    Deregulated expression of TANK in glioblastomas triggers pro-tumorigenic ERK1/2 and AKT signaling pathways

    Signal transmission by the noncanonical IkappaB kinases (IKKs), TANK-binding kinase 1 (TBK1) and IKKɛ, requires interaction with adapter proteins such as TRAF associated NF-κB activator (TANK). Although increased expression or dysregulation of both kinases has been described for a variety of human cancers, this study shows that deregulated expression of the TANK protein is frequently occurring in glioblastomas (GBMs). The functional relevance of TANK was analyzed in a panel of GBM-derived cell lines and revealed that knockdown of TANK arrests cells in the S-phase and prohibits tumor cell migration. Deregulated TANK expression affects several signaling pathways controlling cell proliferation and the inflammatory response. Interference with stoichiometrically assembled signaling complexes by overexpression or silencing of TANK prevented constitutive interferon-regulatory factor 3 (IRF3) phosphorylation. Knockdown of TANK frequently prevents constitutive activation of extracellular signal-regulated kinases 1 and 2 (ERK1/2). TANK-mediated ERK1/2 activation is independent from the canonical MAP kinase or ERK kinase (MEK) 1/2-mediated pathway and utilizes an alternative pathway that uses a TBK1/IKKɛ/Akt signaling axis, thus identifying a novel pathway suitable to block constitutive ERK1/2 activity.Peer reviewe

    Estudos têm demonstrado que o exercício físico regular melhora as condições do diabetes, facilitando a captação periférica da glicose e o metabolismo de glicogênio, proteínas, etc. Por outro lado, pouco se conhece sobre os efeitos do exercício intenso em diabéticos, principalmente com relação ao sistema imune desses organismos. O presente estudo teve como objetivo verificar os efeitos de um treinamento físico de alta intensidade sobre a contagem total e diferencial de leucócitos em ratos diabéticos. Ratos machos jovens Wistar foram distribuídos em quatro grupos: controle sedentário (CS), controle treinado (CT), diabético sedentário (DS) e diabético treinado (DT). O diabetes foi induzido por aloxana (35mg/kg de peso corporal). Durante seis semanas os animais dos grupos CT e DT realizaram um protocolo de treinamento físico, que consistiu na realização de quatro séries de 10 saltos (intercaladas por um minuto de intervalo) em piscina, com o nível da água correspondendo a 150% do comprimento corporal e sobrecarga equivalente a 50% da massa corporal dos animais. Ao final do período experimental, amostras de sangue foram coletadas para a contagem total e diferencial dos leucócitos. Os resultados foram avaliados estatisticamente por ANOVA com um nível de significância de 5%. A glicemia foi aumentada entre os diabéticos e a insulinemia diminuída. Não foram observadas diferenças significativas na contagem diferencial dos linfócitos, neutrófilos, eosinófilos e contagem total de leucócitos entre os grupos estudados. Houve aumento dos monócitos entre os treinados (CS = 10,0 ± 4,5, CT* = 25,4 ± 7,9, DS = 19,75 ± 7,4, DT* = 25,8 ± 4,4%). O peso relativo do timo foi reduzido pelo treinamento e pelo diabetes (CS = 125,0 ± 37,7, CT* = 74,6 ± 8,2, DS* = 47,5 ± 12,2, DT* = 40,1 ± 16,9mg/100g). Esses resultados permitem concluir que o treinamento físico de alta intensidade não alterou o estado geral do diabetes, mas aumentou os monócitos, o que pode representar um efeito positivo sobre a resposta imunológica desses animais.<br>Several studies have demonstrated that regular physical activity improves diabetes conditions, favoring the peripheral glucose uptake, glycogen and protein metabolism. However, the effects of high intensity physical training on the immune system of diabetic organisms are not totally clear. The aim of this study was to verify the effects of high intensity physical training on the total and differential leukocyte count of diabetic rats. Male young Wistar rats were distributed into four groups: sedentary control (SC), trained control (TC), sedentary diabetic (SD) and trained diabetic (TD). Diabetes was induced by alloxan (30 mg/kg body weight i.v.). During six weeks the animals of TC and TD groups followed a high intensity physical training protocol which consisted of four sets of 10 jumps/day (interrupted by one minute of rest interval) in a swimming pool, with the water level corresponding to 150% of the body length and overload equivalent to 50% of the body weight. At the end of the experimental period, blood samples were collected for total and differential leukocyte count. The results were analyzed by ANOVA at a significance level of 5%. Serum glucose was increased in diabetic groups while the insulin level was reduced in these groups. There were no significant differences in lymphocytes, neutrophils, and eosinophils, and in total leukocyte count when the groups were compared. Monocytes count was higher in both trained groups (SC = 10.0 ± 4.5, TC* = 25.4 ± 7.9, SD = 19.75 ± 7.4, TD* = 25.8 ± 4.4%). The relative weight of the thymus was reduced by diabetes and training (SC = 125.0 ± 37.7, TC* =74.6 ± 8.2, SD* = 47.5 ± 12.2, TD* = 40.1 ± 16.9). In conclusion, the high intensity physical training protocol did not change the general diabetes conditions, but improved relative monocytes. These results can represent a positive effect on the immune response

    Autonomia funcional de idosas praticantes de Pilates Functional autonomy of elderly women practicing Pilates

    O objetivo foi avaliar o efeito do método Pilates na autonomia funcional de idosas. Foram selecionadas 52 voluntárias, submetidas à avaliação geral e ao protocolo de avaliação da autonomia funcional do Grupo Latino-Americano de Desenvolvimento para Maturidade (GDLAM), que consiste em cinco testes (caminhada de 10 m, levantar-se da posição sentada, levantar-se da cadeira e locomover-se pela casa, levantar-se da posição de decúbito ventral e vestire tirar uma camiseta); em seguida, foram divididas em dois grupos: grupo Pilates (GP, n=27; idade 66,9±5,3 anos) e grupo controle (GC, n=25; idade 65,2±3,9 anos). O GP foi submetido a uma série de dez exercícios de Pilates, por oito semanas, duas vezes por semana. Os dois grupos foram reavaliados após esse período. O nível de significância considerado foi de p?0,05. O GP obteve resultados significativamente melhores em todos os testes e no índice geral do GDLAM (p=0,035) após a intervenção. O GC obteve escore significativamente melhor (p=0,042) apenas no teste de caminhada de 10 m, tendo mantido sua classificação inicial de funcionalidade regular. Comparando-se os escores dos grupos após a intervenção, encontraram-se diferenças significativas em favor do GP, inclusive no índice GDLAM (p<0,05), tendo este passado da classificação de regular a bom. A prática do método Pilates gerou pois melhora significativa do desempenho funcional das idosas estudadas.<br>The purpose of the study was to assess the effect of the Pilates method on elderly women's functional autonomy. Fifty-two volunteers were submitted to the evaluation protocol of the Latin-American Group for Maturity Development (GDLAM), which consists of five tests: a 10-meter walk, get up from the sitting and lying down positions, get up and walk around at home, and to put on and take off a t-shirt. They were then divided into Pilates group (PG, n=27, mean age 66.9±5.3 years) and control group (CG, n=25, mean age 65.2±3.9 years old). PG underwent a series of 10 Pilates exercises twice-weekly for eight weeks, both groups being reassessed thereafter. The level of significance was set at p?0.05. Results showed PG's better scores in all tests and at the GDLAM index (p=0.035) after the intervention; control group showed a better score ((p=0.042) only at the 10-meter walk test, thus keeping the same initial classification of "regular" functionality.. Comparison between the groups after the intervention showed significant advantages for PG, including at the GDLAM index (p<0.05), whose classification rose from "regular" to "good". It may thus be said that practice of the Pilates method significantly enhanced functional performance of the elderly women studied
