    Age-related Resistance in Bell Pepper to \u3ci\u3eCucumber mosaic virus \u3c/i\u3e

    We demonstrated the occurrence of mature plant resistance in Capsicum annuum \u27Early Calwonder\u27 to Cucumber mosaic virus (CMV) under greenhouse conditions. When Early Cal wonder plants were sown at 10 day intervals and transplanted to 10-cm square pots, three distinct plant sizes were identified that were designated small, medium and large. Trials conducted during each season showed that CMV accumulated in inoculated leaves of all plants of each size category. All small plants (with the exception of the winter trial) developed a systemic infection that included accumulation of CMV in uninoculated leaves and severe systemic symptoms. Medium plants had a range of responses that included no systemic infection to detection of CMV in uninoculated leaves with the systemically infected plants being either symptomless or expressing only mild symptoms. None of the large plants contained detectable amounts of CMV in uninoculated leaves or developed symptoms. When plants were challenged by inoculation of leaves positioned at different locations along the stem or different numbers of leaves were inoculated, large plants continued to accumulate CMV in inoculated leaves but no systemic infection was observed. When systemic infection of large plants did occur, e.g. when CMV-infected pepper was used as a source of inoculum, virus accumulation in uninoculated leaves was relatively low and plants remained symptomless. A time-course study of CMV accumulation in inoculated leaves revealed no difference between small and large plants. Analyses to examine movement of CMV into the petiole of inoculated leaves and throughout the stem showed a range in the extent of infection. While all large plants contained CMV in inoculated leaves, some had no detectable amounts of virus beyond the leaf blade, whereas others contained virus throughout the length of the stem but with limited accumulation relative to controls

    TeV gamma-rays from photo-disintegration/de-excitation of cosmic-ray nuclei

    It is commonly assumed that high-energy gamma-rays are made via either purely electromagnetic processes or the hadronic process of pion production, followed by decay. We investigate astrophysical contexts where a third process (A*) may dominate, namely the photo-disintegration of highly boosted nuclei followed by daughter de-excitation. Starbust regions such as Cygnus OB2 appear to be promising sites for TeV gamma-ray emission via this mechanism. A unique feature of the A* process is a sharp energy minimum ~ 10 TeV/(T/eV) for gamma-ray emission from a thermal region of temperature T. We also check that a diffuse gamma-ray component resulting from the interaction of a possible extreme-energy cosmic-ray nuclei with background radiation is well below the observed EGRET data. The A* mechanism described herein offers an important contribution to gamma-ray astronomy in the era of intense observational activity.Comment: To be published in Phys. Rev. Let

    Next-to-Minimal Supersymmetric Model Higgs Scenarios for Partially Universal GUT Scale Boundary Conditions

    We examine the extent to which it is possible to realize the NMSSM "ideal Higgs" models espoused in several papers by Gunion et al in the context of partially universal GUT scale boundary conditions. To this end we use the powerful methodology of nested sampling. We pay particular attention to whether ideal-Higgs-like points not only pass LEP constraints but are also acceptable in terms of the numerous constraints now available, including those from the Tevatron and BB-factory data, (g−2)ÎŒ(g-2)_\mu and the relic density Ωh2\Omega h^2. In general for this particular methodology and range of parameters chosen, very few points corresponding to said previous studies were found, and those that were found were at best 2σ2\sigma away from the preferred relic density value. Instead, there exist a class of points, which combine a mostly singlet-like Higgs with a mostly singlino-like neutralino coannihilating with the lightest stau, that are able to effectively pass all implemented constraints in the region 80<mh<10080<m_h<100. It seems that the spin-independent direct detection cross section acts as a key discriminator between ideal Higgs points and the hard to detect singlino-like points.Comment: 22 pages, 15 figure

    SREBP1 regulates mitochondrial metabolism in oncogenic KRAS expressing NSCLC [preprint]

    Cancer cells require extensive metabolic reprogramming in order to provide the bioenergetics and macromolecular precursors needed to sustain a malignant phenotype. Mutant KRAS is a driver oncogene that is well known for its ability to regulate the ERK and PI3K signaling pathways. However, it is now appreciated that KRAS can promote tumor growth via upregulation of anabolic metabolism. We recently showed that oncogenic KRAS promotes a gene expression program of de novo lipogenesis in non-small cell lung cancer (NSCLC). To define the mechanism(s) responsible, we focused on the lipogenic transcription factor SREBP1. We observed that KRAS increases SREBP1 expression and genetic knockdown of SREBP1 significantly inhibited cell proliferation of mutant KRAS-expressing cells. Unexpectedly, lipogenesis was not significantly altered in cells subject to SREBP1 knockdown. Carbon tracing metabolic studies showed a significant decrease in oxidative phosphorylation and RNA-seq data revealed a significant decrease in mitochondrial encoded subunits of the electron transport chain (ETC). Taken together, these data support a novel role, distinct from lipogenesis, of SREBP1 on mitochondrial function in mutant KRAS NSCLC

    Dissecting the role of histidine kinase and Hog1 mitogen-activated protein kinase signalling in stress tolerance and pathogenicity of Parastagonospora nodorum on wheat.

    The HOG1 mitogen-activated protein kinase (MAPK) pathway is activated through two-component histidine kinase (HK) signalling. This pathway was first characterized in the budding yeast Saccharomyces cerevisiae as a regulator of osmotolerance. The fungus Parastagonospora nodorum is the causal agent of septoria nodorum blotch of wheat. This pathogen uses host-specific effectors in tandem with general pathogenicity mechanisms to carry out its infection process. Genes showing strong sequence homology to S. cerevisiae HOG1 signalling pathway genes have been identified in the genome of P. nodorum. In this study, we examined the role of the pathway in the virulence of P. nodorum on wheat by disrupting putative pathway component genes: HOG1 (SNOG_13296) MAPK and NIK1 (SNOG_11631) hybrid HK. Mutants deleted in NIK1 and HOG1 were insensitive to dicarboximide and phenylpyrrole fungicides, but not a fungicide that targets ergosterol biosynthesis. Furthermore, both Δnik1 and Δhog1 mutants showed increased sensitivity to hyperosmotic stress. However, HOG1, but not NIK1, is required for tolerance to elevated temperatures. HOG1 deletion conferred increased tolerance to 6-methoxy-2-benzoxazolinone, a cereal phytoalexin. This suggests that the HOG1 signalling pathway is not exclusively associated with NIK1. Both Δnik1 and Δhog1 mutants retained the ability to infect and cause necrotic lesions on wheat. However, we observed that the Δhog1 mutation resulted in reduced production of pycnidia, asexual fruiting bodies that facilitate spore dispersal during late infection.     Measurement of the core-collapse progenitor mass distribution of the Small Magellanic Cloud

    The physics of core-collapse (CC) supernovae (SNe) and how the explosions depend on progenitor properties are central questions in astronomy. For only a handful of SNe, the progenitor star has been identified in pre-explosion images. Supernova remnants (SNRs), which are observed long after the original SN event, provide a unique opportunity to increase the number of progenitor measurements. Here, we systematically examine the stellar populations in the vicinities of 23 known SNRs in the Small Magellanic Cloud (SMC) using the star formation history (SFH) maps of Harris & Zaritsky (2004). We combine the results with constraints on the SNR metal abundances and environment from X-ray and optical observations. We find that 22 SNRs in the SMC have local SFHs and properties consistent with a CC explosion, several of which are likely to have been high-mass progenitors. This result supports recent theoretical findings that high-mass progenitors can produce successful explosions. We estimate the mass distribution of the CC progenitors and find that this distribution is similar to a Salpeter IMF (within the uncertainties), while this result is shallower than the mass distribution found in M31 and M33 by Jennings et al. (2014) and D\'{\i}az-Rodr\'{\i}guez et al. (2018) using a similar approach. Additionally, we find that a number of the SMC SNRs exhibit a burst of star formation between 50-200 Myr ago. As these sources are likely CC, this signature may be indicative of massive stars undergoing delayed CC as a consequence of binary interaction, rapid rotation, or low metallicity. In addition, the lack of Type Ia SNRs in the SMC is possibly a result of the short visibility times of these sources as they may fall below the sensitivity limits of current radio observations.Comment: Accepted to ApJ, 24 pages, 7 Figures, 3 Table

    Variability in concentrations of potentially toxic elements in urban parks from six European cities

    Use of a harmonised sampling regime has allowed comparison of concentrations of copper, chromium, nickel, lead and zinc in six urban parks located in different European cities differing markedly in their climate and industrial history. Wide concentrations ranges were found for copper, lead and zinc at most sites, but for chromium and nickel a wide range was only seen in the Italian park, where levels were also considerably greater than in other soils. As might be expected, the soils from older cities with a legacy of heavy manufacturing industry (Glasgow, Torino) were richest in potentially toxic elements (PTEs); soils from Ljubljana, Sevilla and Uppsala had intermediate metal contents, and soils from the most recently established park, in the least industrialised city (Aveiro), displayed lowest concentrations. When principal component analysis was applied to the data, associations were revealed between pH and organic carbon content; and between all five PTEs. When pH and organic carbon content were excluded from the PCA, a distinction became clear between copper, lead and zinc (the "urban" metals) on the one hand, and chromium and nickel on the other. Similar results were obtained for the surface (0-10 cm depth) and sub-surface (10-20 cm depth) samples. Comparisons with target or limit concentrations were limited by the existence of different legislation in different countries and the fact that few guidelines deal specifically with public-access urban soils intended for recreational use

    Dynamic earthquake triggering response tracks evolving unrest at Sierra Negra volcano, GalĂĄpagos Islands

    The propensity for dynamic earthquake triggering is thought to depend on the local stress state and amplitude of the stress perturbation. However, the nature of this dependency has not been confirmed within a single crustal volume. Here, we show that at Sierra Negra volcano, GalĂĄpagos Islands, the intensity of dynamically triggered earthquakes increased as inflation of a magma reservoir elevated the stress state. The perturbation of short-term seismicity within teleseismic surface waves also increased with peak dynamic strain. Following rapid coeruptive subsidence and reduction in stress and background seismicity rates, equivalent dynamic strains no longer triggered detectable seismicity. These findings offer direct constraints on the primary controls on dynamic triggering and suggest that the response to dynamic stresses may help constrain the evolution of volcanic unrest

    Podoconiosis in East and West Gojam Zones, Northern Ethiopia

    Background: Podoconiosis is a neglected tropical disease (NTD) that is prevalent in red clay soil-covered highlands of tropical Africa, Central and South America, and northern India. It is estimated that up to one million cases exist in Ethiopia. This study aimed to estimate the prevalence of podoconiosis in East and West Gojam Zones of Amhara Region in northern Ethiopia. Methodology/Principal Findings: A cross-sectional household survey was conducted in Debre Eliyas and Dembecha woredas (districts) in East and West Gojam Zones, respectively. The survey covered all 17,553 households in 20 kebeles (administrative subunits) randomly selected from the two woredas. A detailed structured interview was conducted on 1,704 cases of podoconiosis identified in the survey. Results: The prevalence of podoconiosis in the population aged 15 years and above was found to be 3.3% (95% CI, 3.2% to 3.6%). 87% of cases were in the economically active age group (15–64 years). On average, patients sought treatment five years after the start of the leg swelling. Most subjects had second (42.7%) or third (36.1%) clinical stage disease, 97.9% had mossy lesions, and 53% had open wounds. On average, patients had five episodes of acute adenolymphangitis (ALA) per year and spent a total of 90 days per year with ALA. The median age of first use of shoes and socks were 22 and 23 years, respectively. More men than women owned more than one pair of shoes (61.1% vs. 50.5%; x2 = 11.6 p = 0.001). At the time of interview, 23.6% of the respondents were barefoot, of whom about two-thirds were women. Conclusions: This study showed high prevalence of podoconiosis and associated morbidities such as ALA, mossy lesions and open wounds in northern Ethiopia. Predominance of cases at early clinical stage of podoconiosis indicates the potential for reversing the swelling and calls for disease prevention interventions
