    Protein spot arrays on graphene oxide coatings for efficient single-cell capture

    Biomedical applications such as cell screening or cell–cell interaction studies require placement and adhesion of cells on surfaces with controlled numbers and location. In particular, single-cell arraying and positioning has come into focus as a basis of such applications. An ideal substrate would combine biocompatibility with favorable attributes such as pattern stability and easy processing. Here, we present a simple yet effective approach to single-cell arraying based on a graphene oxide (GO) surface carrying protein (fibronectin) microarrays to define cell adhesion points. These capture NIH-3T3 cells, resulting in cell arrays, which are benchmarked against analogous arrays on silanized glass samples. We reveal significant improvement in cell-capture performance by the GO coating with regards to overall cell adhesion and single-cell feature occupancy. This overall improvement of cell-arraying combined with retained transparency of substrate for microscopy and good biocompatibility makes this graphene-based approach attractive for single-cell experiments

    The role of combining national official statistics with global monitoring to close the data gaps in the environmental SDGs

    The Sustainable Development Goals (SDGs) have elevated the profile of the environmental dimension of development – and how we monitor this dimension. However, they have also challenged national statistical systems and the global statistical community to put in place both the methodologies and mechanisms for data collection and reporting on environmental indicators. According to a recent analysis, there is too little data to formally assess the status of 68% of the environment-related SDGs [1]. Many environment-related indicators were not part of the purview of national statistical systems and did not have a methodology or data collection system in place prior to the adoption of the SDG indicator framework [2]. Moderate improvements have been made, as evidenced by the reduced proportion of environment-related SDG indicators classified as Tier III between the original classification in 2016 and May 2019 – dropping from 50% to 28% [3]. As of March 2020, there are currently no Tier III indicators; however, as many of the SDG indicators have been recently reclassified the data availability and experience in compiling these indicators is severely limited. Socioeconomic indicators have far outpaced environmental indicators in this shift, with only 7% of non-environmental indicators classified as Tier III in May 2019 [1,4,5]. As the custodian agency for 26 of the environment-related SDG indicators, UN Environment is establishing methodologies and mechanisms to collect country-level data. However, many countries currently do not have national systems in place for monitoring these environmental indicators and thus there is a risk that much of the environmental dimension of development cannot be captured by using reporting mechanisms which only include traditionally collected national official statistics. For many of these indicators, UN Environment is exploring new data sources, such as data from citizen science. Citizen science has the potential to contribute to global and local level SDG monitoring. Realizing its full potential however, would require building key partnerships around citizen science data and creating an enabling environment. Global modelling is another approach to fill data gaps. These new types of data could not only improve global estimations but could be incorporated in national official statistics in order to improve nationally relevant data and analysis [6]. The Global Material Flow database, which estimates Domestic Material Consumption (covering SDG indicators 8.4.2 and 12.2.2), and the Global Surface Water Explorer application (covering SDG indicator 6.6.1) are a couple of examples of where UN Environment is complementing national data with global data products in the official SDG reporting process. In these cases the use of globally-derived data has been agreed by the Inter-Agency and Expert Group on SDG Indicators (IAEG-SDGs) [7]. Expanding globally-estimated or -modelled data to cover environment-related SDG indicators could build the foundation for a digital ecosystem for the planet, which would provide a basis for developing integrated analysis and insights. A Sustainability Gap Index could be one mechanism to bring together the environmental dimension of development into a single metric, which could inform the achievement of the SDGs, environmental assessments and national policy.     Coulomb plasmas in outer envelopes of neutron stars

    Outer envelopes of neutron stars consist mostly of fully ionized, strongly coupled Coulomb plasmas characterized by typical densities about 10^4-10^{11} g/cc and temperatures about 10^4-10^9 K. Many neutron stars possess magnetic fields about 10^{11}-10^{14} G. Here we briefly review recent theoretical advances which allow one to calculate thermodynamic functions and electron transport coefficients for such plasmas with an accuracy required for theoretical interpretation of observations.Comment: 4 pages, 2 figures, latex2e using cpp2e.cls (included).     Treating children traumatized by war and Tsunami: A comparison between exposure therapy and meditation-relaxation in North-East Sri Lanka

    <p>Abstract</p> <p>Background</p> <p>The North-Eastern part of Sri Lanka had already been affected by civil war when the 2004 Tsunami wave hit the region, leading to high rates of posttraumatic stress disorder (PTSD) in children. In the acute aftermath of the Tsunami we tested the efficacy of two pragmatic short-term interventions when applied by trained local counselors.</p> <p>Methods</p> <p>A randomized treatment comparison was implemented in a refugee camp in a severely affected community. 31 children who presented with a preliminary diagnosis of PTSD were randomly assigned either to six sessions Narrative Exposure Therapy for children (KIDNET) or six sessions of meditation-relaxation (MED-RELAX). Outcome measures included severity of PTSD symptoms, level of functioning and physical health.</p> <p>Results</p> <p>In both treatment conditions, PTSD symptoms and impairment in functioning were significantly reduced at one month post-test and remained stable over time. At 6 months follow-up, recovery rates were 81% for the children in the KIDNET group and 71% for those in the MED-RELAX group. There was no significant difference between the two therapy groups in any outcome measure.</p> <p>Conclusion</p> <p>As recovery rates in the treatment groups exceeded the expected rates of natural recovery, the study provides preliminary evidence for the effectiveness of NET as well as meditation-relaxation techniques when carried out by trained local counselors for the treatment of PTSD in children in the direct aftermath of mass disasters.</p> <p>Trial registration</p> <p>ClinicalTrials.gov Identifier:NCT00820391</p

    Genetic background modifies CNS-mediated sensorimotor decline in the AD-BXD mouse model of genetic diversity in Alzheimer\u27s disease.

    Many patients with Alzheimer\u27s dementia (AD) also exhibit noncognitive symptoms such as sensorimotor deficits, which can precede the hallmark cognitive deficits and significantly impact daily activities and an individual\u27s ability to live independently. However, the mechanisms underlying sensorimotor dysfunction in AD and their relationship with cognitive decline remains poorly understood, due in part to a lack of translationally relevant animal models. To address this, we recently developed a novel model of genetic diversity in Alzheimer\u27s disease, the AD-BXD genetic reference panel. In this study, we investigated sensorimotor deficits in the AD-BXDs and the relationship to cognitive decline in these mice. We found that age- and AD-related declines in coordination, balance and vestibular function vary significantly across the panel, indicating genetic background strongly influences the expressivity of the familial AD mutations used in the AD-BXD panel and their impact on motor function. Although young males and females perform comparably regardless of genotype on narrow beam and inclined screen tasks, there were significant sex differences in aging- and AD-related decline, with females exhibiting worse decline than males of the same age and transgene status. Finally, we found that AD motor decline is not correlated with cognitive decline, suggesting that sensorimotor deficits in AD may occur through distinct mechanisms. Overall, our results suggest that AD-related sensorimotor decline is strongly dependent on background genetics and is independent of dementia and cognitive deficits, suggesting that effective therapeutics for the entire spectrum of AD symptoms will likely require interventions targeting each distinct domain involved in the disease

Nun hat sie beschlossen, eine Liste mit fünf Untersuchungen auf dem Gebiet der ambulanten allgemeinen inneren Medizin zusammenzustellen, die ohne oder mit nur geringem Nutzen bei zahlreichen Patienten durchgeführt werden, gleichzeitig jedoch unerwünschte Nebenwirkungen haben können und zum Anstieg der Gesundheitskosten beitragen

    Translational approaches to understanding resilience to Alzheimer\u27s disease.

    Individuals who maintain cognitive function despite high levels of Alzheimer\u27s disease (AD)-associated pathology are said to be \u27resilient\u27 to AD. Identifying mechanisms underlying resilience represents an exciting therapeutic opportunity. Human studies have identified a number of molecular and genetic factors associated with resilience, but the complexity of these cohorts prohibits a complete understanding of which factors are causal or simply correlated with resilience. Genetically and phenotypically diverse mouse models of AD provide new and translationally relevant opportunities to identify and prioritize new resilience mechanisms for further cross-species investigation. This review will discuss insights into resilience gained from both human and animal studies and highlight future approaches that may help translate these insights into therapeutics designed to prevent or delay AD-related dementia

    Unsilencing of native leptin receptors (LepR) in hypothalamic SF1 neurons does not rescue obese phenotype in LepR-deficient mice

    Leptin receptor (LepR) signaling in neurons of the ventromedial nucleus of the hypothalamus (VMH), specifically those expressing steroidogenic factor-1 (SF1), have been proposed to play a key role in controlling energy balance. By crossing LepR-silenced (LepR) mice to those expressing SF1-Cre, we unsilenced native LepR specifically in the VMH and tested whether SF1 neurons in the VMH are critical mediators of leptin's effect on energy homeostasis. LepR x SF1-Cre (KO/Tg+) mice were metabolically phenotyped and compared to littermate controls that either expressed or were deficient in LepR. Leptin-induced pSTAT3 was present in the VMH of KO/Tg+ mice and absent in other hypothalamic nuclei. VMH leptin signaling did not ameliorate obesity resulting from LepR-deficiency in chow-fed mice. There was no change in food intake or energy expenditure when comparing complete LepR-null mice to KO/Tg+ mice, nor did KO/Tg+ show improved glucose tolerance. The presence of functional LepR in the VMH mildly enhanced sensitivity to the pancreatic hormone amylin. When maintained on high fat diet (HFD), there was no reduction in diet-induced obesity in KO/Tg+ mice, but KO/Tg+ mice had improved glucose tolerance after 7 weeks on HFD compared to LepR-null mice. We conclude that LepR signaling in the VMH alone is not sufficient to correct metabolic dysfunction observed in LepR-null mice
