    Combined Hazard Analyses to Explore the Impact of Liquid Hydrogen Fuel on the Civil Aviation Industry

    Interest in green technology in aviation is increasing. To address environmental issues, novel fuels such as cryogenic liquid hydrogen (LH2) are being explored, however there are safety concerns. This work combines safety studies to explore LH2 fuel safety for civil aviation. Preliminary hazard analyses (PHAs) (utilizing over 70 standards and guides) have been performed identifying possible LH2 hazards on-board aircraft. A PHA has also been produced, with industry stakeholder involvement, to understand the major concerns for LH2 use at airports. Gaps in fundamental knowledge and LH2 technology have been identified, and two of these explored. Firstly, work has been started to understand the fundamental flammability of hydrogen in altitude conditions. Secondly, FLACS CFD modelling has been used to simulate large-scale LH2 pool releases to examine behavior and predict pool size, downwind flammable regions, and flammable mass clouds formed for different environmental conditions and release scenarios. This has identified significant effects of wind speed on buoyancy and flammable cloud travel which must be taken into account of any hydrogen fuel facility design. This work (part of the EC funded ENABLEH2 project) is some of the first in over a decade to re-examine the safety of hydrogen propulsion in aircraft.     Factor analyses differentiate clinical phenotypes of idiopathic and interferon-alpha-induced depression

    The discovery that prolonged administration of interferon-alpha (a pro-inflammatory cytokine) readily precipitates depressive symptoms has played a key role in development of the inflammation theory of major depressive disorder (MDD). However, it remains unclear whether the clinical phenotype of patients with inflammation-associated depression significantly overlaps with, or can be distinguished from that of patients with ‘idiopathic’ depression. Here we explored the Hamilton depression scale factor structure of 172 patients undergoing interferon-alpha treatment for hepatitis-C at the point of transition to a depressive episode of DSM IV defined major depression severity. The resulting factor structure was first compared with a model derived from 6 previous studies of ‘idiopathic’ MDD (Cole et al., 2004). This confirmatory factor analysis revealed that the factor structure of HAMD scores in our interferon-alpha treated cohort did not plausibly fit that previously described for ‘idiopathic’ MDD. Instead, subsequent exploratory factor analysis revealed a distinct four factor model with a novel primary factor grouping cognitive symptoms of depression and anxiety (HAMD items 1, 2, 9, 10, 11, 15). The second sleep disorder factor (items 4, 5, 6) replicated previous findings in ‘idiopathic’ depression. A third and unique factor grouped somatic symptoms and function (items 7, 12, 13, 14 and item 1). The final factor (also common in idiopathic depression studies), grouped gastrointestinal symptoms and weight loss (items 12 and 16). Severe depression items (3, 8, and 17) were excluded from analysis due to very low variance. At transition, interferon-alpha induced major depressive episodes therefore appears to have more associated anxiety features that covary with depressed mood than classical or ‘idiopathic’ MDD and a low likelihood of severe features such as suicidal ideation. Identification of this clinical phenotype may help identify patients with an inflammatory depression etiology and support the development of more effective and personalized therapies

    A four mirror anastigmat collimator design for optical payload calibration

    We present here a four mirror anastigmatic optical collimator design intended for the calibration of an earth observation satellite instrument. Specifically, the collimator is to be applied to the ground based calibration of the Sentinel-4/UVN instrument. This imaging spectrometer instrument itself is expected to be deployed in 2019 in a geostationary orbit and will make spatially resolved spectroscopic measurements of atmospheric contaminants. The collimator is to be deployed during the ground based calibration only and does not form part of the instrument itself. The purpose of the collimator is to provide collimated light within the two instrument passbands in the UV-VIS (305 – 500 nm) and the NIR (750 – 775 nm). Moreover, that collimated light will be derived from a variety of slit like objects located at the input focal (object) plane of the collimator which is uniformly illuminated by a number of light sources. The collimator must relay these objects with exceptionally high fidelity. To this end, the wavefront error of the collimator should be less than 30 nm rms across the collimator field of view. This field is determined by the largest object which is a large rectangular slit, 4.4° x 0.25°. Other important considerations affecting the optical design are the requirements for input telecentricity and the size (85 mm) and location (2500 mm ‘back focal distance’) of the exit pupil.     Preliminary design framework for the power gearbox in a contra-rotating open rotor

    This study introduces an innovative approach to sizing a differential planetary gearbox for a counter-rotating open rotor application. An updated methodology is proposed for the design of maximum load capacity gears based on the power transmitted, durability and space-envelope requirements of the application. The reported methodology has been validated by comparing the results to published data, demonstrating a maximum difference of 0.6% in geometry. Parametric analyses have also been performed to assess the impact of the design assumptions on gearbox dimensional trends. The proposed methodology enables the assessment of the impact of the preliminary transmission system design on engine performance and general arrangement. The characteristics of the gearset lead to an unequal torque split between output shafts (i.e. the propeller shafts). Given the design assumptions made, the study indicates that valid torque ratios would lie between 1.1 and 1.33. The impact of the torque ratio on the size of the gearbox has been analysed for equal rotational speeds and for different speeds between the output shafts.     Beaked whales respond to simulated and actual navy sonar

They are difficult to sight but can reliably be detected by listening for echolocation clicks produced during deep foraging dives. Listening for these clicks, we documented Blainville's beaked whales, Mesoplodon densirostris, in a naval underwater range where sonars are in regular use near Andros Island, Bahamas. An array of bottom-mounted hydrophones can detect beaked whales when they click anywhere within the range. We used two complementary methods to investigate behavioral responses of beaked whales to sonar: an opportunistic approach that monitored whale responses to multi-day naval exercises involving tactical mid-frequency sonars, and an experimental approach using playbacks of simulated sonar and control sounds to whales tagged with a device that records sound, movement, and orientation. Here we show that in both exposure conditions beaked whales stopped echolocating during deep foraging dives and moved away. During actual sonar exercises, beaked whales were primarily detected near the periphery of the range, on average 16 km away from the sonar transmissions. Once the exercise stopped, beaked whales gradually filled in the center of the range over 2–3 days. A satellite tagged whale moved outside the range during an exercise, returning over 2–3 days post-exercise. The experimental approach used tags to measure acoustic exposure and behavioral reactions of beaked whales to one controlled exposure each of simulated military sonar, killer whale calls, and band-limited noise. The beaked whales reacted to these three sound playbacks at sound pressure levels below 142 dB re 1 µPa by stopping echolocation followed by unusually long and slow ascents from their foraging dives.     Cryogenic Fuel Storage Modelling And Optimisation For Aircraft Applications

    Designing commercial aircraft to use liquid hydrogen (LH2) is one way to substantially reduce their life-cycle CO2 emissions. The merits of hydrogen as an aviation fuel have long been recognized, however, the handling of a cryogenic fuel adds complexity to aircraft and engine systems, operations, maintenance and storage. The and fuel tanks could account for 8-10% of an aircraft’s operating empty weight, as such designing them for the least added weight is of high significance. This paper describes the heat transfer model developed in the EU Horizon 2020 project that is used to predict heat ingress to a cylindrical tank with hemispherical end caps with external foam insulation. It accounts for heat transfer according to the state of the tank contents, the insulation material properties, the environment and the dimensions of the tank. The model also estimates the rate of pressure change according to the state of the fuel and the rate at which fuel is withdrawn from the tank. In addition, a methodology is presented, that allows for tank sizing taking into consideration the requirements of a design flight mission, the maximum pressure developed and the fuel evaporated. Finally, the study demonstrates how to select optimal insulation material and thickness that provides the lightest design for the case where gaseous hydrogen is not vented and when venting during cruise is applied, leading to gravimetric efficiencies as high as 74%

    Structural basis of the anti-ageing effects of polyphenolics: mitigation of oxidative stress

    Ageing, and particularly the onset of age-related diseases, is associated with tissue dysfunction and macromolecular damage, some of which can be attributed to accumulation of oxidative damage. Polyphenolic natural products such as stilbenoids, flavonoids and chalcones have been shown to be effective at ameliorating several age-related phenotypes, including oxidative stress, inflammation, impaired proteostasis and cellular senescence, both in vitro and in vivo. Here we aim to identify the structural basis underlying the pharmacology of polyphenols towards ROS and related biochemical pathways involved in age-related disease. We compile and describe SAR trends across different polyphenol chemotypes including stilbenoids, flavonoids and chalcones, review their different molecular targets and indications, and identify common structural ground between chemotypes and mechanisms of action. In particular, we focus on the structural requirements for the direct scavenging of reactive oxygen/nitrogen species such as radicals as well as coordination of a broader antioxidant response. We further suggest that it is important to consider multiple (rather than single) biological activities when identifying and developing new medicinal chemistry entities with utility in modulating complex biological properties such as cell ageing

    Optimisation of a screening platform for determining IL-6 inflammatory signalling in the senescence-associated secretory phenotype (SASP)

    Cellular senescence has been shown to be sufficient for the development of multiple age-related pathologies. Senescent cells adopt a secretory phenotype (the SASP) which comprises a large number of pro-inflammatory cytokines, chemokines and proteases. The SASP itself is thought to be causative in many pathologies of age-related diseases, and there is growing interest in developing seno-modifying agents that can suppress the SASP. However, in order to identify new agents, it is necessary to conduct moderate to high throughput screening with robust assays for the required outcome. Here, we describe optimisation and validation of a cell-based biosensor HEK cell line for measurement of IL-6 concentrations within the range secreted into conditioned medium by primary senescent fibroblasts, adapted for a 384 well plate format suitable for library screening applications. We further show that the assay can measure changes in IL-6 secretion dependent on cell population age, and that the assay is responsive to mTOR inhibition in the senescent cells, which reduces the SASP, including IL-6. Hence, we propose that this optimised biosensor, which we term HEK-SASP, may prove of value in studies requiring robust, renewable and relatively inexpensive assays for measuring SASP factors
