuals’ mass, breeding status and environmental drivers in a capital breeding phocid, the grey seal. 3. Bayesian modelling considered interacting processes jointly: temporal changes in a phenotypic covariate (mass); relationship of mass to breeding probability; effects of maternal breeding state and mark type on resighting. Full reproductive histories were imputed, with the status of unobserved animals estimated as breeding or non‐breeding, accounting for local environmental variation. Overall fecundity was then derived for Scottish breeding colonies with contrasting pup production trends. 4. Maternal mass affected breeding likelihood. Mothers with low body mass at the end of breeding were less likely to bear a pup the following year. Successive breeding episodes incurred a cost in reduced body mass which was more pronounced for North Rona, Outer Hebrides (NR) mothers. Skipping breeding increased subsequent pupping probability substantially for low mass females. Poor environmental conditions were associated with declines in breeding probability at both colonies. Seal mass gain between breeding seasons was (a) negatively associated with lagged North Atlantic Oscillation for seals at NR and (b) positively associated with an index of seal prey (Ammodytes spp) abundance at Isle of May, Firth of Forth (IM). Overall fecundity was marginally greater at IM (increasing/stable pup production) than at NR (decreasing). No effects of mass were detected on maternal survival. 5. Skipping breeding in female grey seals appears to be an individual mass‐dependent constraint moderated by previous reproductive output and local environmental conditions. Different demographic trends at breeding colonies were consistent with the fecundities estimated using this method, which is general and adaptable to other situations.PostprintPeer reviewe

    Water resource assessment of karst islands and the development of a freshwater lens assessment protocol

    Karst islands like those found in the Philippine Archipelago present extreme challenges for stakeholders to manage their water resources in a sustainable manner. Anthropogenic Climate Change, land development, point source pollution and increased population have all combined to alter the water balance on these fragile islands. Karst features, shallow depth to groundwater and the potential for dissolution, contribute greatly to these challenges. Combined, these factors pose an array of complex research questions. A field reconnaissance and semi-structured interviews were completed on Bantayan Island in 2016. Drawing upon current groundwater characterization practices the research posits a novel approach for these hydrogeologic environs. With the application of the Freshwater Lens Assessment Protocol (FLAP) credible output for the characterization of the islands groundwater resources will be determined and integrated into an Adaptive Water Resource Management framework, empowering stakeholders to make informed decisions on sustainable abstraction strategies

    Identification of lead chemotherapeutic agents from medicinal plants against blood flukes and whipworms

    Schistosomiasis and trichuriasis are two of the most common neglected tropical diseases (NTD) that affect almost a billion people worldwide. There is only limited number of effective drugs to combat these NTD. Medicinal plants are a viable source of parasiticides. In this study, we have investigated six of the 19 phytochemicals isolated from two Bhutanese medicinal plants, Corydalis crispa and Pleurospermum amabile, for their anthelmintic properties. We used xWORM technique and Scanning Electron Microscope-based imaging to determine the activity of the compounds. Of the six compounds tested, isomyristicin and bergapten showed significant anthelmintic activity against Schistosoma mansoni and Trichuris muris with bergapten being the most efficacious one against both parasites (S. mansoni IC50 = 8.6 ÎŒg/mL and T. muris IC50 = 10.6 ÎŒg/mL) and also against schistosomula stage of S. mansoni. These two compounds induced tegumental damage to S. mansoni and affected the cuticle, bacillary bands and bacillary glands of T. muris. The efficacy against multiple phylogenetically distinct parasites and different life stages, especially the schistosomulum where praziquantel is ineffective, makes isomyristicin and bergapten novel scaffolds for broad-spectrum anthelmintic drug development that could be used for the control of helminths infecting humans and animals

    Excretory/secretory products of the carcinogenic liver fluke are endocytosed by human cholangiocytes and drive cell proliferation and IL6 production

Liver fluke infection caused by Opisthorchis viverrini remains a major public health problem in many parts of Asia including Thailand, Lao PDR, Vietnam and Cambodia, where there is a strikingly high incidence of cholangiocarcinoma (CCA - hepatic cancer of the bile duct epithelium). Among other factors, uptake of O. viverrini excretory/secretory products (OvES) by biliary epithelial cells has been postulated to be responsible for chronic inflammation and proliferation of cholangiocytes, but the mechanisms by which cells internalise O. viverrini excretory/secretory products are still unknown. Herein we incubated normal human cholangiocytes (H69), human cholangiocarcinoma cells (KKU-100, KKU-M156) and human colon cancer (Caco-2) cells with O. viverrini excretory/secretory products and analysed the effects of different endocytic inhibitors to address the mechanism of cellular uptake of ES proteins. Opisthorchis viverrini excretory/secretory products was internalised preferentially by liver cell lines, and most efficiently/rapidly by H69 cells. There was no evidence for trafficking of ES proteins to cholangiocyte organelles, and most of the fluorescence was detected in the cytoplasm. Pretreatment with clathrin inhibitors significantly reduced the uptake of O. viverrini excretory/secretory products, particularly by H69 cells. Opisthorchis viverrini excretory/secretory products induced proliferation of liver cells (H69 and CCA lines) but not intestinal (Caco-2) cells, and proliferation was blocked using inhibitors of the classical endocytic pathways (clathrin and caveolae). Opisthorchis viverrini excretory/secretory products drove IL6 secretion by H69 cells but not Caco-2 cells, and cytokine secretion was significantly reduced by endocytosis inhibitors. This the first known study to address the endocytosis of helminth ES proteins by host epithelial cells and sheds light on the pathways by which this parasite causes one of the most devastating forms of cancer in south-eastern Asia

    Estimating fishing effort from highly resolved geospatial data : focusing on passive gears

Project N°: EAPA_134/2018”.Increasing competition for marine space requires the appropriate development of indicators to best represent the use of marine areas and the value (whether economic, social and/or cultural) derived from such use. Fishers (the largest group of users) are often under-represented in marine spatial planning processes. Highly-resolved vessel tracking data provide opportunities to map the activities of fishing vessels at a level of detail never before available. Most effort mapping methods have focused on active gears such as trawls or dredges in large scale fisheries. For these fisheries, the time spent fishing at sea (hours) is usually a representative indicator of fishing effort, enabling a straightforward mapping of the most important fishing grounds. However, for passive gears generally used in small-scale fisheries, we show that spatial indicators of effort (here, length of vessel track) greatly outperform time-at-sea as an indicator of fishing effort. We further demonstrate and validate a method to estimate gear soak time from vessel tracking data and show how maps of effort that account for soak time can be different from those solely based on time spent fishing at sea. The development of adequate methods to quantify the spatial distribution of passive gear effort is particularly relevant to fisheries management because globally about a fifth of all catches (by weight) are landed by passive gears. Appropriate, fine scale effort maps will provide better tools for spatial planning to support sustainable fishing.Publisher PDFPeer reviewe

    Structural characterisation of predicted helical regions in the Chironex fleckeri CfTX-1 toxin

    The Australian jellyfish Chironex fleckeri, belongs to a family of cubozoan jellyfish known for their potent venoms. CfTX-1 and -2 are two highly abundant toxins in the venom, but there is no structural data available for these proteins. Structural information on toxins is integral to the understanding of the mechanism of these toxins and the development of an effective treatment. Two regions of CfTX-1 have been predicted to have helical structures that are involved with the mechanism of action. Here we have synthesized peptides corresponding to these regions and analyzed their structures using NMR spectroscopy. The peptide corresponding to the predicted N-terminal amphiphilic helix appears unstructured in aqueous solution. This lack of structure concurs with structural disorder predicted for this region of the protein using the Protein DisOrder prediction System PrDOS. Conversely, a peptide corresponding to a predicted transmembrane region is very hydrophobic, insoluble in aqueous solution and predicted to be structured by PrDOS. In the presence of SDS-micelles both peptides have well-defined helical structures showing that a membrane mimicking environment stabilizes the structures of both peptides and supports the prediction of the transmembrane region in CfTX-1.     Deaths, late deaths, and role of infecting dose in Ebola virus disease in Sierra Leone: retrospective cohort study.

    OBJECTIVES:  To assess the frequency of fatal recrudescence from Ebola virus disease after discharge from treatment centres, and explore the influence of infecting dose on case fatality rates. DESIGN:  Retrospective cohort study. SETTING:  Western Area, Sierra Leone. PARTICIPANTS:  151 survivors treated for Ebola virus disease at the Kerry Town treatment centre and discharged. Survivors were followed up for a vital status check at four to nine months after discharge, and again at six to 13 months after discharge. Verbal autopsies were conducted for four survivors who had died since discharge (that is, late deaths). Survivors still living in Western Area were interviewed together with their household members. Exposure level to Ebola virus disease was ascertained as a proxy of infecting dose, including for those who died. MAIN OUTCOME MEASURES:  Risks and causes of late death; case fatality rates; odds ratios of death from Ebola virus disease by age, sex, exposure level, date, occupation, and household risk factors. RESULTS:  Follow-up information was obtained on all 151 survivors of Ebola virus disease, a mean of 10 months after discharge. Four deaths occurred after discharge, all within six weeks: two probably due to late complications, one to prior tuberculosis, and only one after apparent full recovery, giving a maximum estimate of recrudescence leading to death of 0.7%. In these households, 395 people were reported to have had Ebola virus disease, of whom 227 died. A further 53 people fulfilled the case definition for probable disease, of whom 11 died. Therefore, the case fatality rate was 57.5% (227/395) for reported Ebola virus disease, or 53.1% (238/448) including probable disease. Case fatality rates were higher in children aged under 2 years and adults older than 30 years, in larger households, and in infections occurring earlier in the epidemic in Sierra Leone. There was no consistent trend of case fatality rate with exposure level, although increasing exposure increased the risk of Ebola virus disease. CONCLUSIONS:  In this study of survivors in Western Area, Sierra Leone, late recrudescence of severe Ebola virus disease appears to be rare. There was no evidence for an effect of infecting dose (as measured by exposure level) on the severity of disease

    Evaluating strategies for managing anthropogenic mortality on marine mammals : an R implementation with the package RLA

    Funding: ADERA provided support for salaries (MA).Bycatch, the undesirable and non-intentional catch of non-target species in marine fisheries, is one of the main causes of mortality of marine mammals worldwide. When quantitative conservation objectives and management goals are clearly defined, computer-based procedures can be used to explore likely population dynamics under different management scenarios and estimate the levels of anthropogenic removals, including bycatch, that marine mammal populations may withstand. Two control rules for setting removal limits are the Potential Biological Removal (PBR) established under the US Marine Mammal Protection Act and the Removals Limit Algorithm (RLA) inspired from the Catch Limit Algorithm (CLA) developed under the Revised Management Procedure of the International Whaling Commission. The PBR and RLA control rules were tested in a Management Strategy Evaluation (MSE) framework. A key feature of PBR and RLA is to ensure conservation objectives are met in the face of the multiple uncertainties or biases that plague real-world data on marine mammals. We built a package named RLA in the R software to carry out MSE of control rules to set removal limits in marine mammal conservation. The package functionalities are illustrated by two case studies carried out under the auspices of the Oslo and Paris convention (OSPAR) (the Convention for the Protection of the Marine Environment of the North-East Atlantic) Marine Mammal Expert Group (OMMEG) in the context of the EU Marine Strategy Framework Directive. The first case study sought to tune the PBR control rule to the conservation objective of restoring, with a probability of 0.8, a cetacean population to 80% of carrying capacity after 100 years. The second case study sought to further develop a RLA to set removals limit on harbor porpoises in the North Sea with the same conservation objective as in the first case study. Estimation of the removals limit under the RLA control rule was carried out within the Bayesian paradigm. Outputs from the functions implemented in the package RLA allows the assessment of user-defined performance metrics, such as time to reach a given fraction of carrying capacity under a given level of removals compared to the time needed given no removals.Publisher PDFPeer reviewe
