    Strategic planning optimisation of "Napoli Est" water distribution system

    The District Meter Areas (DMA) design is an innovative methodology of water networks management, based on the pressure patterns control and on the water flows monitoring, in order to reduce water losses and to optimize the water systems management. A District Meter Area is an area supplied from few water inputs, into which discharges can be easily measured to determine leaks. So, the DMA design represents an alternative to the traditional approach based on heavy looped distribution network. In the present paper the DMA design of the “Napoli Est” water distribution system (approximately 65.000÷70.000 customers), performed with the support of the Water Agency ARIN S.p.A., is discussed. After analysis of authorized consumption, by means of a monitoring campaign of water flows over the area, the system water balance was performed, showing significant water losses, as a consequence of high pressure patterns. This situation was confirmed by the high number of maintenance operations performed in the area during the year 2005. In order to characterize the piezometric heads on the network, ARIN S.p.A. supplied to the installation of six pressure transducers in the most vulnerable areas. The water level in the supply reservoir was also measured in order to estimate its influence on the network pressure heads. Hydraulic simulations were carried out with the EPANET software version 2.0 applied to a network layout resulted from the system “skeletonization”, achieved by eliminating out of order pipes, integrating pipelines of same diameter and roughness, replacing dead-end branches and small networks supplied by a single junction with an equivalent discharge. After the skeletonizated network was calibrated, several hypothesis of designing and implementing DMA to reduce physical losses were performed, providing adequate operating pressure of the system. Many numerical simulations were performed to guarantee adequate head pressure especially for peak hours demand, break of transmission mains and fire hydrant service. A chlorine residuals analysis was also effected, by simulating the transport and decay of chlorine through the network. District Meter Areas, therefore, were designed, and the corresponding hydraulic and water quality investigations and simulations were carried out. Six District Meter Areas were planned, assembling 14 intercepting valves and 9 pressure reducing valves to prevent the downstream pressure head from exceeding the set value, achieving a remarkable water saving, approximately equal to 34% of the physical losses, corresponding to 16% of system input volume

    Effect of current corrugations on the stability of the tearing mode

    The generation of zonal magnetic fields in laboratory fusion plasmas is predicted by theoretical and numerical models and was recently observed experimentally. It is shown that the modification of the current density gradient associated with such corrugations can significantly affect the stability of the tearing mode. A simple scaling law is derived that predicts the impact of small stationary current corrugations on the stability parameter Δ\Delta'. The described destabilization mechanism can provide an explanation for the trigger of the Neoclassical Tearing Mode (NTM) in plasmas without significant MHD activity.Comment: Accepted to Physics of Plasma

    Distribution of chirality in the quantum walk: Markov process and entanglement

    The asymptotic behavior of the quantum walk on the line is investigated focusing on the probability distribution of chirality independently of position. The long-time limit of this distribution is shown to exist and to depend on the initial conditions, and it also determines the asymptotic value of the entanglement between the coin and the position. It is shown that for given asymptotic values of both the entanglement and the chirality distribution it is possible to find the corresponding initial conditions within a particular class of spatially extended Gaussian distributions. Moreover it is shown that the entanglement also measures the degree of Markovian randomness of the distribution of chirality.Comment: 5 pages, 3 figures, It was accepted in Physcial Review

    A combined model of pressure variations in Titan's plasma environment

    In order to analyze varying plasma conditions upstream of Titan, we have combined a physical model of Saturn?s plasma disk with a geometrical model of the oscillating current sheet. During modeled oscillation phases where Titan is farthest from the current sheet, the main sources of plasma pressure in the near-Titan space are the magnetic pressure and, for disturbed conditions, the hot plasma pressure. When Titan is at the center of the sheet, the main sources are the dynamic pressure associated with Saturn?s cold, subcorotating plasma and the hot plasma pressure under disturbed conditions. Total pressure at Titan (dynamic plus thermal plus magnetic) typically increases by a factor of up to about 3 as the current sheet center is approached. The predicted incident plasma flow direction deviates from the orbital plane of Titan by ≲ 10◦ . These results suggest a correlation between the location of magnetic pressure maxima and the oscillation phase of the plasma sheet.     Tailoring discrete quantum walk dynamics via extended initial conditions: Towards homogeneous probability distributions

    We study the evolution of initially extended distributions in the coined quantum walk on the line by analyzing the dispersion relation of the process and its associated wave equations. This allows us, in particular, to devise an initially extended condition leading to a uniform probability distribution whose width increases linearly with time, with increasing homogeneity.Comment: 4 pages, 2 figure

    Adjustments, extinction, and remains of selenocysteine incorporation machinery in the nematode lineage

After 12 months, it is available under a Creative Commons License (Attribution-NonCommercial 4.0 International).Selenocysteine (Sec) is encoded by an UGA codon with the help of a SECIS element present in selenoprotein mRNAs. SECIS-binding protein (SBP2/SCBP-2) mediates Sec insertion, but the roles of its domains and the impact of its deficiency on Sec insertion are not fully understood. We used Caenorhabditis elegans to examine SBP2 function since it possesses a single selenoprotein, thioredoxin reductase-1 (TRXR-1). All SBP2 described so far have an RNA-binding domain (RBD) and a Secincorporation domain (SID). Surprisingly, C. elegans SBP2 lacks SID and consists only of an RBD. An sbp2 deletion mutant strain ablated Sec incorporation demonstrating SBP2 essentiality for Sec incorporation. Further in silico analyses of nematode genomes revealed conservation of SBP2 lacking SID and maintenance of Sec incorporation linked to TRXR-1. Remarkably, parasitic plant nematodes lost the ability to incorporate Sec, but retained SecP43, a gene associated with Sec incorporation. Interestingly, both selenophosphate synthetase (SPS) genes are absent in plant parasitic nematodes, while only Cys-containing SPS2 is present in Sec-incorporating nematodes. Our results indicate that C. elegans and the nematode lineage provide key insights into Sec incorporation and the evolution of Sec utilization trait, selenoproteomes, selenoproteins, and Sec residues.     New Histamine-Related Five-Membered N-Heterocycle Derivatives as Carbonic Anhydrase I Activators

    A series of histamine (HST)-related compounds were synthesized and tested for their activating properties on five physiologically relevant human Carbonic Anhydrase (hCA) isoforms (I, II, Va, VII and XIII). The imidazole ring of HST was replaced with different 5-membered heterocycles and the length of the aliphatic chain was varied. For the most interesting compounds some modifications on the terminal amino group were also performed. The most sensitive isoform to activation was hCA I (K(A) values in the low micromolar range), but surprisingly none of the new compounds displayed activity on hCA II. Some derivatives (1, 3a and 22) displayed an interesting selectivity for activating hCA I over hCA II, Va, VII and XIII

    A novel synthetic peptide from a tomato defensin exhibits antibacterial activities against Helicobacter pylori

    Defensins are a class of cysteine-rich proteins, which exert broad spectrum antimicrobial activity. In this work, we used a bioinformatic approach to identify putative defensins in the tomato genome. Fifteen proteins had a mature peptide that includes the well-conserved tetradisulfide array. We selected a representative member of the tomato defensin family; we chemically synthesized its gamma-motif and tested its antimicrobial activity. Here, we demonstrate that the synthetic peptide exhibits potent antibacterial activity against Gram-positive bacteria, such as Staphylococcus aureus A170, Staphylococcus epidermidis, and Listeria monocytogenes, and Gram-negative bacteria, including Salmonella enterica serovar Paratyphi, Escherichia coli, and Helicobacter pylori. In addition, the synthetic peptide shows minimal (<5%) hemolytic activity and absence of cytotoxic effects against THP-1 cells. Finally, SolyC exerts an anti-inflammatory activity in vitro, as it downregulates the level of the proinflammatory cytokines TNF-alpha and IFN-gamma
