    Diversity of Tephritidae and Agromyzidae (Diptera: Brachycera) in Flower Heads of Asteraceae in the Chaco

    The Chaco is an international biome, connecting four countries: Paraguay (230,000 km2), Bolivia (90,000 km2), Argentina (520,000 km2), and Brazil (Mato Grosso do Sul state (MS), with around 9,000 km2 and in the middle of South America. Brazilian Chaco is restricted to Porto Murtinho region, MS. The daisies (Asteraceae) with near 24,000 species worldwide is characterized by herbs and shrubs that coevolved with several taxa of endophagous insects: dipterans Agromyzidae, Ceciidomyidae and Tephritidae; Coleoptera (Apionidae), Hemiptera (Miridae), Lepidoptera (Blastobasidae, Gelechiidae, Pterophoridae, Pyralidae, and Tortricidae) and the parasitoids of this endophagous insects, which found in the daisies’s flower heads ideal conditions for food, breeding site and shelter. The Neotropical florivorous flies are the Agromyzinae (Agromyzidae), and Tephritinae (Tephritidae), which in their larval stage feed on Asteraceae inflorescences. To report the species of florivore flies, their host plants and parasitoids in flower heads of Asteraceae from the Brazilian Chaco, we sampled inflorescences of 25 species (± 500 flower heads/species) that were kept in containers to the emergence of the florivorous flies or their parasitoids sampled in the three phytophysiognomies. The adult insects after 48 hours of their emergence were fixed in 80% ethanol for later identification. A total 25 species of Asteraceae were evaluated in the Brazilian Chaco, being collected 17,000 flower heads. Nine tribes of two Asteraceae subfamilies were sampled, from which 15 species of florivorous flies were recovered. We found 5 genera with 9 of Tephritinae (Tephritidae), 6 species of Melanagromyza (Agromyzinae, Agromyzidae), and 104 parasitoids (Hymenoptera) of the florivorous flies

    Entende-se por preconceito de gênero, as atitudes sociais que discriminam as pessoas de acordo com o seu sexo. Em geral, as mulheres são mais afetadas através de idéias, palavras e atos, determinando diferentes comportamentos sociais quando comparadas aos homens. Os preconceitos de gênero variam de acordo com o momento histórico e a cultura de cada local, e a sua identificação auxilia na superação dos mesmos. O objetivo desta pesquisa foi identificar estereótipos e preconceitos de gênero em alguns aspectos da vida social (trabalho, comportamento, linguagem e religião), entre estudantes de enfermagem. Foram entrevistados, com roteiro semiestruturado, 76 alunos da graduação, regularmente matriculados na Escola de Enfermagem da UFBA, aleatoriamente escolhidos entre os diversos semestres. A análise dos dados revelou que as profissões podem ser exercidas por homens e mulheres, dependendo apenas da capacidade de quem as exerce. Entretanto, descriminam atividades masculinas e femininas em cada uma. Identificou-se a existência de estereótipos e preconceitos sexistas relacionados ao trabalho, à religião e a outros aspectos da vida social

    Oxyanion flux characterization using passive flux meters: Development and field testing of surfactant-modified granular activated carbon

    Abstract We report here on the extension of Passive Flux Meter (PFM) applications for measuring fluxes of oxyanions in groundwater, and present results for laboratory and field studies. Granular activated carbon, with and without impregnated silver (GAC and SI-GAC, respectively), was modified with a cationic surfactant, hexadecyltrimethylammonium (HDTMA), to enhance the anion exchange capacity (AEC). Langmuir isotherm sorption maxima for oxyanions measured in batch experiments were in the following order: perchlorate NN chromate N selenate, consistent with their selectivity. Linear sorption isotherms for several alcohols suggest that surfactant modification of GAC and SI-GAC reduced (∼ 30-45%) sorption of alcohols by GAC. Water and oxyanion fluxes (perchlorate and chromate) measured by deploying PFMs packed with surfactant-modified GAC (SM-GAC) or surfactant-modified, silver-impregnated GAC (SM-SI-GAC) in laboratory flow chambers were in close agreement with the imposed fluxes. The use of SM-SI-GAC as a PFM sorbent was evaluated at a field site with perchlorate contamination of a shallow unconfined aquifer. PFMs packed with SM-SI-GAC were deployed in three existing monitoring wells with a perchlorate concentration range of ∼ 2.5 to 190 mg/L. PFM-measured, depth-averaged, groundwater fluxes ranged from 1.8 to 7.6 cm/day, while depth-averaged perchlorate fluxes varied from 0.22 to 1.7 g/m 2 /day. Groundwater and perchlorate flux distributions measured in two PFM deployments closely matched each other. Depth-averaged Darcy fluxes measured with PFMs were in line with an estimate from a borehole dilution test, but much smaller than those based on hydraulic conductivity and head gradients; this is likely due to flow divergence caused by well-screen clogging. Flux-averaged perchlorate concentrations measured with PFM deployments matched concentrations in groundwater samples taken from one well, but not in two Journal of Contaminant Hydrology 92 (2007) other wells, pointing to the need for additional field testing. Use of the surfactant-modified GACs for measuring fluxes of other anions of environmental interest is discussed

    The State of the Art in Language Workbenches. Conclusions from the Language Workbench Challenge

    Language workbenches are tools that provide high-level mechanisms for the implementation of (domain-specific) languages. Language workbenches are an active area of research that also receives many contributions from industry. To compare and discuss existing language workbenches, the annual Language Workbench Challenge was launched in 2011. Each year, participants are challenged to realize a given domain-specific language with their workbenches as a basis for discussion and comparison. In this paper, we describe the state of the art of language workbenches as observed in the previous editions of the Language Workbench Challenge. In particular, we capture the design space of language workbenches in a feature model and show where in this design space the participants of the 2013 Language Workbench Challenge reside. We compare these workbenches based on a DSL for questionnaires that was realized in all workbenches

    IκBβ acts to inhibit and activate gene expression during the inflammatory response

    The activation of pro-inflammatory gene programs by nuclear factor-κB (NF-κB) is primarily regulated through cytoplasmic sequestration of NF-κB by the inhibitor of κB (IκB) family of proteins1. IκBβ, a major isoform of IκB, can sequester NF-κB in the cytoplasm2, although its biological role remains unclear. Although cells lacking IκBβ have been reported3, 4, in vivo studies have been limited and suggested redundancy between IκBα and IκBβ5. Like IκBα, IκBβ is also inducibly degraded; however, upon stimulation by lipopolysaccharide (LPS), it is degraded slowly and re-synthesized as a hypophosphorylated form that can be detected in the nucleus6, 7, 8, 9, 10, 11. The crystal structure of IκBβ bound to p65 suggested this complex might bind DNA12. In vitro, hypophosphorylated IκBβ can bind DNA with p65 and c-Rel, and the DNA-bound NF-κB:IκBβ complexes are resistant to IκBα, suggesting hypophosphorylated, nuclear IκBβ may prolong the expression of certain genes9, 10, 11. Here we report that in vivo IκBβ serves both to inhibit and facilitate the inflammatory response. IκBβ degradation releases NF-κB dimers which upregulate pro-inflammatory target genes such as tumour necrosis factor-α (TNF-α). Surprisingly, absence of IκBβ results in a dramatic reduction of TNF-α in response to LPS even though activation of NF-κB is normal. The inhibition of TNF-α messenger RNA (mRNA) expression correlates with the absence of nuclear, hypophosphorylated-IκBβ bound to p65:c-Rel heterodimers at a specific κB site on the TNF-α promoter. Therefore IκBβ acts through p65:c-Rel dimers to maintain prolonged expression of TNF-α. As a result, IκBβ^(−/−) mice are resistant to LPS-induced septic shock and collagen-induced arthritis. Blocking IκBβ might be a promising new strategy for selectively inhibiting the chronic phase of TNF-α production during the inflammatory response

    Architecture of the RNA polymerase II–TFIIF complex revealed by cross-linking and mass spectrometry

    Higher-order multi-protein complexes such as RNA polymerase II (Pol II) complexes with transcription initiation factors are often not amenable to X-ray structure determination. Here, we show that protein cross-linking coupled to mass spectrometry (MS) has now sufficiently advanced as a tool to extend the Pol II structure to a 15-subunit, 670 kDa complex of Pol II with the initiation factor TFIIF at peptide resolution. The N-terminal regions of TFIIF subunits Tfg1 and Tfg2 form a dimerization domain that binds the Pol II lobe on the Rpb2 side of the active centre cleft near downstream DNA. The C-terminal winged helix (WH) domains of Tfg1 and Tfg2 are mobile, but the Tfg2 WH domain can reside at the Pol II protrusion near the predicted path of upstream DNA in the initiation complex. The linkers between the dimerization domain and the WH domains in Tfg1 and Tfg2 are located to the jaws and protrusion, respectively. The results suggest how TFIIF suppresses non-specific DNA binding and how it helps to recruit promoter DNA and to set the transcription start site. This work establishes cross-linking/MS as an integrated structure analysis tool for large multi-protein complexes

    Comparative proximity biotinylation implicates the small GTPase RAB18 in sterol mobilization and biosynthesis

    Loss of functional RAB18 causes the autosomal recessive condition Warburg Micro syndrome. To better understand this disease, we used proximity biotinylation to generate an inventory of potential RAB18 effectors. A restricted set of 28 RAB18-interactions were dependent on the binary RAB3GAP1-RAB3GAP2 RAB18-guanine nucleotide exchange factor (GEF) complex. 12 of these 28 interactions are supported by prior reports and we have directly validated novel interactions with SEC22A, TMCO4 and INPP5B. Consistent with a role for RAB18 in regulating membrane contact sites (MCSs), interactors included groups of microtubule/membrane-remodelling proteins, membrane-tethering and docking proteins, and lipid-modifying/transporting proteins. Two of the putative interactors, EBP and OSBPL2/ORP2, have sterol substrates. EBP is a Δ8-Δ7 sterol isomerase and ORP2 is a lipid transport protein. This prompted us to investigate a role for RAB18 in cholesterol biosynthesis. We find that the cholesterol precursor and EBP-product lathosterol accumulates in both RAB18-null HeLa cells and RAB3GAP1-null fibroblasts derived from an affected individual. Further, de novo cholesterol biosynthesis is impaired in cells in which RAB18 is absent or dysregulated, or in which ORP2 expression is disrupted. Our data demonstrate that GEF-dependent Rab-interactions are highly amenable to interrogation by proximity biotinylation and may suggest that Micro syndrome is a cholesterol biosynthesis disorder
