    Combined effects of salicylic acid and furosemide and noise on hearing

    <p>Abstract</p> <p>Background</p> <p>A major cause of the hearing loss following exposure to intense noise involves release of free radicals resulting from the elevated metabolism. The free radicals induce damage to several of the components of the cochlear amplifier including the outer hair cells and indirectly to the transduction currents. Salicylic acid induces a reversible hearing loss since it binds to the motor protein prestin in the outer hair cells, reducing electromotility. Furosemide also induces a reversible hearing loss since it reduces the endocochlear potential which is a major component of the cochlear transduction currents. On the other hand, each of these drugs also provides protection from a noise induced hearing loss if they are injected just before a noise exposure, probably as a result of the decreased metabolism induced in their presence, with release of lower levels of free radicals. In this study, both drugs were administered in order to assess whether their protective effects would be additive.</p> <p>Methods</p> <p>The study was conducted on normal hearing albino mice of the Sabra strain. They were injected with either salicylic acid alone (N = 11), or furosemide alone (N = 14), or both together (N = 14), or with saline control (N = 11) and exposed to broad band noise for 3.5 hours. An additional group of 9 mice was injected with both salicylic acid and furosemide, but not exposed to noise. The degree of the resulting hearing loss was assessed by recording thresholds of the auditory nerve brainstem evoked responses to broad band clicks before the injections and noise, and 7, 14 and 21 days after.</p> <p>Results</p> <p>The noise induced hearing loss in the mice injected with salicylic acid alone or furosemide alone was smaller than in those injected with saline, i.e. these drugs provided protection, as in previous studies in this laboratory. There was no threshold elevation after two weeks in the mice injected with both drugs without noise exposure, i.e. the effects of the two drugs given together was reversible. On the other hand, there was a significant hearing loss (i.e. threshold elevation) in the group which received both drugs and was also exposed to noise, with mean threshold elevations of 38.8 ± 19.0 dB and 28.3 ± 11.7 dB 7 days after noise exposure.</p> <p>Conclusions</p> <p>This result is very surprising, if not paradoxical. Drugs which provide protection from a noise induced hearing loss when administered alone, not only do not provide protection when given together, but also induce a greater hearing loss when accompanied by noise. This observation may be related to the finding that the depression of the endocochlear potential normally caused by furosemide is reduced in the presence of salicylic acid, so that the protection usually provided by furosemide is not present when it is administered together with salicylic acid. Thus it seems that each drug may interfere with the protective action of the other when coupled with noise.</p

    Weight Stigmatization Among Hispanic American Children

    This study was designed to examine weight stigmatization among Hispanic American children. Fifty-five fifth grade students from a large, urban school district in Southern California were asked to rank six samesex drawings of children with various physical characteristics (related to weight or disability) in order of friend preference (1 = the most preferred, and 6 = the least preferred friend). Positive and negative adjectives were then assigned to the average-weight and obese drawings using the Adjective Checklist (ACL). The majority of the participants (60%) chose the average-weight child as the most preferred and 46% identified the obese child as the least preferred friend. In addition, the average-weight child was assigned more positive and fewer negative adjectives compared to the obese child. Significant differences in ACL composite scores between normal weight and overweight drawings were also found (p = 0.00). It appears that weight stigmatization is present in the current sample, which suggests that Hispanic children living in the U.S. may adopt negative attitudes about weight that are similar to American culture

    Geologic and Structural Evolution of the NE Lau Basin, Tonga: Morphotectonic Analysis and Classification of Structures Using Shallow Seismicity

    The transition from subduction to transform motion along horizontal terminations of trenches is associated with tearing of the subducting slab and strike-slip tectonics in the overriding plate. One prominent example is the northern Tonga subduction zone, where abundant strike-slip faulting in the NE Lau back-arc basin is associated with transform motion along the northern plate boundary and asymmetric slab rollback. Here, we address the fundamental question: how does this subduction-transform motion influence the structural and magmatic evolution of the back-arc region? To answer this, we undertake the first comprehensive study of the geology and geodynamics of this region through analyses of morphotectonics (remote-predictive geologic mapping) and fault kinematics interpreted from ship-based multibeam bathymetry and Centroid-Moment Tensor data. Our results highlight two unique features of the NE Lau Basin: (1) the occurrence of widely distributed off-axis volcanism, in contrast to typical ridge-centered back-arc volcanism, and (2) fault kinematics dominated by shallow-crustal strike slip-faulting (rather than normal faulting) extending over ~120 km from the transform boundary. The orientations of these strike-slip faults are consistent with reactivation of earlier-formed normal faults in a sinistral megashear zone. Notably, two distinct sets of Riedel megashears are identified, indicating a recent counter-clockwise rotation of part of the stress field in the back-arc region closest to the arc. Importantly, these structures directly control the development of complex volcanic-compositional provinces, which are characterized by variably-oriented spreading centers, off-axis volcanic ridges, extensive lava flows, and point-source rear-arc volcanoes that sample a heterogenous mantle wedge, with sharp gradients and contrasts in composition and magmatic affinity. This study adds to our understanding of the geologic and structural evolution of modern backarc systems, including the association between subduction-transform motions and the siting and style of seafloor volcanism

    Experiential education and outreach based on nearshore monitoring of the Elwha River restoration project

    Nearshore monitoring of benthic habitats and the coastal environment following the Elwha River Restoration project has engaged students and citizens with coastal science and management issues. In the post-dam-removal period, the lessons learned will continue to be disseminated via a UW undergraduate course and an interactive digital map, both designed to engage students and communities in restoration science. The research-focused course developed at the UW Friday Harbor Labs has allowed us to engage diverse undergraduate students (and graduate teaching assistants) in the research process. The course integrates interdisciplinary lectures and workshops on data analysis and laboratory methods, with the research process; from proposal to oceanographic data collection to analysis to publication. The course provides opportunities for student creativity and leadership. Outcome tracking indicates that these undergraduate (and post-bac) students are generally attending graduate school at a high rate, and launching careers in education, coastal management, and other STEM fields. To engage a broader segment of the community and to support decision-making about large-scale coastal restoration projects, we have developed an interactive digital map that will be available on-line, and will also be piloted as a physical interpretive display at the Feiro Marine Life Center in Port Angeles, WA. The interactive digital map is designed to effectively tell the story of the Elwha restoration in the coastal environment through the compilation and display of multiple data sets, some of which have never before been publicly available. Ultimately, the result of long-term monitoring of the Elwha nearshore system will provide a better understanding of the effects of restoration activities, such as dam removal on benthic habitats, and this knowledge will be passed to future managers and citizens through educational and outreach activities that captivate and inspire a broad audience

    Changing the Principal Supervisor Role to Better Support Principals: Evidence from the Principal Supervisor Initiative

    The principal supervisor job has traditionally revolved around administration, operations and compliance, but as principals have increasingly been called on in recent years to concentrate on supporting high-quality teaching, the idea of a complementary makeover of the supervisor job has gained attention. In 2014, with funding from The Wallace Foundation, six large school districts around the country embarked on a four-year, $24-million initiative to redesign the supervisor position so it focused primarily on supporting principals in their role as instructional leaders.This report, which looks at the final year and the effects of the Principal Supervisor Initiative, concludes that the effort succeeded in changing the job so that it centered on developing and evaluating principals to help them promote effective teaching and learning in their schools. Over the course of the initiative, principals' ratings of their supervisors' effectiveness rose from 3.88 to 4.10 on a scale of 1-to-5, a statistically significant increase. Principals reported greater frequency of supervisor practices to develop school leadership—such as helping principals with data analysis, providing them with useful feedback and working with them to assess teacher effectiveness

    Changes in Kelp and Other Seaweeds Following Elwha Dam Removal

    Kelps are ecologically important seaweeds that dominated the nearshore vegetation community prior to dam removal on the Elwha River. Dam removal is expected to trigger a shift from kelps to vegetation types that are characteristic of soft-sediment communities through restoring natural sediment supply. This study is investigating how nearshore vegetation responds to restoration of the natural sediment regime, both initially when large amounts of sediment entrained in the reservoirs are released and over longer time periods. We assessed vegetation at multiple spatial scales using three approaches. First, we measured floating kelp canopy area using aerial photography. Second, we assessed the abundance of understory kelp and seagrasses with towed videography along 50 km of shoreline. Third, scuba divers recorded density of kelp species and other seaweeds along 10 km of shoreline bracketing the river mouth. Results show profound changes in vegetation and a strong gradient in magnitude of impact related to distance from the river mouth. Floating kelp canopy area decreased 74% in the Elwha Drift Cell in the first year following project initiation (year 1), with lower magnitude losses throughout the Strait of Juan de Fuca. Area of prostrate kelps decreased by 45% (400 ha) and of stipitate kelps by 30% (130 ha) in the Elwha Drift Cell in year 1. Mean kelp density near the river mouth decreased 77% in year 1 and 95% in year 2. While all 10 kelp species declined, annuals were more impacted than perennials. In contrast to the general decline, juveniles of several kelp species appeared in late August of year 2, a substantial delay compared to typical spring timing of juvenile growth. What caused the large kelp losses and apparent delay of juvenile growth? Likely candidates include light reduction from the river plume and scour, burial or settlement inhibition from deposition. These candidates will be explored using physical data from the multidisciplinary research effort

    Nutritional and metabolic status of children with autism vs. neurotypical children, and the association with autism severity

    <p>Abstract</p> <p>Background</p> <p>The relationship between relative metabolic disturbances and developmental disorders is an emerging research focus. This study compares the nutritional and metabolic status of children with autism with that of neurotypical children and investigates the possible association of autism severity with biomarkers.</p> <p>Method</p> <p>Participants were children ages 5-16 years in Arizona with Autistic Spectrum Disorder (n = 55) compared with non-sibling, neurotypical controls (n = 44) of similar age, gender and geographical distribution. Neither group had taken any vitamin/mineral supplements in the two months prior to sample collection. Autism severity was assessed using the Pervasive Development Disorder Behavior Inventory (PDD-BI), Autism Treatment Evaluation Checklist (ATEC), and Severity of Autism Scale (SAS). Study measurements included: vitamins, biomarkers of vitamin status, minerals, plasma amino acids, plasma glutathione, and biomarkers of oxidative stress, methylation, sulfation and energy production.</p> <p>Results</p> <p>Biomarkers of children with autism compared to those of controls using a t-test or Wilcoxon test found the following statistically significant differences (p < 0.001): Low levels of biotin, plasma glutathione, RBC SAM, plasma uridine, plasma ATP, RBC NADH, RBC NADPH, plasma sulfate (free and total), and plasma tryptophan; also high levels of oxidative stress markers and plasma glutamate. Levels of biomarkers for the neurotypical controls were in good agreement with accessed published reference ranges. In the Autism group, mean levels of vitamins, minerals, and most amino acids commonly measured in clinical care were within published reference ranges.</p> <p>A stepwise, multiple linear regression analysis demonstrated significant associations between several groups of biomarkers with all three autism severity scales, including vitamins (adjusted R<sup>2 </sup>of 0.25-0.57), minerals (adj. R<sup>2 </sup>of 0.22-0.38), and plasma amino acids (adj. R<sup>2 </sup>of 0.22-0.39).</p> <p>Conclusion</p> <p>The autism group had many statistically significant differences in their nutritional and metabolic status, including biomarkers indicative of vitamin insufficiency, increased oxidative stress, reduced capacity for energy transport, sulfation and detoxification. Several of the biomarker groups were significantly associated with variations in the severity of autism. These nutritional and metabolic differences are generally in agreement with other published results and are likely amenable to nutritional supplementation. Research investigating treatment and its relationship to the co-morbidities and etiology of autism is warranted.</p
