    Acute Treadmill Exercise Discriminately Improves the Skeletal Muscle Insulin-Stimulated Growth Signaling Responses in Mice Lacking REDD1

    A loss of the regulated in development and DNA damage 1 (REDD1) hyperactivates mechanistic Target of Rapamycin Complex 1 (mTORC1) reducing insulin‐stimulated insulin signaling, which could provide insight into mechanisms of insulin resistance. Although aerobic exercise acutely inhibits mTORC1 signaling, improvements in insulin‐stimulated signaling are exhibited. The goal of this study was to determine if a single bout of treadmill exercise was sufficient to improve insulin signaling in mice lacking REDD1. REDD1 wildtype (WT) and REDD1 knockout (KO) mice were acutely exercised on a treadmill (30 min, 20 m/min, 5% grade). A within animal noninsulin‐to‐insulin‐stimulated percent change in skeletal muscle insulin‐stimulated kinases (IRS‐1, ERK1/2, Akt), growth signaling activation (4E‐BP1, S6K1), and markers of growth repression (REDD1, AMPK, FOXO1/3A) was examined, following no exercise control or an acute bout of exercise. Unlike REDD1 KO mice, REDD1 WT mice exhibited an increase (P \u3c 0.05) in REDD1 following treadmill exercise. However, both REDD1 WT and KO mice exhibited an increase (P \u3c 0.05) AMPK phosphorylation, and a subsequent reduction (P \u3c 0.05) in mTORC1 signaling after the exercise bout versus nonexercising WT or KO mice. Exercise increased (P \u3c 0.05) the noninsulin‐to‐insulin‐stimulated percent change phosphorylation of mTORC1, ERK1/2, IRS‐1, and Akt on S473 in REDD1 KO mice when compared to nonexercised KO mice. However, there was no change in the noninsulin‐to‐insulin‐stimulated percent change activation of Akt on T308 and FOXO1/3A in the KO when compared to WT or KO mouse muscle after exercise. Our data show that a bout of treadmill exercise discriminately improves insulin‐stimulated signaling in the absence of REDD1

    Perkinsus Sp Infection Risk For Manila Clams, Venerupis Philippinarum (A. Adams And Reeve, 1850) On The Pacific Coast Of North And Central America

    Manila clams (Venerupis philippinarum, A. Adams and Reeve 1850) are an important aquaculture species on the west coast of North America and are also cultured in Europe, Asia, and other locations. Clams cultured on the west coast of North America are free of Perkinsus sp. infections, while clams from certain Asian and European sources are infected. Infection in Korean Manila clams is reportedly associated with high morbidity and mortality. We evaluated the health status of readily accessible Manila clam juveniles from Korea that were proposed for importation into Mexican waters where they would increase in size, and then be shipped into the United States, either to market destinations or to receiving waters. The examination of the clams was performed as a preliminary assessment for a producer considering the importation of Korean Manila clams. We report finding a high prevalence of a Perkinsus sp. causing significant tissue damage in juvenile Korean Manila clams. Parasite taxonomic verification was made using a genus-Perkinsus SSUrRNA gene-specific DNA probe for in situ hybridization. The use of this probe is validated and reported for the first time. As a result of this finding, no importation of this clam stock took place. It is urgently important to make widely known the risk of the spread of this disease into the clam stocks of the west coast of North and Central America to prevent such an introduction. In addition, we report new information regarding the prevalence and intensity of this disease in juvenile clams available for export, as well as pathologic features of the disease

    Sino malaya?: Melodrama and the Integration of Contemporary Social Issues in Philippine Independent Films

    Various film critics have written about the dominant themes found in independent films, such as gender and sexuality, culture, and sociopolitical issues. Independent filmmakers have distinctive ways of portraying these issues. Guided by Pathak (2017)’s framework on melodrama, the researchers chose to use melodramatic conventions and techniques to analyze further how these social issues are portrayed in independent films. Findings found that the independent films used melodramatic conventions in portraying social issues. Analyses showed that (1) the films made use of issues such as family relationships and understanding reality, (2) various cinematic elements were applied in the context of melodrama to deliver the film’s message, and (3) setting and physical characteristics were utilized in independent films to display social issues better. This study may be relevant to future research on independent film analysis, the development of national cinema, and the progress of local independent films as a whole

    Assessment of Bioaerosols at a Concentrated Dairy Operation.

    Increased bioaerosol loadings in downwind plumes from concentrated animal feeding operations (CAFOs) may increase the risk for allergy and infection in humans. In this study we monitored airborne concentrations of culturable bacteria and fungi at upwind (background) and downwind sites at a 10,000 milking cow dairy over the course of a year. The average bacterial concentrations at the upwind site were 8,400 colony forming units (CFU)/cubic meter and increased to 990,000 CFU/cubic meter at the downwind edge of the cattle pens, decreasing to 63,000 CFU/cubic meter 200 m farther downwind. At the same sites, the average fungal concentrations were 515, 945, and 1010 CFU/cubic meter, respectively. Significant correlations between the ambient weather data and airborne fungal concentrations were identified, but not with bacteria. Sequence analysis of PCR-amplified DNA from bacterial clones and fungal isolates revealed genus and species level differences between upwind and downwind sites. Although we could not cultivate gram-negative bacteria, bacterial clones at downwind sites identified as being gram-negative matched with the following genera: Acinetobacter, Bradyrhizobium, Escherichia, Idiomarina, Methylobacterium, Ralstonia, and Novosphingobium. Fungal isolates from downwind matched with the following genera: Acremonium, Alternaria, Ascomycte, Aspergillus, Basidiomycete, Cladosporium, Davidiella, Doratomyces, Emericella, Lewia, Onygenales, Penicillium, Rhizopus, and Ulocladium. None of the bacterial and fungal sequence matches were affiliated with genera and species known to be pathogenic to humans. Overall, the concentrated dairy operation does not appear to increase the risk of exposure to bioaerosols, especially when an individual increases their downwind distance from the facility
