    Determinationof oxidation induction time and temperature bY DSC: Results of round robin tests

    Results obtained from three round robin tests, performed in 1998, 2000 and 2004, are summarized and presented. Oxidation Induction Time (OITtime) and Oxidation Induction Temperature (OITtemp) on six different grades of polyethylene were measured by DSC. The measured data were collected by Empa and evaluated using a robust statistical method concerning repeatability and reproducibility standard deviation. The results of the tests indicate that the determination of OITtime is associated with a high degree of uncertainty, especially for low OITtime values. Thus, OITtemp measurements could be a valuable alternative for testing less stabilized or non-stabilized polyolefin in particular.     Zusammenfassung: Der Nutzungsdruck auf Grundwasserressourcen in intensiv genutzten Flussebenen wird zunehmend größer. Ziel dieses Beitrages ist, anhand eines repräsentativen, instationären Datensatzes eines urbanen Grundwassersystems (Unteres Birstal CH) aufzuzeigen, wie mit Ansätzen des adaptiven Grundwassermanagements die Voraussetzungen für die nachhaltige Nutzung von Grundwasserressourcen geschaffen werden können. Mithilfe eines instationären Grundwassermodells können spezifische Fragen der Fluss-Grundwasser-Interaktion und dem Betrieb künstlicher Grundwasseranreicherung beantwortet werden. Die Instationarität der Fluss-Grundwasser-Interaktion und eine damit zusammenhängende Änderung von In - und Exfiltrationsverhältnissen konnte für verschiedene Flussabschnitte ermittelt werden. Die Datenauswertung eines Jahrhunderthochwassers trug wesentlich zum Verständnis dieser Prozesse bei. Durch ein Experiment mit der längerfristigen Außerbetriebnahme einer künstlichen Grundwasseranreicherung im Untersuchungsgebiet konnten die Auswirkungen von geplanten Nutzungsänderungen abgeschätzt werden. Die Untersuchungen tragen zum Prozessverständnis des Grundwassersystems bei und liefern die Grundlage für eine Diskussion über lang-, mittel- und kurzfristige Ziele hinsichtlich der regionalen Bewirtschaftung urbaner Wasserressource

    Moisture and Seasonality Shifts Recorded in Holocene and Pleistocene Speleothems From Southeastern Arabia

    The source and seasonality of rainfall in southern Arabia during the early- to mid-Holocene and preceding humid periods are controversial because fossil lacustrine sediments provide solely indirect information on the amount of rainfall. Hydrogen and oxygen isotope measurements on fluid inclusion water trapped in Holocene and Pleistocene stalagmites from Hoti Cave in Northern Oman are direct indicators of the isotopic composition of paleoprecipitation. Isotope values of fluid inclusions formed during peak interglacial periods plot along monsoonal water lines and are indicative of a southern monsoonal moisture source. The last monsoon-dominated period lasting from ∼10,100 to 6,300 years before present was terminated within a few decades in southeastern Arabia. The subsequent reduction in rainfall amount and change from predominantly summer to predominantly winter rainfall had a profound impact on human communities living in this area and triggered migration from inland to coastal areas where resources were more abundant

    From awareness raising to sustainable behaviour change in school: the WASH in school road map

    Development organisations working in the WASH sector tend to focus on their preferred methodologies for Hygiene and Sanitation (H&S) promotion, while the sustainability of an intervention depends much more on how the methodology is implemented and whether an enabling environment is in place. In 2014, Caritas Switzerland in Kenya started a learning and critical review process on how to improve its WASH in school interventions to finally make the step from raising students’ awareness on good H&S practices to sustainable behaviour change. As a result of this process, a number of lessons on how to foster the enabling environment were learned, based on which the WASH in school road map was developed. This article presents, for each phase of the WASH in school road map, the main lessons learned and how they have been implemented in Kenya

    Variable response in alpine tree-ring stable isotopes following volcanic eruptions in the tropics and iceland

    The importance of the stable isotopes in tree rings for the study of the climate variations caused by volcanic eruptions is still unclear. We studied δ18O, δD, δ13C stable isotopes of larch and cembran pine cellulose around four major eruptions with annual resolution, along with a superposed epoch analysis of 34 eruptions with 5-year resolution. Initial analysis of the tropical Tambora (1815 CE) and Samalas (1257 CE) eruptions showed a post-eruption decrease in δ18O values attributed to post-volcanic cooling and increased summer precipitation in Southern Europe, as documented by observations and climate simulations. The post-volcanic cooling was captured by the δD of speleothem fluid inclusion. The δ18O decrease was also observed in the analysis of 34 major tropical eruptions over the last 2000 years. In contrast, the eruptions of c. 750, 756, and 764 CE attributed to Icelandic volcanoes left no significant responses in the cellulose isotopes. Further analysis of all major Icelandic eruptions in the last 2000 years showed no consistent isotopic fingerprints, with the exception of lower post-volcanic δ13C values in larch. In summary, the δ18O values of cellulose can provide relevant information on climatic and hydroclimatic variations following major tropical volcanic eruptions, even when using the 5-year resolution wood samples of the Alpine Tree-Ring Isotope Record database

    Phase-coherence threshold and vortex-glass state in diluted Josephson-junction arrays in a magnetic field

    We study numerically the interplay of phase coherence and vortex-glass state in two-dimensional Josephson-junction arrays with average rational values of flux quantum per plaquette ff and random dilution of junctions. For f=1/2f=1/2, we find evidence of a phase coherence threshold value xsx_s, below the percolation concentration of diluted junctions xpx_p, where the superconducting transition vanishes. For xs<x<xpx_s < x < x_p the array behaves as a zero-temperature vortex glass with nonzero linear resistance at finite temperatures. The zero-temperature critical currents are insensitive to variations in ff in the vortex glass region while they are strongly ff dependent in the phase coherent region.Comment: 6 pages, 4 figures, to appear in Phys.     Infarct-remodelled hearts with limited oxidative capacity boost fatty acid oxidation after conditioning against ischaemia/reperfusion injury

    Aims Infarct-remodelled hearts are less amenable to protection against ischaemia/reperfusion. Understanding preservation of energy metabolism in diseased vs. healthy hearts may help to develop anti-ischaemic strategies effective also in jeopardized myocardium. Methods and results Isolated infarct-remodelled/sham Sprague-Dawley rat hearts were perfused in the working mode and subjected to 15 min of ischaemia and 30 min of reperfusion. Protection of post-ischaemic ventricular work was achieved by pharmacological conditioning with sevoflurane. Oxidative metabolism was measured by substrate flux in fatty acid and glucose oxidation using [3H]palmitate and [14C]glucose. Mitochondrial oxygen consumption was measured in saponin-permeabilized left ventricular muscle fibres. Activity assays of citric acid synthase, hydroxyacyl-CoA dehydrogenase, and pyruvate dehydrogenase and mass spectrometry for acylcarnitine profiling were also performed. Six weeks after coronary artery ligation, the hearts exhibited macroscopic and molecular signs of hypertrophy consistent with remodelling and limited respiratory chain and citric acid cycle capacity. Unprotected remodelled hearts showed a marked decline in palmitate oxidation and acetyl-CoA energy production after ischaemia/reperfusion, which normalized in sevoflurane-protected remodelled hearts. Protected remodelled hearts also showed higher β-oxidation flux as determined by increased oxygen consumption with palmitoylcarnitine/malate in isolated fibres and a lower ratio of C16:1+C16OH/C14 carnitine species, indicative of a higher long-chain hydroxyacyl-CoA dehydrogenase activity. Remodelled hearts exhibited higher PPARα-PGC-1α but defective HIF-1α signalling, and conditioning enabled them to mobilize fatty acids from endogenous triglyceride stores, which closely correlated with improved recovery. Conclusions Protected infarct-remodelled hearts secure post-ischaemic energy production by activation of β-oxidation and mobilization of fatty acids from endogenous triglyceride store

    Mal/SRF Is Dispensable for Cell Proliferation in Drosophila

    The Mal/SRF transcription factor is regulated by the level of G-actin in cells and has important roles in cell migration and other actin-dependent processes in Drosophila. A recent report suggests that Mal/SRF and an upstream regulator, Pico, are required for cell proliferation and tissue growth in Drosophila. I find otherwise. Mutation of Mal or SRF does not affect cell proliferation in the fly wing. Furthermore, I cannot reproduce the reported effects of Pico RNAi or Pico overexpression on body size. Nevertheless, I can confirm that overexpression of Pico or Mal causes tissue overgrowth specifically in the fly wing - where SRF is most highly expressed. My results indicate that Mal/SRF can promote tissue growth when abnormally active, but is not normally required for tissue growth during development

    Mechanical Stress Inference for Two Dimensional Cell Arrays

    Many morphogenetic processes involve mechanical rearrangement of epithelial tissues that is driven by precisely regulated cytoskeletal forces and cell adhesion. The mechanical state of the cell and intercellular adhesion are not only the targets of regulation, but are themselves likely signals that coordinate developmental process. Yet, because it is difficult to directly measure mechanical stress {\it in vivo} on sub-cellular scale, little is understood about the role of mechanics of development. Here we present an alternative approach which takes advantage of the recent progress in live imaging of morphogenetic processes and uses computational analysis of high resolution images of epithelial tissues to infer relative magnitude of forces acting within and between cells. We model intracellular stress in terms of bulk pressure and interfacial tension, allowing these parameters to vary from cell to cell and from interface to interface. Assuming that epithelial cell layers are close to mechanical equilibrium, we use the observed geometry of the two dimensional cell array to infer interfacial tensions and intracellular pressures. Here we present the mathematical formulation of the proposed Mechanical Inverse method and apply it to the analysis of epithelial cell layers observed at the onset of ventral furrow formation in the {\it Drosophila} embryo and in the process of hair-cell determination in the avian cochlea. The analysis reveals mechanical anisotropy in the former process and mechanical heterogeneity, correlated with cell differentiation, in the latter process. The method opens a way for quantitative and detailed experimental tests of models of cell and tissue mechanics
