    Effect of type of food and culture density on growth and lipid composition of Seriola dumerili juveniles

    The aim of this study was to determine the effect of three different types of food and two stocking densities on growth, survival and lipid composition of muscle and liver of Seriola dumerili juveniles. Juvenile of S. dumerili were randomly distributed in groups of 15 fish/tank (low stocking density, LD) and groups of 25 fish/tank (high stocking density, HD) and fed with three different types of food: commercial pellets for Sparidae (SP), commercial pellets for S. dumerili (SE) and frozen Mackerel (MA). Samples of muscle and liver were taken for lipid analysis at the end of trial (86 days). Fish fed with SP showed growth significantly lower than the groups fed with SE and MA for both stocking densities. Higher survival rates was observed in fish fed diet SE and MA held at LD. Lipid composition analyses revealed important differences between fish fed with different diets but no difference between groups with different stocking density were detected. Total lipid content in muscle and liver from fish fed with SE was very much higher than in fish fed with SP and MA, which was reflected in higher tryglicerides and lower cholesterol contents. The fatty acid composition of tissues clearly reflected the composition of the diet supplied. The groups fed with MA presented the highest proportion of docosahexaenoic acid (DHA), arachidonic acid (ARA) and the lower amounts of eicosapentaenoic acid (EPA). In contrast, fish fed with SE presented the lower amounts of ARA and DHA and higher content in 18:2n-6 specifically in the liver. In consequence, fish fed with MA presented a higher DHA/EPA ratio and a lower EPA/AA ratio in both tissues while the opposite was observed in group SE. These preliminary results indicate that SE diet allows get better performance than SP diet obtaining growth and survival rates comparable to fish fed with MA, although with a higher lipid deposition in tissues

    AKT1 and AKT2 isoforms play distinct roles during breast cancer progression through the regulation of specific downstream proteins

    The purpose of this study was to elucidate the mechanisms associated with the specific effects of AKT1 and AKT2 isoforms in breast cancer progression. We modulated the abundance of specific AKT isoforms in IBH-6 and T47D human breast cancer cell lines and showed that AKT1 promoted cell proliferation, through S6 and cyclin D1 upregulation, but it inhibited cell migration and invasion through β1-integrin and focal adhesion kinase (FAK) downregulation. In contrast, AKT2 promoted cell migration and invasion through F-actin and vimentin induction. Thus, while overexpression of AKT1 promoted local tumor growth, downregulation of AKT1 or overexpression of AKT2 promoted peritumoral invasion and lung metastasis. Furthermore, we evaluated The Cancer Genome Atlas (TCGA) dataset for invasive breast carcinomas and found that increased AKT2 but not AKT1 mRNA levels correlated with a worse clinical outcome. We conclude that AKT isoforms play specific roles in different steps of breast cancer progression, with AKT1 involved in the local tumor growth and AKT2 involved in the distant tumor dissemination, having AKT2 a poorer prognostic value and consequently being a worthwhile target for therapy.Facultad de Ciencias Naturales y MuseoCentro de Investigaciones Inmunológicas Básicas y Aplicada

    Emotional intelligence impairments in women with fibromyalgia: Associations with widespread pain.

    This study aimed at testing the differences in emotional intelligence ability between women with fibromyalgia (cases) and their age-matched counterparts not with fibromyalgia from the general population (controls) and analysing the association between emotional intelligence ability and widespread pain in women with fibromyalgia. A total of 133 cases and 77 controls participated in this cross-sectional study. Controls performed better than cases on emotion understanding. Higher emotion perception and management were significantly associated with lower widespread pain. Therefore, women with fibromyalgia may experience difficulties in understanding emotional information. In fibromyalgia, higher emotion perception and management abilities are independently related to lower widespread pain.pre-print424 K

    Phosphoinositide 3-kinase beta controls replication factor C assembly and function.

    Genomic integrity is preserved by the action of protein complexes that control DNA homeostasis. These include the sliding clamps, trimeric protein rings that are arranged around DNA by clamp loaders. Replication factor C (RFC) is the clamp loader for proliferating cell nuclear antigen, which acts on DNA replication. Other processes that require mobile contact of proteins with DNA use alternative RFC complexes that exchange RFC1 for CTF18 or RAD17. Phosphoinositide 3-kinases (PI3K) are lipid kinases that generate 3-poly-phosphorylated-phosphoinositides at the plasma membrane following receptor stimulation. The two ubiquitous isoforms, PI3Kalpha and PI3Kbeta, have been extensively studied due to their involvement in cancer and nuclear PI3Kbeta has been found to regulate DNA replication and repair, processes controlled by molecular clamps. We studied here whether PI3Kbeta directly controls the process of molecular clamps loading. We show that PI3Kbeta associated with RFC1 and RFC1-like subunits. Only when in complex with PI3Kbeta, RFC1 bound to Ran GTPase and localized to the nucleus, suggesting that PI3Kbeta regulates RFC1 nuclear import. PI3Kbeta controlled not only RFC1- and RFC-RAD17 complexes, but also RFC-CTF18, in turn affecting CTF18-mediated chromatid cohesion. PI3Kbeta thus has a general function in genomic stability by controlling the localization and function of RFC complexes

    Positive-negative tunable liquid crystal lenses based on a microstructured transmission line

    In this work, a novel technique to create positive-negative tunable liquid crystal lenses is proposed and experimentally demonstrated. This structure is based on two main elements, a transmission line acting as a voltage divider and concentric electrodes that distribute the voltage homogeneously across the active area. This proposal avoids all disadvantages of previous techniques, involving much simpler fabrication process (a single lithographic step) and voltage control (one or two sources). In addition, low voltage signals are required. Lenses with switchable positive and negative focal lengths and a simple, low voltage control are demonstrated.
