    Development of supramolecular peptide-based magnetolipogels: towards on-demand drug delivery

    Solid and aqueous magnetoliposomes were encapsulated in supramolecular naproxen N-capped dehydrodipeptide-based hydrogels. Fluorescence spectroscopy was used to assess the dynamics of magnetoliposomes during gelation through co-encapsulation of Nile Red and curcumin.     Brazilian Version of the Foot Health Status Questionnaire (FHSQ-Br): Cross-Cultural Adaptation and Evaluation of Measurement Properties

    OBJECTIVE: To conduct a cross-cultural adaptation of the Foot Health Status Questionnaire into Brazilian-Portuguese and to assess its measurement properties. INTRODUCTION: This instrument is an outcome measure with 10 domains with scores ranging from 0-100, worst to best, respectively. The translated instrument will improve the examinations and foot care of rheumatoid arthritis patients. METHODS: The questions were translated, back-translated, evaluated by a multidisciplinary committee and pre-tested (n = 40 rheumatoid arthritis subjects). The new version was submitted to a field test (n = 65) to evaluate measurement properties such as test-retest reliability, internal consistency and construct validity. The Health Assessment Questionnaire, Numeric Rating Scale for foot pain and Sharp/van der Heijde scores for foot X-rays were used to test the construct validity. RESULTS: The cross-cultural adaptation was completed with minor wording adaptations from the original instrument. The evaluation of measurement properties showed high reliability with low variation coefficients between interviews. The a-Cronbach coefficients varied from 0.468 to 0.855, while correlation to the Health Assessment Questionnaire and Numeric Rating Scale was statistically significant for five out of eight domains. DISCUSSION: Intra- and inter-observer correlations showed high reliability. Internal consistency coefficients were high for all domains, revealing higher values for less subjective domains. As for construct validity, each domain revealed correlations with a specific group of parameters according to what the domains intended to measure. CONCLUSION: The FHSQ was cross-culturally adapted, generating a reliable, consistent, and valid instrument that is useful for evaluating foot health in patients with rheumatoid arthritis

    New magnetogels based on manganese ferrite nanoparticles and self-assembled peptide hydrogels as drug nanocarriers

    In this work, superparamagnetic manganese ferrite (MnFe2O4) nanoparticles were successfully incorporated in self-assembled peptide-derived hydrogels. The new magnetogels were tested as nanocarriers for two fluorescent drugs, curcumin (an anticancer and neuroprotective drug) and a new antitumor thienopyridine derivative. Fluorescence-based techniques (fluorescence emission, FRET and fluorescence anisotropy) were used to assess incorporation of these drugs in the magnetogels and their transport towards models of biological membranes.     Nano-scale hydrogen-bond network improves the durability of greener cements

    More than ever before, the world's increasing need for new infrastructure demands the construction of efficient, sustainable and durable buildings, requiring minimal climate-changing gas-generation in their production. Maintenance-free “greener” building materials made from blended cements have advantages over ordinary Portland cements, as they are cheaper, generate less carbon dioxide and are more durable. The key for the improved performance of blends (which substitute fine amorphous silicates for cement) is related to their resistance to water penetration. The mechanism of this water resistance is of great environmental and economical impact but is not yet understood due to the complexity of the cement's hydration reactions. Using neutron spectroscopy, we studied a blend where cement was replaced by ash from sugar cane residuals originating from agricultural waste. Our findings demonstrate that the development of a distinctive hydrogen bond network at the nano-scale is the key to the performance of these greener materials

    Liposomal formulations loaded with a eugenol derivative for application as insecticides: encapsulation studies and In silico identification of protein targets

    Supplementary Materials can be downloaded at: https://www.mdpi.com/article/10.3390/nano12203583/s1,A recently synthesized new eugenol derivative, ethyl 4-(2-methoxy-4-(oxiran-2-ylmethyl)phenoxy)butanoate, with a high insecticidal activity against Sf9 (Spodoptera frugiperda) insect cells, was encapsulated in the liposomal formulations of egg-phosphatidylcholine/cholesterol (Egg-PC:Ch) 70:30 and 100% dioleoylphosphatidylglycerol (DOPG), aiming at the future application as insecticides. Compound-loaded DOPG liposomes have sizes of 274 ± 12 nm, while Egg-PC:Ch liposomes exhibit smaller hydrodynamic diameters (69.5 ± 7 nm), high encapsulation efficiency (88.8% ± 2.7%), higher stability, and a more efficient compound release, thus, they were chosen for assays in Sf9 insect cells. The compound elicited a loss of cell viability up to 80% after 72 h of incubation. Relevantly, nanoencapsulation maintained the toxicity of the compound toward insect cells while lowering the toxicity toward human cells, thus showing the selectivity of the system. Structure-based inverted virtual screening was used to predict the most likely targets and molecular dynamics simulations and free energy calculations were used to demonstrate that this molecule can form a stable complex with insect odorant binding proteins and/or acetylcholinesterase.     Management of Psoriasis by Family Physicians: Referral Algorithm and Shared Management with Dermatology

    Introduction: The implementation of models capable of improving referral quality, limiting the growth of waiting lists in hospitals, and ensuring the best possible treatment and follow-up of the psoriatic patient is of the utmost importance. Material and methods: A panel of Family Physicians and Dermatologists discussed and created a simple and effective algorithm of referral for patients with psoriasis. Results: The proposed algorithm starts when the Family Physician suspects of psoriasis. In case of diagnostic doubt, the patient should be referred to Dermatology. In case of a confirmed diagnosis, the Family Physician should assess the patient's severity and responder profile, evaluate comorbidities and assess the presence of psoriatic arthritis. If psoriasis is mild, topical treatments should be initiated, and if there is no clinical improvement or worsening of the disease, the patient should be referred to Dermatology. If psoriasis is moderate to severe, is located in high impact locations, or in pediatric age, the patient should be referred to Dermatology. In order to enable shared management in terms of follow-up and treatment of these patients, it is critical that the Family Physician has the necessary knowledge regarding the systemic treatments used in psoriasis and their side effects. Discussion and conclusion: Only a shared management of the psoriatic patient can allow for the best treatment and follow-up of these patients, a more rational use of available medical resources, thus giving the patient the best possible quality of life.Introdução: A implementação de modelos capazes de melhorar a referenciação, por forma a garantir a qualidade e precocidade do diagnóstico, o melhor tratamento e seguimento do doente psoriático é fundamental.Material e Métodos: Um painel de médicos de Medicina Geral e Familiar e Dermatologia discutiu e criou um algoritmo de referenciação simples, eficaz e célere de doentes com psoríase.Resultados: O algoritmo proposto inicia-se quando o clínico de Medicina Geral e Familiar suspeita de psoríase. Caso haja dúvidas no diagnóstico, o doente deve ser referenciado para a dermatologia. No caso de um diagnóstico confirmado, compete ao clínico de Medicina Geral e Familiar avaliar a gravidade e perfil de respondedor do doente, rastrear comorbilidades e a possibilidade de artrite psoriática. Se a psoríase for ligeira, deverão ser iniciados tratamentos tópicos, sendo que, se não houver melhoria clínica ou ocorrer agravamento da doença, o doente deverá ser referenciado para a dermatologia. Se a psoríase for considerada moderada a grave, em localizações de elevado impacto, ou em idade pediátrica, o doente deverá ser referenciado para a dermatologia. Para que o seguimento e tratamento destes doentes seja partilhado, é fundamental que o médico de Medicina Geral e Familiar tenha o conhecimento necessário sobre os tratamentos sistémicos que o doente está a fazer e os seus efeitos adversos.Discussão e Conclusão: Apenas uma gestão partilhada do doente psoriático poderá tornar possível o melhor tratamento e seguimento destes doentes, a utilização mais racional dos recursos médicos disponíveis, proporcionando ao doente a melhor qualidade devida possível.info:eu-repo/semantics/publishedVersio

    Magnetoliposomes incorporated in peptide-based hydrogels: towards development of magnetolipogels

    A major problem with magnetogels is the encapsulation of hydrophobic drugs. Magnetoliposomes not only provide these domains but also improve drug stability and avert the aggregation of the magnetic nanoparticles. In this work, two magnetoliposome architectures, solid and aqueous, were combined with supramolecular peptide-based hydrogels, which are of biomedical interest owing to their biocompatibility, easy tunability, and wide array of applications. This proof-of-concept was carried out through combination of magnetoliposomes (loaded with the model drug curcumin and the lipid probe Nile Red) with the hydrogels prior to pH triggered gelation, and fluorescence spectroscopy was used to assess the dynamics of the encapsulated molecules. These systems allow for the encapsulation of a wider array of drugs. Further, the local environment of the encapsulated molecules after gelation is unaffected by the used magnetoliposome architecture.     Synthesis, computational and nanoencapsulation studies on eugenol-derived insecticides

    A new set of alkoxy alcohols were synthesised by reaction of eugenol oxirane with aliphatic and aromatic alcohols. These eugenol derivatives were evaluated against their effect upon the viability of the insect cell line Sf9 (Spodoptera frugiperda). The most promising compounds, 4-(3-(tert-butoxy)-2-hydroxypropyl)-2-methoxyphenol and 4-(2-((4-fluorobenzyl)oxy)-3-hydroxypropyl)-2-methoxyphenol were submitted to in silico assays to predict possible targets. Throught an Inverted Virtual Screening approach, 23 common pesticide targets were screened and the top 2 targets predicted were further analyzed through molecular dynamics simulations and free energy calculations. In addition, these eugenol derivatives were subjected to encapsulation and release assays using liposome-based nanosystems of egg phosphatidylcholine/cholesterol (7:3), with encapsulation efficiencies higher than 90% and release profiles well described by both Korsmeyer-Peppas and Weibull models.This research was funded by the project PTDC/ASP-AGR/30154/2017 (POCI-01-0145-FEDER-030154) of the COMPETE 2020 program, co-financed by the FEDER and the European Union.
