    Effects of polylinker uATGs on the function of grass HKT1 transporters expressed in yeast cells

    HvHKT1 mediates K+ or Na+ uniport in yeast cells if the expression promoter is joined directly to the HvHKT1 cDNA, and Na+-K+ symport if a 59-nt polylinker is inserted. Our results show that three ATG triplets in the polylinker decreased the synthesis of the transporter and that the lower amount of transporter caused the functional change. With the rice HKT1 cDNA, the 59-nt polylinker changed the mode of Na+ uptake from K+-insensitive to K+-inhibitable. These two modes of Na+ uptake also occurred in rice plant

    Mineralization of Clapper Rail Eggshell from a Contaminated Salt Marsh System

    The effect of contamination on eggshell mineralization has been studied for clapper rails (Rallus longirostris) inhabiting a contaminated salt marsh in coastal Georgia. To assess the impact of contaminants, the thickness, microstructure (crystal orientation), mineral composition, and chemistry of shell material were analyzed from a contaminated site and a nearby reference site using optical microscopy, X-ray diffraction, inductively coupled plasma mass spectrometry, and gas chromatography with electron capture detector. Eggshells from the contaminated site were generally thinner than those from the reference site. Also, eggshells from the contaminated site were abnormally brittle and contained anomalous microstructural attributes. The combination of reduced shell thickness and anomalous microstructure resulted in weaker eggshells, which in turn could pose a significant threat to the reproductive success of the affected population. PCB concentrations in eggshells were at background levels in both sites. Eggshells from the contaminated site had higher concentrations of heavy metals, specifically mercury, than the reference site. The structural changes observed in eggshells may be related to the concentration of specific metals (e.g., Mg, Cu, Zn, Pb, and Hg) in shell, however, statistical analyses indicated that metals only explained a small portion of the observed variation in properties (i.e., thickness, crystal orientation). Further analysis is required to better constrain the factors leading to unusually weak eggshells in the contaminated site

    Health care experiences of Black cancer survivors: A qualitative study exploring drivers of low and high Consumer Assessment of Healthcare Providers and Systems global ratings of care

    Background: The purpose of this study was to understand the experiences of Black patients with cancer in health care by comparing drivers of high and low ratings. / Methods: Semistructured in-depth interviews were conducted with 18 Black patients with cancer recruited from cancer survivorship support groups and Facebook between May 2019 and March 2020. Interviews were coded across all transcripts by using a thematic analysis approach before comparing low- and high-rating groups. / Results: There were three major themes that influenced whether patients rated their care as low or high, which included the patient–provider relationship, health care staff interactions, and cancer care coordination. For example, the high-rating group described good communication with the health care team as physicians listening to their needs, being responsive to their concerns, and providing recommendations on how to address side effects. In contrast, the low-rating group described poor communication with their health care team as their needs being dismissed and being excluded from decision-making processes. Additionally, there were two distinct themes that influenced patients’ low ratings: insurance and financial toxicity issues and experiences of health care discrimination. / Conclusions: In an effort to promote equitable cancer care experiences for Black patients, it is important that health systems work to prioritize patient interactions with health care providers and staff, comprehensive care management for patients with cancer, and reductions in the financial burden of caring for cancer

    Clapper Rails (Rallus longirostris) can potentially serve as an indicator species of estuarinemarsh health because of their strong site fi delity and predictable diet consisting predominantly of benthic organisms. These feeding habits increase the likelihood of individuals accumulating signifi - cant amounts of contaminants associated with coastal sediments. Moreover, since Clapper Rails are threatened in most of their western range, additional study of the effects of potential toxins on these birds is essential to conservation programs for this species. Here we present techniques (DNA strand breakage, eggshell structure, and human-consumption risk) that can be used to quantify detrimental effects to Clapper Rails exposed to multiple contaminants in disturbed ecosystems as well as humans who may eat them. Adult birds collected near a site contaminated with polychlorinated biphenyls (PCBs) and metals in Brunswick, Georgia had a high degree of strand breakage, while those collected from a nearby reference area had no strand breakage. Although, results showed that eggshell integrity was compromised in eggs from the contaminated sites, these results were more diffuse, reemphasizing that multiple endpoints should be used in ecological assessments. This study also shows that techniques such as eggshell integrity on hatched eggs and DNA strand breakage in adults can be used as non-lethal mechanisms to monitor the population health of more threatened populations such as those in the western US. We also present results from human-based risk assessment for PCBs as a third toxicological endpoint, since these species are hunted and consumed by the public in the southeastern US. Using standard human-risk thresholds, we show a potential risk to hunters who consume Clapper Rails shot near the contaminated site from PCBs because of the additional lifetime cancer risk associated with that consumption

    Fenugreek proteins and their hydrolysates prevent hypercholesterolemia and enhance the HDL antioxidant properties in rats

    Purpose This paper aims to investigate the in vivo hypocholesterolemic property of fenugreek proteins (FP), Purafect-fenugreek protein hydrolysate (PFPH) and Esperase-fenugreek protein hydrolysate (EFPH) on high cholesterol (HC)-fed rats. Design/methodology/approach Rats were randomized into five groups: four were fed for four weeks a hypercholesterolemic diet and the tested products were given by gavage. The fifth group was taken as control (C) receiving the same diet without cholesterol. Findings Results showed that the elevated aspartate aminotransferase activity in HC group plasma was significantly corrected by FP and EFPH administration (-33 per cent; p = 0.0003). HC liver lipids and total cholesterol (TC) contents were not markedly affected by FP and EFPH. However, liver triglycerides (TG) contents trended to decrease in FP rats vs HC (p = 0.07), while, the TG decrease was significant in groups fed the proteins hydrolysates (p = 0.02). On the other hand, serum TC and TG decreased by 53 per cent (p = 0.0003) and 20 per cent (p = 0.04), respectively, in FP treated rats compared to HC group. This decrease was associated with a high fecal cholesterol excretion (2.5-fold higher in FP vs HC; p = 0.0001). Likewise, EFPH-treated rats exhibited lower TC compared to HC rats (p = 0.004). The very low density lipoprotins was the main affected fraction in these two groups, while there were no significant difference in apolipoproteins (Apo) B, A-I and A-IV contents between the different groups, except in FP group, where Apo A-I and A-IV decreased by 26 and 17 per cent, respectively, compared to C rats (p = 0.02). The high density lipoproteins (HDL) of rats treated with proteins hydrolysates showed a better antioxidant property compared to those of HC rats, which was accompanied with an increase in paraoxonase activity when compared to HC group. Originality/value Unlike PFPH which had almost no effect, FPs and EFPH could constitute a nutraceutical ingredient in cardiovascular disease management

    Dyskeratosis congenita: natural history of the disease through the study of a cohort of patients diagnosed in childhood

The main objective of this study is to describe the natural history of DC through a cohort of patients diagnosed in childhood and followed up for a long period of time. Material and methods: Multicenter, retrospective, longitudinal study conducted in patients followed up to 24 years since being diagnosed in childhood (between 1998 and 2020). Results: Fourteen patients were diagnosed with DC between the ages of 3 and 17 years (median, 8.5 years). They all had hematologic manifestations at diagnosis, and nine developed mucocutaneous manifestations during the first decade of life. Seven presented severe DC variants. All developed non-hematologic manifestations during follow-up. Mutations were identified in 12 patients. Thirteen progressed to bone marrow failure at a median age of 8 years [range, 3–18 years], and eight received a hematopoietic stem cell transplant. Median follow-up time was 9 years [range, 2–24 years]. Six patients died, the median age was 13 years [range, 6–24 years]. As of November 2022, eight patients were still alive, with a median age of 18 years [range, 6–32 years]. None of them have developed myeloblastic syndrome or cancer. Conclusions: DC was associated with high morbidity and mortality in our series. Hematologic manifestations appeared early and consistently. Non-hematologic manifestations developed progressively. No patient developed cancer possibly due to their young age. Due to the complexity of the disease multidisciplinary follow-up and adequate transition to adult care are essential

    The role of OCT-A in retinal disease management

    Optical coherence tomography angiography (OCT-A) is a non-invasive, non-dye-based imaging modality that has the potential to enhance our understanding of retinal diseases. While this rapidly advancing imaging modality offers great potential, there is a need for community-wide understanding of the range of technologies and methods for interpreting the images, as well as a need to enhance understanding of images from disease-free eyes for reference when screening for retinal diseases. Importantly, clinical trials have been designed without OCT-A-based endpoints; therefore, caution is required when making treatment decisions based on OCT-A imaging alone. With this in mind, a full understanding of the advantages and limitations of OCT-A will be vital for effective development of the technique within the field of ophthalmology. On behalf of the Vision Academy Steering Committee (sponsored by Bayer), this publication summarizes the views of the authors on the current use of OCT-A imaging and explores its potential for future applications in research and clinical practice

    The CARMA Study: Early Infant Antiretroviral Therapy - Timing Impacts on Total HIV-1 DNA Quantitation 12 Years Later

    Background. Strategies aimed at antiretroviral therapy (ART)-free remission will target individuals with a limited viral reservoir. We investigated factors associated with low reservoir measured as total human immunodeficiency virus type 1 (HIV-1) DNA in peripheral blood mononuclear cells (PBMCs) in perinatal infection (PaHIV).Methods. Children from 7 European centers in the Early Treated Perinatally HIV Infected Individuals: Improving Children's Actual Life (EPIICAL) consortium who commenced ART aged <2 years, and remained suppressed (viral load [VL] <50 copies/mL) for >5 years were included. Total HIV-1 DNA was measured by quantitative polymerase chain reaction per million PBMCs. Factors associated with total HIV-1 DNA were analyzed using generalized additive models. Age, VL at ART initiation, and baseline CD4% effects were tested including smoothing splines to test nonlinear association.Results. Forty PaHIV, 27 (67.5%) female 21 (52.5%) Black/Black African, had total HIV-1 DNA measured; median 12 (IQR, 7.3-15.4) years after ART initiation. Eleven had total HIV-1 DNA <10 copies/10(6) PBMCs. HIV-1 DNA levels were positively associated with age and VL at ART initiation, baseline CD4%, and Western blot antibody score. Age at ART initiation presented a linear association (coefficient = 0.10 +/- 0.001, P <= .001), the effect of VL (coefficient = 0.35 +/- 0.1, P <= .001) noticeable >6 logs. The effect of CD4% (coefficient = 0.03 +/- 0.01, P <= .049) was not maintained >40%.Conclusions. In this PaHIV cohort, reduced total HIV-1 DNA levels were associated with younger age and lower VL at ART initiation. The impact of early-infant treatment on reservoir size persists after a decade of suppressive therapy
