    Fine tuning and interference study in the determination of heavy metals in fishery products by ICP-MS technique

    Fish is one of the main protein sources for human diet which is usually affected by the pollution of aquatic ecosystem due to bioaccumulation of heavy metals, among others. For this reason, these pollutants are included in the regulation of the European Community Commission which set the maximum levels for certain contaminants in foodstuff. In order to quantify heavy metals, Inductively Coupled Plasma Mass Spectrometry (ICP-MS) is a great analytical technique due to its high accuracy and precision in detecting metals at low concentrations. The aim of this project is to carry out a fine tuning procedure and interference elimination studies during the determination of mercury (Hg), cadmium (Cd) and lead (Pb) in fishery products using ICP-MS. Linearity was studied by reduction of the curve range in order to increase the accuracy of the method. Moreover, selection of the optimum internal standard in order to correct matrix effect during the standardization process was evaluated. Residuals and percentage of internal standard recovery are the quality controls established to determine if the selected standard (Rh-103) was adequate. Finally, results indicate that Cd-111 was the isotope more suitable in order to avoid the spectral interferences

    Background: Adverse drug reactions (ADRs) have a high impact on morbidity and mortality of the population, becoming a public health issue. Studying and publishing about these is referred as pharmacovigilance.Objective: To describe and compare the adverse reactions produced by drugs of nervous system action (CNS-D) and neurological ADRs produced by drugs of systemic action (Sys-D). To further develop the need of reporting adverse reactions. Methods: An observational, cross-sectional, retrospective study performed on a database of neurological consultations which took place at the Neurology department. Patients meeting the inclusion criteria were selected and divided into two groups: Sys-D and CNS-D. Demographic and neurological variables were analyzed. Parametric and non-parametric statistics were used according to distribution. The Naranjo Algorithm (NA) was used to define causality.Results: 71 ADRs were described, from which 63.38% (n=45) were produced by CNS-D, especially antiepileptics by 47% (n=21) and psycholeptics by 44%. Of the total, 36.62% (n=26) were caused by Sys-D, such as antineoplastics (n=9) and antibiotics (n=9), being Cefepime the most frequent. The diagnosis of ADRs caused by a Sys-D was delayed prolonging hospitalization (p 0.05) due to a lower NA score (p 0.003) compared to the CNS-D group.Conclusion: Multiple frequently used drugs of systemic action, such as antineoplastics and antibiotics, generate neurological adverse effects. From our analysis, it was presumed that the suspicion of a neurological ADR caused by these drugs was scarce, thus causing a higher morbidity for the patient

    Cell viability assay in corneal endothelium

The limited availability of corneas makes necessary the developing of preservation techniques that allow a long storage without losing endothelial viability.Objectives: Optimization of a cell viability assay in preserved corneas.Methods: One half of an endothelium from a cornea that was storage in hypothermic conditions and an endothelium of a cryopreserved cornea were stained with LIVE/DEAD imaging kit and Hoechst. The other half of endothelium was the negative control. Corneal endothelia were imaged using a fluorescence microscope.Results: Four sort of cells were visualized on both endothelia: viable cells with high esterase activity, intermediate cells with low esterase activity, non-viable cells without esterase activity, and cells only stained by Hoechst. Conclusions: Triple stain is effectiveto detect different sort of cells in endothelium of preserved corneas, included viable cells, depending on their esterase enzymatic activity and on cell and nuclear membrane damage

    Long runs of homozygosity are associated with Alzheimer’s disease

    Long runs of homozygosity (ROH) are contiguous stretches of homozygous genotypes, which are a footprint of inbreeding and recessive inheritance. The presence of recessive loci is suggested for Alzheimer’s disease (AD); however, their search has been poorly assessed to date. To investigate homozygosity in AD, here we performed a finescale ROH analysis using 10 independent cohorts of European ancestry (11,919 AD cases and 9181 controls.) We detected an increase of homozygosity in AD cases compared to controls [βAVROH (CI 95%) = 0.070 (0.037–0.104); P = 3.91 × 10−5; βFROH (CI95%) = 0.043 (0.009–0.076); P = 0.013]. ROHs increasing the risk of AD (OR > 1) were significantly overrepresented compared to ROHs increasing protection (p < 2.20 × 10−16). A significant ROH association with AD risk was detected upstream the HS3ST1 locus (chr4:11,189,482‒11,305,456), (β (CI 95%) = 1.09 (0.48 ‒ 1.48), p value = 9.03 × 10−4), previously related to AD. Next, to search for recessive candidate variants in ROHs, we constructed a homozygosity map of inbred AD cases extracted from an outbred population and explored ROH regions in wholeexome sequencing data (N = 1449). We detected a candidate marker, rs117458494, mapped in the SPON1 locus, which has been previously associated with amyloid metabolism. Here, we provide a research framework to look for recessive variants in AD using outbred populations. Our results showed that AD cases have enriched homozygosity, suggesting that recessive effects may explain a proportion of AD heritability.Consejería de Salud de la Junta de Andalucía PI-0001/201

    Subjectivity and symptom in Uruguayan interior hospitals: scope of the role of the psychology resident

    El presente trabajo pretende dar visibilidad al complejo y novedoso escenario en el que se inserta el Residente de Psicología en Uruguay, cuando ingresa a trabajar en un Hospital General, tomando las experiencias en cuatro hospitales del interior del país. Las experiencias a transmitir son fruto de nuestra inserción en ASSE (Administración de los Servicios de Salud del Estado) a través del Programa de Practicantes y Residentes de Psicología (PPRP) por el convenio entre ASSE y la Facultad de Psicología de la Universidad de la República (2009). Conjuntamente transitando la Especialización de Psicología en Servicios de Salud (Muniz, 2013) que se enmarca en el Sistema Nacional Integrado de Salud (2007) y la nueva Ley de Salud Mental (2017). Interesa problematizar acerca de los obstáculos y facilitadores para el ejercicio del rol de Residente de Psicología en los Servicios de Salud. Así como también intercambiar en relación al valioso aporte que podemos realizar desde nuestra disciplina en los equipos de salud hospitalarios, contribuyendo a un abordaje interdisciplinario. Se propone tomar como líneas de análisis de las subjetividades y presentaciones sintomáticas actuales, los efectos que produce la proximidad del fallecimiento de un paciente en la institución y quienes trabajan en ella, pero sobre todo en los usuarios y sus familias. ¿Cómo se transitan los procesos de enfermedad, la aceptación de un diagnóstico médico?, ¿Qué lugar se le da a la expresión y al trabajo sobre el sufrimiento, las emociones, la angustia, el dolor psíquico? ¿Cuál es el rol del Residente de Psicología ante estos nuevos escenarios en Uruguay?The present work aims to give visibility to the complex and novel scenario in which the Psychology Resident in Uruguay is inserted, when he enters to work in a General Hospital, taking the experiences in four hospitals of the interior of the country.The experiences to be transmitted are the result of our insertion in ASSE (Administration of State Health Services) through the Program of Practitioners and Residents of Psychology (PPRP) by the agreement between ASSE and the Faculty of Psychology of the University of the Republic (2009). Jointly transiting the Specialization of Psychology in Health Services (Muniz, 2013), that is part of the National Integrated Health System (2007) and the new Mental Health Law (2017). It is interesting to problematize about the obstacles and facilitators for the exercise of the role of Resident of Psychology in Health Services. As well as exchange in relation to the valuable contribution we can make from our discipline in hospital health teams, contributing to an interdisciplinary approach. It is proposed to take as a line of analysis the subjectivities and current symptomatic presentations, the effects produced by the proximity of the death of a patient in the institution and those who work in it, but especially in users and their families. How is the disease processes, the acceptance of a medical diagnosis transited? What place is given to the expression and work on suffering, emotions, anguish, psychic pain?     Development of a disease-specific quality of life questionnaire for adult patients with hereditary angioedema due to C1 inhibitor deficiency (HAE-QoL): Spanish multi-centre research project

    BACKGROUND: There is a need for a disease-specific instrument for assessing health-related quality of life in adults with hereditary angioedema due to C1 inhibitor deficiency, a rare, disabling and life-threatening disease. In this paper we report the protocol for the development and validation of a specific questionnaire, with details on the results of the process of item generation, domain selection, and the expert and patient rating phase. METHODS/DESIGN: Semi-structured interviews were completed by 45 patients with hereditary angioedema and 8 experts from 8 regions in Spain. A qualitative content analysis of the responses was carried out. Issues raised by respondents were grouped into categories. Content analysis identified 240 different responses, which were grouped into 10 conceptual domains. Sixty- four items were generated. A total of 8 experts and 16 patients assessed the items for clarity, relevance to the disease, and correct dimension assignment. The preliminary version of the specific health-related quality of life questionnaire for hereditary angioedema (HAE-QoL v 1.1) contained 44 items grouped into 9 domains. DISCUSSION: To the best of our knowledge, this is the first multi-centre research project that aims to develop a specific health-related quality of life questionnaire for adult patients with hereditary angioedema due to C1 inhibitor deficiency. A preliminary version of the specific HAE-QoL questionnaire was obtained. The qualitative analysis of interviews together with the expert and patient rating phase helped to ensure content validity. A pilot study will be performed to assess the psychometric properties of the questionnaire and to decide on the final version

    Expression of p53 protein isoforms predicts survival in patients with multiple myeloma

    Loss and/or mutation of the TP53 gene are associated with short survival in multiple myeloma, but the p53 landscape goes far beyond. At least 12 p53 protein isoforms have been identified as a result of a combination of alternative splicing, alternative promoters and/or alternative transcription site starts, which are grouped as α, β, γ, from transactivation domain (TA), long, and short isoforms. Nowadays, there are no studies evaluating the expression of p53 isoforms and its clinical relevance in multiple myeloma (MM). We used capillary nanoimmunoassay to quantify the expression of p53 protein isoforms in CD138‐purified samples from 156 patients with newly diagnosed MM who were treated as part of the PETHEMA/GEM2012 clinical trial and investigated their prognostic impact. Quantitative real‐time polymerase chain reaction was used to corroborate the results at RNA levels. Low and high levels of expression of short and TAp53β/γ isoforms, respectively, were associated with adverse prognosis in MM patients. Multivariate Cox models identified high levels of TAp53β/γ (hazard ratio [HR], 4.49; p < .001) and high‐risk cytogenetics (HR, 2.69; p < .001) as independent prognostic factors associated with shorter time to progression. The current cytogenetic‐risk classification was notably improved when expression levels of p53 protein isoforms were incorporated, whereby high‐risk MM expressing high levels of short isoforms had significantly longer survival than high‐risk patients with low levels of these isoforms. This is the first study that demonstrates the prognostic value of p53 isoforms in MM patients, providing new insights on the role of p53 protein dysregulation in MM biology
