    Molecular cloning and expression of a cold-adapted lipase gene from an antarctic deep sea psychrotrophic bacterium Pseudomonas sp. 7323

The final version is available from Springer via the DOI in this record .A psychrotrophic bacterium producing a cold-adapted lipase was isolated from the deep-sea sediment of Prydz Bay, Antarctic and identified as a Pseudomonas strain. Determination of the nucleotide sequence of the gene encoding a lipase from Pseudomonas sp. 7323 (lipA) revealed that LipA is composed of 617 amino acid residues with a calculated molecular weight of 64,466 Da. LipA has a GXSXG motif, which is conserved in lipases/esterases and generally contains the active-site serine. The lipase purified from the Escherichia coli transformant (rLipA) by metal-chelating chromatography exhibited the same electrophoretic mobility as did the wild-type lipase (wLipA) purified from strain 7323, and both enzymes were quite similar in physicochemical properties. The optimal temperature and pH value for the lipases activity were 30°C and 9.0, respectively. They were unstable at temperatures above 25°C and only retained half of their highest activity after incubation at 60°C for 5 min. These results indicated that the enzymes were typical alkaline cold-adapted enzymes. Both enzymes were particularly activated by Ca2+.     Purification and characterization of a cold-adapted α-amylase produced by Nocardiopsis sp. 7326 isolated from Prydz Bay, Antarctic

The final version is available from Springer via the DOI in this record. An actinomycete strain 7326 producing cold-adapted α-amylase was isolated from the deep sea sediment of Prydz Bay, Antarctic. It was identified as Nocardiopsis based on morphology, 16S rRNA gene sequence analysis, and physiological and biochemical characteristics. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and zymogram activity staining of purified amylase showed a single band equal to a molecular mass of about 55 kDa. The optimal activity temperature of Nocardiopsis sp. 7326 amylase was 35°C, and the activity decreased dramatically at temperatures above 45°C. The enzyme was stable between pH 5 and 10, and exhibited a maximal activity at pH 8.0. Ca2+, Mn2+, Mg2+, Cu2+, and Co2+stimulated the activity of the enzyme significantly, and Rb2+, Hg2+, and EDTA inhibited the activity. The hydrolysates of soluble starch by the enzyme were mainly glucose, maltose, and maltotriose.     Prepulse Inhibition of Auditory Cortical Responses in the Caudolateral Superior Temporal Gyrus in Macaca mulatta

    Prepulse inhibition (PPI) refers to a decreased response to a startling stimulus when another weaker stimulus precedes it. Most PPI studies have focused on the physiological startle reflex and fewer have reported the PPI of cortical responses. We recorded local field potentials (LFPs) in four monkeys and investigated whether the PPI of auditory cortical responses (alpha, beta, and gamma oscillations and evoked potentials) can be demonstrated in the caudolateral belt of the superior temporal gyrus (STGcb). We also investigated whether the presence of a conspecific, which draws attention away from the auditory stimuli, affects the PPI of auditory cortical responses. The PPI paradigm consisted of Pulse-only and Prepulse + Pulse trials that were presented randomly while the monkey was alone (ALONE) and while another monkey was present in the same room (ACCOMP). The LFPs to the Pulse were significantly suppressed by the Prepulse thus, demonstrating PPI of cortical responses in the STGcb. The PPI-related inhibition of the N1 amplitude of the evoked responses and cortical oscillations to the Pulse were not affected by the presence of a conspecific. In contrast, gamma oscillations and the amplitude of the N1 response to Pulse-only were suppressed in the ACCOMP condition compared to the ALONE condition. These findings demonstrate PPI in the monkey STGcb and suggest that the PPI of auditory cortical responses in the monkey STGcb is a pre-attentive inhibitory process that is independent of attentional modulation.Peer reviewe

    Consolidation effect of visual function training on children with different degrees of amblyopia

    AIM: To observe the consolidation effect of visual function training in amblyopic therapy on children with amblyopia of different degrees(mild, moderate and severe)which had been normalized. <p>METHODS: Totally 78 amblyopic children were divided into two groups: visual function training group(<i>n</i>=36, 70 eyes)and control group(<i>n</i>=42, 67 eyes). The rollback situation of the two groups in 6, 12, 24 and 36 months were observed after visual acuity reached 0.9 during treatment.<p>RESULTS: The rollback rates were both 0 in visual function training group and control group with different degrees of amblyopia after 6 months. There was no significant difference in rollback rate between the 2 groups with different degrees of amblyopia after 6 and 12 months. There was no significant difference in rollback rate between the two groups with mild amblyopia after 24 and 36 months(<i>P</i>=0.269, 0.269). However, the rollback rate in training group with moderate amblyopia was significantly different from in control group after 24 and 36 months(<i>P</i>=0.004, 0.002). There was no significant difference in rollback rate between the two groups with severe amblyopia after 24 and 36 months. <p>CONCLUSION: Visual function training can reduce the rollback rate and consolidate the effect of amblyopic treatment effectively for children with moderate amblyopia. However, the effect is not as good for children with mild and severe amblyopia

    Digitalization of regional public health information is an irreversible trend in the course of China\u27s social development. This is also one of the key tasks to reform China\u27s medicine and public health system. To learn more about the influence of using information technology for medical personnel and hospital performance, this study reviews in the personal perspective to know more about the sustainable action of using IT in hospitals based on the Theory of Planned Bahavior. We proposed a research model which expanded TPB and tested it based on 278 valid data collected from employees in a large-scale hospital in Anhui Province, East China. The results of study reveal: 1.We verified TPB in the Chinese context and found that attitude and subjective norm not only have a directly influence on behavioral intention, but also can influence the use behavior.2.The habit of using IT of medical personnel has a directly influence on the attitude of using IT, and it also has significant influence on the behavioral intention. Our research model discovered the relationship between subjective factors of IT users’ and their behaviour. The founding of this study not only can provide references regarding the direction of public hospitals reform in China for decision makers of hospitals, but also is very helpful for management teams in hospitals to adopt more rational management measures for better IT use among medical personnel, and better hospital performance
