pendiente autorización de la revistaThe Environment Department of the Government of Andalusia (Spain) has recently assumed the responsibility of integrating extensive livestock grazing as a tool for fire prevention. The present study aims to analyse the sustainability of 60 farms (mainly small ruminant farms) belonging to the Andalusian Network of Grazed Fuel breaks. Farms are located mainly in Natural Areas of Eastern Andalusia. 9291 goats, 24491 ewes and 170 cows grazed in 260 km of fuel breaks. 27 farms have only sheep (mainly meat purpose), 18 only goats (mainly dairy purpose), 10 goat and sheep, 3 small ruminants and suckling cows and 2 small ruminants, suckling cows and Iberian pigs; only 4 goat farms produce cheese. Meat and milk production has a marked seasonal variation. The feed supplied in the manger is relatively high, especially in goat herds. Hard work, small farm profitability and lack of assurance of farm continuity are the most important aspects that negatively affect the sustainability of these farms. For improving sustainability, farmers are to be able to exploit strengths, absorb shocks, adjust following a disturbance and transform their farm to take advantage of new opportunities.

Wnt/β-Catenin Signaling Contributes to Paclitaxel Resistance in Bladder Cancer Cells with Cancer Stem Cell-Like Properties

The Wnt/β-catenin pathway plays an important role in tumor progression and chemother apy resistance and seems to be essential for the maintenance of cancer stem cells (CSC) in several tumor types. However, the interplay of these factors has not been fully addressed in bladder cancer. Here, our goal was to analyze the role of the Wnt/β-catenin pathway in paclitaxel resistance and to study the therapeutic efficacy of its inhibition in bladder cancer cells, as well as to determine its influence in the maintenance of the CSC-like phenotype in bladder cancer. Our results show that paclitaxel-resistant HT1197 cells have hyperactivation of the Wnt/β-catenin pathway and increased CSC-like properties compared with paclitaxel-sensitive 5637 cells. Paclitaxel sensitivity diminishes in 5637 cells after β-catenin overexpression or when they are grown as tumorspheres, enriched for the CSC-like phenotype. Additionally, downregulation of β-catenin or inhibition with XAV939 sensitizes HT1197 cells to paclitaxel. Moreover, a subset of muscle-invasive bladder carcinomas shows aberrant expression of β-catenin that associates with positive expression of the CSC marker ALDH1A1. In conclusion, we demonstrate that Wnt/β-catenin signaling contributes to paclitaxel resistance in bladder cancer cells with CSC-like properties.Instituto de Salud Carlos III FIS-PI17/1240Instituto de Salud Carlos III FIS-PI20/1641Ministry of Economy, Industry and Competitiveness SAF2017-87358-C2-1-R and -2-RMinisterio de Ciencia e Innovación PID2020-118774RB-C21 and -C22Consejería de Salud y Familias PI-0213-2020Consejería de Salud y Familias OH-0017-201

Obatoclax and Paclitaxel Synergistically Induce Apoptosis and Overcome Paclitaxel Resistance in Urothelial Cancer Cells

Paclitaxel is a treatment option for advanced or metastatic bladder cancer after the failure of first-line cisplatin and gemcitabine, although resistance limits its clinical benefits. Mcl-1 is an anti-apoptotic protein that promotes resistance to paclitaxel in different tumors. Obatoclax, a BH3 mimetic of the Bcl-2 family of proteins, antagonizes Mcl-1 and hence may reverse paclitaxel resistance in Mcl-1-overexpressing tumors. In this study, paclitaxel-sensitive 5637 and -resistant HT1197 bladder cancer cells were treated with paclitaxel, obatoclax, or combinations of both. Apoptosis, cell cycle, and autophagy were measured by Western blot, flow cytometry, and fluorescence microscopy. Moreover, Mcl-1 expression was analyzed by immunohistochemistry in bladder carcinoma tissues. Our results confirmed that paclitaxel alone induced Mcl-1 downregulation and apoptosis in 5637, but not in HT1197 cells; however, combinations of obatoclax and paclitaxel sensitized HT1197 cells to the treatment. In obatoclax-treated 5637 and obatoclax + paclitaxel-treated HT1197 cells, the blockade of the autophagic flux correlated with apoptosis and was associated with caspase-dependent cleavage of beclin-1. Obatoclax alone delayed the cell cycle in 5637, but not in HT1197 cells, whereas combinations of both retarded the cell cycle and reduced mitotic slippage. In conclusion, obatoclax sensitizes HT1197 cells to paclitaxel-induced apoptosis through the blockade of the autophagic flux and effects on the cell cycle. Furthermore, Mcl-1 is overexpressed in many invasive bladder carcinomas, and it is related to tumor progression, so Mcl-1 expression may be of predictive value in bladder cancer.España, Sistema Público Andaluz Biobanco y ISCIII-Red de Biobancos PT17/0015/004

Phase angle and COVID-19: a systematic review with meta-analysis

    Get PDF
Phase angle (PhA) has been identified as a poor prognostic factor in patients with COVID-19. This study aimed to achieve a systematic review, where we discussed the potential role of PhA value as a prognostic marker of adverse clinical outcomes such as mortality and complication in hospitalized with SARS-CoV2 infection and established the strength of recommendations for use. A systematic literature review with meta-analysis was done in the main electronic databases from 2020 to January 2023. The selected articles had to investigate adverse consequences of the COVID-19 population and raw bioimpedance parameters such as PhA and published in peer-reviewed journals. GRADE tools regarded the quality of the methodology. The review protocol was registered in PROSPERO. Only eight studies, 483 studies, were eligible for the analysis. In general, differences in PhA were seen between the comparative study groups. Patients with a low PhA experienced poor outcomes. A low PhA was associated with a significantly increased mortality risk [RR: 2.44; 95% CI (1.20–4.99), p = 0.01; I2 = 79% (p = 0.0008)] and higher complications risk [OR: 3.47, 95% CI (1.16 – 10.37), p = 0.03; I2 = 82% (p = 0.004)] in COVID-19 patients. Our analysis showed four evidence-based recommendations on the prognostic value of PhA with two strong recommendations, one of moderate and another of low-moderate quality, for predicting mortality and complications, respectively. We recommend using PhA as a prognostic marker for mortality and complications in this population. Although the results are promising, future studies must identify the PhA cut-off to guide therapeutic decisions more precisely.Funding for open access publishing: Universidad Málaga/CBUA. Rice and bean AHL-mimic quorum-sensing signals specifically interfere with the capacity to form biofilms by plant-associated bacteria

    Get PDF
Many bacteria regulate their gene expression in response to changes in their population density in a process called quorum sensing (QS), which involves communication between cells mediated by small diffusible signal molecules termed autoinducers. n-acyl-homoserine-lactones (AHLs) are the most common autoinducers in proteobacteria. QS-regulated genes are involved in complex interactions between bacteria of the same or different species and even with some eukaryotic organisms. Eukaryotes, including plants, can interfere with bacterial QS systems by synthesizing molecules that interfere with bacterial QS systems. In this work, the presence of AHL-mimic QS molecules in diverse Oryza sativa (rice) and Phaseolus vulgaris (bean) plant-samples were detected employing three biosensor strains. A more intensive analysis using biosensors carrying the lactonase enzyme showed that bean and rice seed-extract contain molecules that lack the typical lactone ring of AHLs. Interestingly, these molecules specifically alter the QS-regulated biofilm formation of two plant-associated bacteria, Sinorhizobium fredii SMH12 and Pantoea ananatis AMG501, suggesting that plants are able to enhance or to inhibit the bacterial QS systems depending on the bacterial strain. Further studies would contribute to a better understanding of plant–bacteria relationships at the molecular level

[EN]This paper presents the results of the skill evaluation of the members of two ensembles of regional climate simulations in reproducing precipitation and maximum and minimum temperatures using three different grid databases (SPAIN02, AEMET and E-OBS ), which have similar spatial resolution (approximately 25 km). The first set is multiphysics (30 km) and consists of eight members. The second set is multi-model (ESCENA) and consists of 5 members (25 km resolution). The results indicate that the skill of the ensemble members in reproducing the observed climate over Spain depends on the selected database

Factores de riesgo cardiovascular: desde los clásicos hasta los emergentes a la luz de las nuevas evidencias

    Get PDF
Cardiovascular diseases (CVD) are the main cause of morbidity and mortality worldwide, especially affecting the population over 60 years of age, they can manifest depending on their origin in coronary disease (acute myocardial infarction, angina pectoris, sudden cardiac arrest, heart failure and sudden death) and cerebrovascular disease (strokes and transient ischemic events). Several risk factors associated with the development of CVD have been described, the classic factors (modifiable and non-modifiable), and others that have been added to the list as a result of new evidence, such as inflammation, thrombosis, stress, alterations in the levels of natriuretic peptides, heavy metals/environmental pollutants, renal insufficiency, and some infectious agents. The objective of this article is to present the state of the art associated with classic cardiovascular risk factors (CRFs) and the evidence that supports emerging CRFs. &nbsp
