    Performance based earthquake assessment of an industrial silos structure and retrofit with sliding isolators

    Recent seismic events pointed out the high vulnerability of existing industrial facilities, stressing on safety and high losses inherent to interruption of economic activities and release of environmentally hazardous materials. These structures often have irregular geometry and structural configuration, are subject to aging and corrosion, and are designed without specific performance-based or seismic design criteria. Due to these inherent complexities, retrofit using friction isolators can be a viable and practical solution for performance improvements. This work presents a case study of irregular industrial storage plant structure consisting of a group of six elevated silos resting on a steel frame on one side and connected to a vaulted RC structure on the other. A computational model is built incorporating nonlinearities from the components (braces, beams, columns, etc.) and from the mitigation devices. Retrofit using friction isolators is analyzed and evaluated through linear and nonlinear dynamic analyses under a set of natural ground motions. Results show the effectiveness of the mitigation strategy in terms of performance improvement

    First report of Leptosphaeria biglobosa ‘brassicae’ as causal agent of phoma leaf spot in Brassica napus (Canola) in Argentina

    Canola(Brassicanapus L.) is the second largest oilseed crop in the worldproviding 13% of the world´s oil supply. This crop has been grown in Argentinasince the 1930s, and the area devoted to its cultivation varies every year,reaching a maximum of 95000 Ha in the 2012/2013 growing season. Because of theoccurrence of optimal weather conditions and soils with high fertility, theaverage yield in this region is about 2000 kg/Ha. Phoma leaf spot and Phomastem canker are considered the most important and devastating diseases in Brassica napus andother Brassicaespecies [1]. In both cases, the causal agent is a complexof two closely related fungal species, Leptosphaeria maculans and L. biglobosa. In Argentina,the presence of L. maculans incanola plants was reported for the first time in 2004 [2], but the existence ofL.biglobosa has not been recorded so far. During the 2015/2016season, we collected several samples with typical Phoma leaf spot symptoms fromcanola plants growing in fields from the north and northeastern regions of the Buenos Aires province.The necrotic lesions were circular to irregularly oval, 8 to 15 mm in diameter,pale brown in the center, grayish green at the margin and characterized withthe presence of pycnidia. Several leaf pieces with lesions were rinsed twicewith deionized sterile water and placed in a humid chamber (90 mm diameterPetri dish with a layer of filter paper soaked in deionized sterile water) during2-3 days to induce the pycnidia to exude cirri of conidia. After this period,one cirrus per sample was transferred onto PDA plates supplemented withantibiotics (15 mg/L streptomycin, 15 mg/L gentamicin and 12 mg/L tetracycline)using an inoculation needle under stereoscopic microscope. Thus, severalisolates were obtained, some of them showing rapid mycelial growth rate andpigment production on PDA medium, as showed by the isolate Tapidor of L. biglobosa thatwe used as control (kindly provided by Professor Bruce Fitt, University ofHertfordshire-UK). In order to confirm the identity of these isolates, a PCRassay using genomic DNA as template was performed to distinguish L. maculans from L. biglobosa withthe species-specific primers LmacR, LmacF,and LbigF ina three-primers strategy described by Gaetan (2005)[3]. These reactions gave a444-bp amplicon as expected for L. biglobosa ´brassicae´.In addition, these results were confirmed by sequencing the nuclear ribosomalinternal transcribed spacer (ITS) region, which showed a 99% of identity withthe sequence of L. biglobosa ´brassicae´at the GenBank database (FO905468). L. biglobosa isolateswere then tested for pathogenicity on the canola cultivars Westar and Bioaureo2286 (Nuseed). With this purpose, cotyledons of 10-day-old seedlings werepricked with a needle, and each wound inoculated with 10 μl ofa conidial suspension (107 42conidia/ml). Sterilized distilled water was used as control. Developing primaryleaves were removed every 2-3 days in order to ensure that cotyledons continueto expand. Fourteen days after inoculation, irregular and brown necroticlesions were visible at the site of inoculation. These cotyledons were detachedand placed in a humid chamber to induce pycnidia formation. After three dayscirri of conidia were transferred to a plate with PDA supplemented with antibioticsas mentioned above. The identity of these isolates of L. biglobosa wereconfirmed by pigment production on PDA medium and by PCR assay usingspecies-specific primers. To our knowledge, this is the first report of L. biglobosa ´brassicae´as a pathogen of canola in Argentina. This finding shows that in Argentina´scanola cropping areas not 50 only L. maculans but alsoL.biglobosa are the causal agents of Phoma leaf spot disease.Fil: Rossi, Franco Rubén.     Role of perfusion machines in the setting of clinical liver transplantation. A qualitative systematic review

    Growing enthusiasm around machine perfusion (MP) in clinical liver transplantation (LT) may be the preamble for standardized practice to expand the donors' pool. The present systematic review investigated all the liver transplantations performed using grafts treated with MP. A systematic review of 309 papers was performed. Eventually, 27 articles were enrolled for the study. A total number of 173 cases was reported. Only 12 cohort studies were identified: the remaining ones were case reports or case series. Hypothermic machine perfusion was performed in 102 (59.0%), normothermic machine perfusion in 65 (37.6%), and controlled oxygenated rewarming in the remaining 6 (3.4%) cases. Donor characteristics, evaluation of graft quality and end-points were not homogeneous among the studies. Overall, post-LT results were excellent, with 1.2 and 4.0% of patients experienced primary non-function and ischemic-type biliary lesions, respectively

    A New Fast Silicon Photomultiplier Photometer

    The realization of low-cost instruments with high technical performance is a goal which deserves some efforts in an epoch of fast technological developments: indeed such instruments can be easily reproduced and therefore allow to open new research programs in several Observatories. We realized a fast optical photometer based on the SiPM technology, using commercially available modules. Using low-cost components we have developed a custom electronic chain to extract the signal produced by a commercial MPPC module produced by Hamamatsu, in order to obtain sub millisecond sampling of the light curve of astronomical sources, typically pulsars. In the early February 2011 we observed the Crab Pulsar at the Cassini telescope with our prototype photometer, deriving its period, power spectrum and shape of its light curve in very good agreement with the results obtained in the past with other instruments.Comment: Accepted for Publications of the Astronomical Society of Pacific (PASP), 8 pages, 8 figure

    Reactive oxygen species generated in chloroplasts contribute to tobacco leaf infection by the necrotrophic fungus Botrytis cinerea

    Reactive oxygen species (ROS) play fundamental roles in plant responses to pathogen infection, including modulation of cell death processes and defense-related gene expression. Cell death triggered as part of the hypersensitive response enhances resistance to biotrophic pathogens, but favors the virulence of necrotrophs. Even though the involvement of ROS in the orchestration of defense responses is well established, the relative contribution of specific subcellular ROS sources to plant resistance against microorganisms with different pathogenesis strategies is not completely known. The aim of this work was to investigate the role of chloroplastic ROS in plant defense against a typical necrotrophic fungus, Botrytis cinerea. For this purpose, we used transgenic Nicotiana tabacum (tobacco) lines expressing a plastid-targeted cyanobacterial flavodoxin (pfld lines), which accumulate lower chloroplastic ROS in response to different stresses. Tissue damage and fungal growth were significantly reduced in infected leaves of pfld plants, as compared with infected wild-type (WT) counterparts. ROS build-up triggered by Botrytis infection and associated with chloroplasts was significantly decreased (70–80%) in pfld leaves relative to the wild type. Phytoalexin accumulation and expression of pathogenesis-related genes were induced to a lower degree in pfld plants than in WT siblings. The impact of fungal infection on photosynthetic activity was also lower in pfld leaves. The results indicate that chloroplast-generated ROS play a major role in lesion development during Botrytis infection. This work demonstrates that the modulation of chloroplastic ROS levels by the expression of a heterologous antioxidant protein can provide a significant degree of protection against a canonical necrotrophic fungus.Fil: Rossi, Franco Rubén.     P02-187 Assessing initial patient's affective disposition towards the hospitalization and the treatment: A validation study

    The present work shows some preliminary results of a longitudinal study aimed at validating a psychometric instrument - the Subjective Distance Scale (SDS) - developed to assess the patient's affective nearness/remoteness towards/from the psychiatric treatment in a Day Hospital program. 55 day hospital patients were assessed at different moments of the treatment: At admission they were evaluated by means of the SDS, the SCL-90-R (patient's psychiatric symptoms) and the GAS (patient's mental health level). Institutional therapeutic alliance was assessed one week from admission (IWAI-p) and finally, patient's psychiatric symptoms were retested (SCL-90-R) at the end of the treatment and 3 months from discharge. The main results reveal good psychometric properties of the SDS: its factorial structure partially confirms the dimensions theoretically hypothesised; its internal consistency - total and most of its subscales - reach adequate reliability levels; and related to its predicted validity, the scale correlates with some important aspects of the treatment, like the quality of the early institutional alliance, the symptomatic improvement and the stability of the improvements in time. New studies with larger samples and conducted in additional psychiatric settings are necessary to guarantee the validity and reliability of the scale before it could be used as a clinical screening instrument

È di difficile diagnosi pre-operatoria e ha una prognosi sfavorevole in alcuni casi. Questo lavoro riporta il caso di un paziente di 94 anni con istiocitoma ad origine cutanea e sottolinea l’importanza della diagnosi precoce ai fini della curabilità
