    MORPHOLOGICAL ANALYSIS OF ANCIENT GRAPE SEEDS FROM A SINK IN THE MIDDLE-AGE TOWN OF PALERMO

    The archaeological excavations in Piazza della Vittoria, in the Roman-Middle Age town of Palermo (Sicily) put in light a sink 3.20 m deep and 1 square m. large, partially filled by thin organic sediments. Grape seeds (grape-stones), fish scales and few vertebrate bones have been found in specific strata sealed under a stratum chronologically attributed to Islamic Middle-Age period (a post-quem limit). The finding of well preserved grape seeds is peculiar and their study opens the opportunity to improve the actual knowledge about evolution, cultivation, use and trade of Vitis L. in the Mediterranean area. This preliminary work focuses on morphologic and morphometric analysis of the ancient grape seeds with two aims: i) systematically describe the remains collection and, ii) define seeds typology and a consequent morphotaxonomic attribution. Over 200 seeds have been carefully dry cleaned (soft brush), photographed and analyzed for total breadth (B), total length (L) and length of stalk (LS) parameters, the most efficient for typological attribution (1, 2); Stummer index has been also calculated (1). Apical notch length (AN) has been for the first time evaluated. Measurements on digital images have been performed using ImageJ 1.31 platform; morphological parameters have been assembled in a dedicated database. Descriptive analysis and linear correlations have been performed using SYSTAT 10. Analysis of variance (ANOVA) and Tukey’s HSD (5% level of significance, α = 0.05) have been applied. All the parameters approximate a normal distribution. Major variation has been observed in LS (c.v. = 35.6%) and AN (c.v. = 35.6%), while B and L showed a c.v. of 9.5% and 12.6% respectively. All the analyzed parameters behave as independent variables with the exception of a significant correlation between Stummer index and L (R2 = 0.45; y = 8.17-0.047x with y = L and x = Stummer index). This correlation reveals that Stummer index depends more from the L and not from the B parameter. On the base of LS measures three subgroups have been arbitrarily created in relationship with the LS: LS1 0.90 mm (35 seeds). Analyzing together the LS groups toward AN, we have found a proportional and significant correlation (p = 0.05) between the extremes LS1 and LS3. In the entire collection, Stummer index varies from 55.76 to 100.86; in the LS groups, the range is 68.38-97.87 in LS1, 61.02-100.86 in LS2 and 55.76-81.70 in LS3. A small group (17) of seeds has been excluded for the impossibility to measure the stalk. The analyzed ancient grape seeds show a wide range of variability for all the considered parameters, revealing a polymorphic collection. In general, the seeds have a rounded heart-like shape, with a noticeable pointy stalk and a very invaginated apical notch. This typical shape is more marked in LS3 group. On the base of LS measures, LS1 is ascribable to wild grapevines, while LS2 and LS3 seem to be ascribable neither to wild nor to cultivated autochthonous Vitis. Furthermore, these seeds differ from those already described in other archaeological horizons in Italy (3) and in France (2). The Stummer index varies highly, exceeding the known range of wild Vitis vinifera (4), although values close to 100 have been already found in wild grapevines in Spain (5) and values above 80 have been also described in Extra-European Vitis species (6).     Functional characterization of the sea urchin sns chromatin insulator in erythroid cells

    Chromatin insulators are regulatory elements that determine domains of genetic functions. We have previously described the characterization of a 265 bp insulator element, termed sns, localized at the 3' end of the early historic H2A gene of the sea urchin Paracentrotus lividus. This sequence contains three cis-acting elements (Box A, Box B, and Box C+T) all needed for the enhancer-blocking activity in both sea urchin and human cells. The goal of this Study was to further characterize the sea urchin sns insulator in the erythroid environment. We employed colony assays in human (K562) and mouse (MEL) erythroid cell lines. We tested the capability of sns to interfere with the communication between the 5HS2 enhancer of the human beta-globin LCR and the gamma-globin promoter. We found that the sns sequence displays directional enhancer-blocking activity. By the use of antibodies against known DNA binding proteins, in electrophoretic mobility shift assays, we demonstrated the binding of the erythroid-specific GATA-1 and the ubiquitous Oct-1 and Sp1 transcription factors. These factors bind to Box A, Box B, and Box C+T, respectively, in both K562 and MEL nuclear extracts. These results may have significant implications for the conservation of insulator function ill evolutionary distant organisms and may prove to be of practical benefit in gene transfer applications for erythroid disorders such as hemoglobinopathies and thalassemias

    Quaternary marine and continental unconformity-bounded stratigraphic units of the NW Sicily coastal belt

    In the coastal sector of NW Sicily, the regional correlation of relevant unconformities recognised within the Quaternary sedimentary successions allowed the mapping of seven unconformitybounded stratigraphic units (UBSUs). The regional unconformities are marine or subaerial erosional surfaces, as well as non-depositional surfaces, locally marked by paleosoils. The erosional surfaces were produced from marine abrasion, surface water overland/ concentrated flow, river erosion, karst solution, mass movement, or wind erosion. The main lithofacies of the Quaternary UBSUs consist of: (a) marine and coastal bioclastic calcarenites, (b) aeolian sandstones, (c) river deposits, (d) colluvial deposits, (e) talus slope deposits, (f) landslide deposits, and (g) chemical carbonates (travertines and speleothems). Quaternary environmental changes, due to tectonics, climate, and sea-level oscillations, are the causes that favoured the development of erosion/deposition processes responsible for the genesis of unconformities and deposits. As a result, through the UBSU map of the NW Sicilian coastal belt, it is possible to: (i) recognise stratigraphic units controlled by tectonic, climatic, and environmental processes (and their interplay) and (ii) detect Quaternary sedimentary evolution

    Stories of courage in a group of adults with Substance Use Disorder

    The treatment for drug addiction is considered a difficult path for the most of patients. As matter of fact, individuals with Substance Use Disorder (SUD) experience numerous challenges before, during and after the treatment (e.g. tackling an unpredictable, uncertain and paradoxically negative future, tackling the anxiety and anticipatory fear of physical pain associated with abstinence; decide to go beyond self-justification and self-deception). Courage could be considered a positive and functional resource to help people with SUD to face challenges and difficulties related to treatment. In connection therewith, the aim of this study was to examine, using an embedded mix method analysis, the personal stories of courage of 80 individuals with SUD in order to identify the themes and types of courage used in their life. The analysis carried out showed that individuals with SUD reported more frequently stories of courage related to their SUD condition respect to other life situation. Moreover, the quantitative analysis showed that participants used more frequently psychological courage respect to moral and physical courage when these stories were referred to their SUD condition then other life situations

    Synthesis, antiproliferative activity, and mechanism of action of a series of 2-{[2E]-3-phenylprop-2-enoylamino}benzamides

    Several new 2-{[2E]-3-phenylprop-2-enoylamino}benzamides 12a-s and 17t-v were synthesized by stirring in pyridine the (E)-3-(2-R1-3-R2-4-R3-phenyl)acrylic acid chlorides 11c-k and 11t-v with the appropriate anthranilamide derivatives 10a-c or the 5-iodo anthranilic acid 13. Some of synthesized compounds were evaluated for their in vitro antiproliferative activity against the full NCI tumor cell line panel derived from nine clinically isolated cancer types (leukemia, non-small cell lung, colon, CNS, melanoma, ovarian, renal, prostate and breast). COMPARE analysis, effects on tubulin polymerization in cells and with purified tubulin, and effects on cell cycle distribution for 17t, the most active of the series, indicate that these new antiproliferative compounds act as antitubulin agent

    Aerosol-Assisted CVD-Grown PdO Nanoparticle-Decorated Tungsten Oxide Nanoneedles Extremely Sensitive and Selective to Hydrogen

    We report for the first time the successful synthesis of palladium (Pd) nanoparticle (NP)-decorated tungsten trioxide (WO3) nanoneedles (NNs) via a two-step aerosol-assisted chemical vapor deposition approach. Morphological, structural, and elemental composition analysis revealed that a Pd(acac)2 precursor was very suitable to decorate WO3 NNs with uniform and well-dispersed PdO NPs. Gas-sensing results revealed that decoration with PdO NPs led to an ultrasensitive and selective hydrogen (H2) gas sensor (sensor response peaks at 1670 at 500 ppm of H2) with low operating temperature (150 °C). The response of decorated NNs is 755 times higher than that of bare WO3 NNs. Additionally, at a temperature near that of the ambient temperature (50 °C), the response of this sensor toward the same concentration of H2 was 23, which is higher than that of some promising sensors reported in the literature. Finally, humidity measurements showed that PdO/WO3 sensors displayed low-cross-sensitivity toward water vapor, compared to bare WO3 sensors.     This paper describes the design and fabrication of a Ka Band PLL DRO having a fundamental oscillation frequency of 19.250 GHz, used as local oscillator in the low-noise block of a down converter (LNB) for an EHF band receiver. Apposite circuital models have been created to describe the behaviour of the dielectric resonator and of the active component used in the oscillator core. The DRO characterization and measurements have shown very good agreement with simulation results. A good phase noise performance is obtained by using a very highQdielectric resonator
