    Simple versus optimal rules as guides to policy

    This paper contributes to the policy evaluation literature by developing new strategies to study alternative policy rules. We compare optimal rules to simple rules within canonical monetary policy models. In our context, an optimal rule represents the solution to an intertemporal optimization problem in which a loss function for the policymaker and an explicit model of the macroeconomy are specified. We define a simple rule to be a summary of the intuition policymakers and economists have about how a central bank should react to aggregate disturbances. The policy rules are evaluated under minimax and minimax regret criteria. These criteria force the policymaker to guard against a worst-case scenario, but in different ways. Minimax makes the worst possible model the benchmark for the policymaker, while minimax regret confronts the policymaker with uncertainty about the true model. Our results indicate that the case for a model-specific optimal rule can break down when uncertainty exists about which of several models is true. Further, we show that the assumption that the policymaker’s loss function is known can obscure policy trade-offs that exist in the short, medium, and long run. Thus, policy evaluation is more difficult once it is recognized that model and preference uncertainty can interact.

    This paper characterizes the frequency domain properties of feedback control rules in linear systems in order to better understand how different policies affect outcomes frequency by frequency. We are especially concerned in understanding how reductions of variance at some frequencies induce increases in variance at others. Tradeoffs of this type are known in the control literature as design limits. Design limits are important in understanding the full range of effects of macroeconomic stabilization policies. We extend existing results to account for discrete time bivariate systems with rational expectations. Application is made to the evaluation of monetary policy rules.

    SCoRS - a method based on stability for feature selection and mapping in neuroimaging.

    Feature selection (FS) methods play two important roles in the context of neuroimaging based classification: potentially increase classification accuracy by eliminating irrelevant features from the model and facilitate interpretation by identifying sets of meaningful features that best discriminate the classes. Although the development of FS techniques specifically tuned for neuroimaging data is an active area of research, up to date most of the studies have focused on finding a subset of features that maximizes accuracy. However, maximizing accuracy does not guarantee reliable interpretation as similar accuracies can be obtained from distinct sets of features. In the current paper we propose a new approach for selecting features: SCoRS (Survival Count on Random Subsamples) based on a recently proposed Stability Selection theory. SCoRS relies on the idea of choosing relevant features that are stable under data perturbation. Data are perturbed by iteratively subsampling both features (subspaces) and examples. We demonstrate the potential of the proposed method in a clinical application to classify depressed patients versus healthy individuals based on fMRI data acquired during visualization of happy faces

    PSMA-Specific CAR-Engineered T Cells Eradicate Disseminated Prostate Cancer in Preclinical Models.

    Immunology-based interventions have been proposed as a promising curative chance to effectively attack postoperative minimal residual disease and distant metastatic localizations of prostate tumors. We developed a chimeric antigen receptor (CAR) construct targeting the human prostate-specific membrane antigen (hPSMA), based on a novel and high affinity specific mAb. As a transfer method, we employed last-generation lentiviral vectors (LV) carrying a synthetic bidirectional promoter capable of robust and coordinated expression of the CAR molecule, and a bioluminescent reporter gene to allow the tracking of transgenic T cells after in vivo adoptive transfer. Overall, we demonstrated that CAR-expressing LV efficiently transduced short-term activated PBMC, which in turn were readily stimulated to produce cytokines and to exert a relevant cytotoxic activity by engagement with PSMA+ prostate tumor cells. Upon in vivo transfer in tumor-bearing mice, CAR-transduced T cells were capable to completely eradicate a disseminated neoplasia in the majority of treated animals, thus supporting the translation of such approach in the clinical setting

    Pushing the mass limit for intact launch and photoionization of large neutral biopolymers

    Since their first discovery by Louis Dunoyer and Otto Stern, molecular beams have conquered research and technology. However, it has remained an outstanding challenge to isolate and photoionize beams of massive neutral polypeptides. Here we show that femtosecond desorption from a matrix-free sample in high vacuum can produce biomolecular beams at least 25 times more efficiently than nanosecond techniques. While it has also been difficult to photoionize large biomolecules, we find that tailored structures with an abundant exposure of tryptophan residues at their surface can be ionized by vacuum ultraviolet light. The combination of these desorption and ionization techniques allows us to observe molecular beams of neutral polypeptides with a mass exceeding 20,000 amu. They are composed of 50 amino acids – 25 tryptophan and 25 lysine residues – and 26 fluorinated alkyl chains. The tools presented here offer a basis for the preparation, control and detection of polypeptide beams

    Oestrus behaviour and performance in vivo of Saanen goats raised in northeast of Brazil.

    Abstract: In order to verify the oestrus activity and to observe the body weight of Saanen goats, when reared in intensive condition of Northeast Brazil during four different seasons, ten adult Saanen goats were used. In this experiment four climatic seasons were considered; early rainy (ER), late rainy (LR), early dry (ED) and late dry (LD). Oestrus behaviour was monitored twice a day (8:00 a.m. and 4:00 p.m.) during all the experiment, using two vasectomised bucks. The oestrus cycles were quantified within each season and classified as short ( 25 days). The goats were weighed every seven days. During the late rainy season, a significant fall of the mean body weight of the animals was observed when compared to early rainy season. A larger percentage of goats in oestrus were observed during the late rainy season. Oestrus duration was longer in the late rainy and early dry season than in early rainy and late dry season. During the experiment, there were 12.0% of short cycles, 50.3% of normal cycles and 37.7% of long cycles. The percentages of abnormal cycles (short and long cycles), in relation to total cycles monitored in each season were 52, 58, 38 and 50% for early rainy, late rainy, early dry and late dry, respectively. From the results of this study it can be concluded that pen-fed Saanen goats do not show seasonality for oestrus behaviour. Rainfall appeared to be an extero-receptive factor influencing the quality of sexual activity and performance in these goats
