    PCR-RFLP Using BseDI Enzyme for Pork Authentication in Sausage and Nugget Products

    A polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) using BseDI restriction enzyme had been applied for identifying the presence of pork in processed meat (beef sausage and chicken nugget) including before and after frying. Pork sample in various levels (1%, 3%, 5%, 10%, and 25 %) was prepared in a mixture with beef and chicken meats and processed for sausage and nugget. The primers CYTb1 and CYTb2 were designed in the mitochondrial cytochrome b (cyt b) gene and PCR successfully amplified fragments of 359 bp. To distinguish existence of porcine species, the amplified PCR products of mitochondrial DNA were cut by BseDI restriction enzyme. The result showed pig mitochondrial DNA was cut into 131 and 228 bp fragments. The PCR-RFLP species identification assay yielded excellent results for identification of porcine species. It is a potentially reliable technique for pork detection in animal food processed products for Halal authentication

    Studi Karakteristik Kelistrikan Komposit Carbon Nanotube-polyvynilidene-flouride (Cnt-pvdf)

    Development of nanomaterial technology strongly supports advancements in composite technology. Composite that use nanomaterial produce better electrical properties than common composite. Carbon nanotubes (CNT) is one of nanomaterial with high conductivity, so it\u27s potentially used in composite for various aplication. This research combines Carbon Nanotubes (CNT) with Polyvynilidene Flouride (PVDF) for producing CNT-PVDF composite with high conductivity. CNT-PVDF composites were fabricated by solution processing method. PVDF was dissolved into distillated aceton then CNT was dissolved in acetone and added Triton X-100 4 ml. PVDF solution and the solution of CNT were stirred for 1 hour using a magnetic stirrer, then both were mixed and stirred at temperature of 40 ° C until the acetone evaporates. Composites were molded using a hot-pressing at a pressure of 1 MPa and a temperature of 200oC for 10 minutes. CNT-PVDF composite were characterized by a conductivity test to generate conductivity values​​. The results of the analysis of the test showed that the addition of CNT conductivity on CNT-PVDF composite increased conductivity values​​. In the mass percentage of CNT 2%, 4%, 6%, and 8% conductivity increased slowly. Improved high conductivity of CNTs obtained in the mass percentage of 10%

    Pengaruh Fee Audit, Audit Tenure, Dan Rotasi Audit Terhadap Kualitas Audit

    The purpose of this study is to examine the influence of audit fee, audit tenure and audit rotation of audit quality. Audit quality uses proxy size KAP which is measured by dummy variable and using the big four and non big four group of auditors, while tenure is measured by counting the year in which the same auditors has made an engangement with the auditee, audit rotation is measured by dummy variable.The population in this study consists of listed go public firm manufactur in Indonesia Stock Exchange in year 2008-2012. Sampling method used is purposive sampling. In this research, the total of the sample is 645, after data observation then, there are 415 samples that included outlier should be excluded from samples of observation. so, the final amounts of the sample are 230 firms. Logistic regression used to be analysis technique. The empirical result of this study show that audit fee, audit tenure, and audit rotation have significant influenced of audit quality. Audit fee variable positively affects the audit quality, audit tenure negatively affects the audit quality and audit rotation postively affects the audit quality

    Green building merupakan suatu konsep bangunan ramah lingkungan yang sudah menjadi perhatian khusus di berbagai negara dan mulai diterapkan di Indonesia. Konsep Green Building merupakan salah satu upaya penghematan energi yang dapat diterapkan pada suatu gedung. Penulisan Tugas Akhir ini dilakukan untuk mengukur rating/sertifikasi sebagai tolak ukur sudah sejauh mana tingkat green building gedung-gedung di ITS, dengan cara melakukan pengukuran langsung, yang dilakukan oleh peneliti berdasarkan kriteria standar nasional (Greenship-GBCI). Namun penelitian hanya dilakukan pada beberapa kriteria green building. Hasil dari penelitian ini berupa pengukuran kriteria green building sesuai standar Greenship dan penilaian rating sertifikasi green building pada Gedung Teknik Sipil ITS yang kemudian akan menjadi tolak ukur tersendiri untuk kedepan nya. Dalam hasil penelitian terlihat bahwa Gedung Teknik Sipil ITS sudah memenuhi beberapa standar kriteria green building yang tercantum pada Greenship, namun masih ada juga kriteria-kriteria green building yang belum diterapkan ataupun terpenuhi. Konsep Green Building inipun sebaiknya dilakukan sejak awal perencanaan, namun hal ini tidak menutup kemungkinan untuk meningkatkan kualitas green building pada Gedung Teknik Sipil ITS ke depanny

    Optimization of Multiple Response Using Taguchi-WPCA in ST 60 Tool Steel Turning Process with Minimum Quantity Cooling Lubrication (MQCL) Method

    A research was conducted for the optimization of the turning process st 60 tool steel with multiple performance characteristics based on the orthogonal array with Taguchi-WPCA method. Minimum Quantity Cooling Lubrication (MQCL) metode was applied as a coolant. The experimental studies were conducted under varying the cutting speed, feeding, depth of cut and type of coolant. The optimized multiple performance characteristics were surface roughness, and material removal rate. An orthogonal array, signal-to-noise ratio, grey relational analysis, weighted pricipal component analysis and analysis of variance were employed to study the multiple performance characteristics. Experimental results show that cutting speed gives the highest contribution for minimize of surface roughness and maximize of material removal rate, followed by feeding speed, type of coolant and depth of cut. The minimum of surface roughness and maximize of material removal rat could be obtained by using the values of cutting speed, feeding speed, depth of cut and type of coolant of 172.95 m/minute, 0.053 mm/rev, 0.25 mm, and vegetable oil as a coolant respectively
