    Light Assisted Collisional Loss in a 85/87^{85/87}Rb Ultracold Optical Trap

    We have studied hetero- and homonuclear excited state/ground state collisions by loading both 85^{85}Rb and 87^{87}Rb into a far off resonant trap (FORT). Because of the relatively weak confinement of the FORT, we expect the hyperfine structure of the different isotopes to play a crucial role in the collision rates. This dependence on hyperfine structure allows us to measure collisions associated with long range interatomic potentials of different structure: such as long and short ranged; or such as purely attractive, purely repulsive, or mixed attractive and repulsive. We observe significantly different loss rates for different excited state potentials. Additionally, we observe that some collisional channels' loss rates are saturated at our operating intensities (~15 mW/cm2^{2}).     Theory-based design for promoting positive behaviours in an urban blue space: Pre-and-post observations of a community co-created intervention in Plymouth, United Kingdom

    Behaviour observation and mapping are useful planning tools to generate evidence to create people-friendly public spaces. Small-scale interventions are gaining popularity as cost-effective and quick solutions to regenerate degraded urban outdoor environments. Onsite behaviour observation mapping with GIS underpins evidence gathering, analysis, and planning and design decision-making. Using the BlueHealth Behaviour Assessment Tool (BBAT) developed within the EU-funded Horizon 2020 Project “Blue Health”, we carried out pre- and post- intervention observations of visits, activities and visitors' characteristics in a site at Teats Hill, Plymouth, United Kingdom. Pre-post comparison of visits and activities in three target areas the entire site before and the within-site intervention area (a small open-air theatre) and the rest of the site afterwards were examined to analyse the impact of the intervention on socialising and relaxing activities. Behaviour observation data was both spatially and statistically analysed. Key outcomes were sitting, standing, walking, activity with a dog, viewing, using a phone and socialising. Both a logistic regression model and spatial analysis using density maps and hot spot analysis confirmed an increased use of the open-air theatre and its positive impact on the use of the rest of the park (the non-intervention area). Our logit models showed that the intervention promoted positive behaviour for health (i.e. blue space activities, socialisation, and relaxing activities), greater inclusivity and diversity of visitors. We conclude that an evidence-based design approach can increase blue space accessibility, improve place affordances and promote positive behaviour and psychological well-being benefitting local communities

    Comparative study of a vitrinite-rich and an inertinite-rich Witbank coal (South Africa) using pyrolysis-gas chromatography

    Abstract This study aims to compare iso-rank vitrinite-rich and inertinite-rich coal samples to understand the impact of coal-forming processes on pyrolysis chemistry. A medium rank C bituminous coal was density-fractionated to create a vitrinite-rich and an inertinite-rich sub-sample. The vitrinite-rich sample has 83 vol% total vitrinite (mineral-matter-free basis), whereas the inertinite-rich counterpart has 66 vol% total inertinite. The vitrinite-rich sample is dominated by collotelinite and collodetrinite. Fusinite, semifusinite, and inertodetrinite are the main macerals of the inertinite-rich sample. Molecular chemistry was assessed using a pyrolysis gas chromatograph (py-GC) equipped with a thermal desorption unit coupled to a time of flight mass spectrometer (MS) (py-GC/MS) and solid-state nuclear magnetic resonance (13C CP-MAS SS NMR). The pyrolysis products of the coal samples are generally similar, comprised of low and high molecular weight alkanes, alkylbenzenes, alkylphenols, and alkyl-subtituted polycyclic aromatic hydrocarbons, although the vitrinite-rich sample is chemically more diverse. The lack of diversity exhibited by the inertinite-rich sample upon pyrolysis may be interpreted to suggest that major components were heated in their geologic history. Based on the 13C CP-MAS SS NMR analysis, the inertinite-rich sample has a greater fraction of phenolics, reflected in the py-GC/MS results as substituted and unsubstituted derivatives. The greater abundance of phenolics for the inertinite-rich sample may suggest a fire-related origin for the dominant macerals of this sample. The C2-alkylbenzene isomers (p-xylene and o-xylene) were detected in the pyrolysis products for the vitrinite-rich and inertinite-rich samples, though more abundant in the former. The presence of these in both samples likely reflects common source vegetation for the dominant vitrinite and inertinite macerals

    Measurements of methane and nitrous oxide in human breath and the development of UK scale emissions

    Exhaled human breath can contain small, elevated concentrations of methane (CH4) and nitrous oxide (N2O), both of which contribute to global warming. These emissions from humans are not well understood and are rarely quantified in global greenhouse gas inventories. This study investigated emissions of CH4 and N2O in human breath from 104 volunteers in the UK population, to better understand what drives these emissions and to quantify national-scale estimates. A total of 328 breath samples were collected, and age, sex, dietary preference, and smoking habits were recorded for every participant. The percentage of methane producers (MPs) identified in this study was 31%. The percentage of MPs was higher in older age groups with 25% of people under the age of 30 classified as MPs compared to 40% in the 30+ age group. Females (38%) were more likely to be MPs than males (25%), though overall concentrations emitted from both MP groups were similar. All participants were found to emit N2O in breath, though none of the factors investigated explained the differences in emissions. Dietary preference was not found to affect CH4 or N2O emissions from breath in this study. We estimate a total emission of 1.04 (0.86–1.40) Gg of CH4 and 0.069 (0.066–0.072) Gg of N2O in human breath annually in the UK, the equivalent of 53.9 (47.8–60.0) Gg of CO2. In terms of magnitude, these values are approximately 0.05% and 0.1% of the total emissions of CH4 and N2O reported in the UK national greenhouse gas inventories

    Increasing uptake of hepatitis C virus infection case- finding, testing, and treatment in primary care: evaluation of the HepCATT (Hepatitis C Assessment Through to Treatment) trial

    Background Hepatitis C virus (HCV) infection is a key cause of liver disease but can be cured in more than 95% of patients. Around 70 000 people in England may have undiagnosed HCV infection and many more will not have been treated. Interventions to increase case-finding in primary care are likely to be cost-effective; however, evidence of effective interventions is lacking. The Hepatitis C Assessment Through to Treatment (HepCATT) trial assessed whether a complex intervention in primary care could increase case-finding, testing, and treatment of HCV.Aim To investigate the feasibility and acceptability of the HepCATT intervention.Design and setting A qualitative study with primary care practice staff from practices in the south west of England taking part in the HepCATT trial.Method Semi-structured interviews were carried out with GPs, nurses, and practice staff to ascertain their views of the HepCATT intervention at least 1 month after implementing the intervention in their practice. Normalisation process theory, which outlines the social processes involved in intervention implementation, informed thematic data analysis.Results Participants appreciated the HepCATT intervention for increasing knowledge and awareness of HCV. Although some initial technical difficulties were reported, participants saw the benefits of using the audit tool to systematically identify patients with HCV infection risk factors and found it straightforward to use. Participants valued the opportunity to discuss HCV testing with patients, especially those who may not have been previously aware of HCV risk. Future implementation should consider fully integrating software systems and additional resources to screen patient lists and conduct tests.Conclusion When supported by a complex intervention, primary care can play a crucial role in identifying and caring for patients with HCV infection, to help stem the HCV epidemic, and prevent HCV-related illnes

    Functional characterization of two novel 5' untranslated exons reveals a complex regulation of NOD2 protein expression

    <p>Abstract</p> <p>Background</p> <p>NOD2 is an innate immune receptor for the bacterial cell wall component muramyl-dipeptide. Mutations in the leucine-rich repeat region of NOD2, which lead to an impaired recognition of muramyl-dipeptide, have been associated with Crohn disease, a human chronic inflammatory bowel disease. Tissue specific constitutive and inducible expression patterns of NOD2 have been described that result from complex regulatory events for which the molecular mechanisms are not yet fully understood.</p> <p>Results</p> <p>We have identified two novel exons of the <it>NOD2 </it>gene (designated exon 1a and 1b), which are spliced to the canonical exon 2 and constitute the 5' untranslated region of two alternative transcript isoforms (i.e. exon 1a/1b/2 and exon 1a/2). The two novel transcripts are abundantly expressed and seem to comprise the majority of NOD2 transcripts under physiological conditions. We confirm the expression of the previously known canonical first exon (designated exon 1c) of the gene in unstimulated mononuclear cells. The inclusion of the second alternative exon 1b, which harbours three short upstream open reading frames (uORFs), is downregulated upon stimulation with TNF-α or under pro-inflammatory conditions in the inflamed intestinal mucosa <it>in vivo</it>. Using the different 5' UTR splice forms fused to a firefly luciferase (LUC) reporter we demonstrate a rapamycin-sensitive inhibitory effect of the uORFs on translation efficacy.</p> <p>Conclusion</p> <p>The differential usage of two alternative promoters in the <it>NOD2 </it>gene leads to tissue-specific and context-dependent <it>NOD2 </it>transcript isoform patterns. We demonstrate for the first time that context-dependent alternative splicing is linked to uORF-mediated translational repression. The results suggest complex parallel control mechanisms that independently regulate NOD2 expression in the context of inflammatory signaling.</p

    Barriers and Facilitators of Physical Activity in Children of a South Asian Ethnicity

    Children of South Asian ethnicity residing in England have low levels of physical activity. Limited literature exists on correlates, barriers and facilitators to activity in South Asian children. The aim of this study was to fill this gap in the literature. Interviews were conducted with 10 parents of South Asian ethnicity residing in the UK. Interviews covered a description of the family setup, participants’ opinions of physical activity including barriers and facilitators and their children’s participation, as well as approaches to general parenting, and how children spend their free time. Data were analysed using thematic analysis. Key themes identified included (i) restraints on parents’ and children’s time to be physically active, (ii) the role of the family in children’s physical activity participation, (iii) situational barriers to physical activity, (iv) physical activity not a priority, (v) opportunities to be active, (vi) perception of activity level and health. A number of key barriers to South Asian children’s participation in physical activity were identified, including (i) restraints on parents and children’s time, (ii) parents providing limited support for physical activity, (iii) physical activity has a low priority. A number of facilitators were also identified (i) play, (ii) school-time, (iii) extra-curricular clubs. In this sample of South Asian parents residing in the UK several socio-cultural barriers and facilitators of their children’s physical activity have been identified. The study provides preliminary data for a larger study to ascertain if such barriers and facilitators are representative of the wider South Asian community, so that recommendations for intervention and policies can be made
