    Purification and structural characterization of the Na<sup>+</sup>-translocating ferredoxin: NAD<sup>+</sup> reductase (Rnf) complex of Clostridium tetanomorphum

    Various microbial metabolisms use H+/Na+-translocating ferredoxin:NAD+ reductase (Rnf) either to exergonically oxidize reduced ferredoxin by NAD+ for generating a transmembrane electrochemical potential or reversely to exploit the latter for producing reduced ferredoxin. For cryo-EM structural analysis, we elaborated a quick four-step purification protocol for the Rnf complex from Clostridium tetanomorphum and integrated the homogeneous and active enzyme into a nanodisc. The obtained 4.27 Ã… density map largely allows chain tracing and redox cofactor identification complemented by biochemical data from entire Rnf and single subunits RnfB, RnfC and RnfG. On this basis, we postulated an electron transfer route between ferredoxin and NAD via eight [4Fe-4S] clusters, one Fe ion and four flavins crossing the cell membrane twice related to the pathway of NADH:ubiquinone reductase. Redox-coupled Na+ translocation is provided by orchestrating Na+ uptake/release, electrostatic effects of the assumed membrane-integrated FMN semiquinone anion and accompanied polypeptide rearrangements mediated by different redox steps

    Superconducting properties of Nb thin films deposited on porous silicon templates

    Porous silicon, obtained by electrochemical etching, has been used as a substrate for the growth of nanoperforated Nb thin films. The films, deposited by UHV magnetron sputtering on the porous Si substrates, inherited their structure made of holes of 5 or 10 nm diameter and of 10 to 40 nm spacing, which provide an artificial pinning structure. The superconducting properties were investigated by transport measurements performed in the presence of magnetic field for different film thickness and substrates with different interpore spacing. Perpendicular upper critical fields measurements present peculiar features such as a change in the H_c2(T) curvature and oscillations in the field dependence of the superconducting resistive transition width at H=1 Tesla. This field value is much higher than typical matching fields in perforated superconductors, as a consequence of the small interpore distance.Comment: accepted for publication on Journal of Applied Physic

    Unadulterated spectral function of low energy quasiparticles: Bi-2212, nodal direction

    Fitting the momentum distribution photoemission spectra to the Voigt profile appears to be a robust procedure to purify the interaction effects from the experimental resolution. In application to Bi-2212 high-Tc cuprates, the procedure reveals the true scattering rate at low binding energies and temperatures, and, consequently, the true value of the elastic scattering. Reaching the minimal value ~ 16 meV, the elastic scattering does not reveal a systematic dependence on doping level, but is rather sensitive to impurity concentration, and can be explained by the forward scattering on out-of-plane impurities. The inelastic scattering is found to form well-defined quasiparticles with the quadratic and cubic energy dependence of the scattering rate above and below Tc, respectively. The observed energy-temperature asymmetry of the scattering rate is also discussed.Comment: 4 revtex pages, 4 figure

    Inter- and Intragranular Effects in Superconducting Compacted Platinum Powders

    Compacted platinum powders exhibit a sharp onset of diamagnetic screening at T≃1.9T \simeq 1.9 mK in zero magnetic field in all samples investigated. This sharp onset is interpreted in terms of the intragranular transition into the superconducting state. At lower temperatures, the magnetic ac susceptibility strongly depends on the ac field amplitude and reflects the small intergranular critical current density jcj_{c}. This critical current density shows a strong dependence on the packing fraction f of the granular samples. Surprisingly, jcj_{c} increases significantly with decreasing f (jc(B=0,T=0)≃0.07j_{c}(B=0, T=0) \simeq 0.07 A/cm2^{2} for f = 0.67 and jc(B=0,T=0)≃0.8j_{c}(B=0, T=0) \simeq 0.8 A/cm2^{2} for f = 0.50). The temperature dependence of jcj_{c} shows strong positive curvature over a wide temperature range for both samples. The phase diagrams of inter- and intragranular superconductivity for different samples indicate that the granular structure might play the key role for an understanding of the origin of superconductivity in the platinum compacts.Comment: 11 pages including 9 figures.     Relationships between Larval and Juvenile Abundance of Winter-Spawned Fishes in North Carolina, USA

    We analyzed the relationships between the larval and juvenile abundances of selected estuarine-dependent fishes that spawn during the winter in continental shelf waters of the U.S. Atlantic coast. Six species were included in the analysis based on their ecological and economic importance and relative abundance in available surveys: spot Leiostomus xanthurus, pinfish Lagodon rhomboides, southern flounder Paralichthys lethostigma, summer flounder Paralichthys dentatus, Atlantic croaker Micropogonias undulatus, and Atlantic menhaden Brevoortia tyrannus. Cross-correlation analysis was used to examine the relationships between the larval and juvenile abundances within species. Tests of synchrony across species were used to find similarities in recruitment dynamics for species with similar winter shelf-spawning life-history strategies. Positive correlations were found between the larval and juvenile abundances for three of the six selected species (spot, pinfish, and southern flounder). These three species have similar geographic ranges that primarily lie south of Cape Hatteras. There were no significant correlations between the larval and juvenile abundances for the other three species (summer flounder, Atlantic croaker, and Atlantic menhaden); we suggest several factors that could account for the lack of a relationship. Synchrony was found among the three southern species within both the larval and juvenile abundance time series. These results provide support for using larval ingress measures as indices of abundance for these and other species with similar geographic ranges and winter shelf-spawning life-history strategies

    Origin of complex crystal structures of elements at pressure

    We present a unifying theory for the observed complex structures of the sp-bonded elements under pressure based on nearly free electron picture (NFE). In the intermediate pressure regime the dominant contribution to crystal structure arises from Fermi-surface Brillouin zone (FSBZ) interactions - structures which allow this are favoured. This simple theory explains the observed crystal structures, transport properties, the evolution of internal and unit cell parameters with pressure.
