    Gastroprotective potential of frutalin, a d-galactose binding lectin, against ethanol-induced gastric lesions

    AbstractThe present study was designed to verify whether frutalin (FTL) affords gastroprotection against the ethanol-induced gastric damage and to examine the underlying mechanism(s). Gastric damage was induced by intragastric administration of 0.2ml of ethanol (96%). Mice in groups were pretreated with FTL (0.25, 0.5 and 1mg/kg; i.p.), cimetidine (100mg/kg; p.o.), or vehicle (0.9% of NaCl, 10mL/kg; p.o.), 30min before ethanol administration. They were sacrificed 30min later, the stomachs excised, and the mucosal lesion area (mm2) measured by planimetry. Gastroprotection was assessed in relation to inhibition of gastric lesion area. To study the gastroprotective mechanism(s), its relations to capsaicin-sensitive fibers, endogenous prostaglandins, nitric oxide, sulphydryls, ATP-sensitive potassium channels, adrenoceptors, opioid receptors and calcium channels were analyzed. Treatments effects on ethanol-associated oxidative stress markers GSH and MDA were measured in gastric tissue. FTL afforded a dose-unrelated gastroprotection against the ethanol damage. However, it failed to prevent the ethanol-induced changes in the levels of GSH and MDA. It was observed that the gastroprotection by FTL was greatly reduced in animals pretreated with capsazepine, indomethacin, L-NAME or glibenclamide. Considering the results, it is suggested that the FTL could probably be a good therapeutic agent for the development of new medicine for the treatment of gastric ulcer

    Amitriptyline, clomipramine, and maprotiline attenuate the inflammatory response by inhibiting neutrophil migration and mast cell degranulation

    Objective: Despite the recognized anti-inflammatory potential of heterocyclic antidepressants, the mechanisms concerning their modulating effects are not completely known. Thus, we evaluated the anti-inflammatory effect of amitriptyline, clomipramine, and maprotiline and the possible modulating properties of these drugs on neutrophil migration and mast cell degranulation. Methods: The hind paw edema and air-pouch models of inflammation were used. Male Wistar rats were treated with saline, amitriptyline, clomipramine or maprotiline (10, 30, or 90 mg/kg, per os [p.o.]) 1 h before the injection of carrageenan (300 μg/0.1 mL/paw) or dextran (500 μg/0.1 mL/paw). Then, edema formation was measured hourly. Neutrophil migration to carrageenan (500 μg/pouch) and N-formyl-methionyl-leucyl-phenylalanine (fMLP) (10-6 M/mL/pouch) was also investigated in 6-day-old air-pouch cavities. Compound 48/80-induced mast cell degranulation was assessed in the mesenteric tissues of antidepressant-treated rats. Results: All tested antidepressants prevented both carrageenan- and dextran-induced edema. The anti-inflammatory effect of these drugs partially depends on the modulation of neutrophil migration, since they significantly counteracted the chemotactic response of both carrageenan and fMLP (p < 0.01). Furthermore, amitriptyline, clomipramine and maprotiline inhibited compound 48/80-induced mast cell degranulation (p < 0.001). Conclusions: These results suggest an important anti-inflammatory role of heterocyclic antidepressants, which is dependent on the modulation of neutrophil migration and mast cell stabilization

    Polysaccharide-rich extract of Genipa americana leaves protects seizures and oxidative stress in the mice model of pentylenetetrazole-induced epilepsy

    Plant polysaccharides have biological activities in the brain and those obtained from Genipa americana leaves present antioxidant and anticonvulsant effects in the mice model of pentylenetetrazole (PTZ)-induced acute seizures. This study aimed to evaluate the polysaccharide-rich extract of Genipa americana leaves (PRE-Ga) in the models of acute seizures and chronic epilepsy (kindling) induced by PTZ. In the acute seizure model, male Swiss mice (25–35 g) received PRE-Ga (1 or 9 mg/kg; intraperitoneal- IP), alone or associated with diazepam (0.01 mg/kg), 30 min before induction of seizures with PTZ (70 mg/kg; IP). In the chronic epilepsy model, seizures were induced by PTZ (40 mg/kg) 30 min after treatment and in alternated days up to 30 days and evaluated by video. Brain areas (prefrontal cortex, hippocampus, striatum) were assessed for inflammatory and oxidative stress markers. Diazepam associated to PRE-Ga (9 mg/kg; i.p.) increased the latency of seizures in acute (222.4 ± 47.57 vs. saline: 62.00 ± 4.709 s) and chronic models (6.267 ± 0.502 vs. saline: 4.067 ± 0.407 s). In hippocampus, PRE-Ga (9 mg/kg) inhibited TNF-α (105.9 ± 5.38 vs. PTZ: 133.5 ± 7.62 pmol/g) and malondialdehyde (MDA) (473.6 ± 60.51) in the chronic model. PTZ increased glial fibrillar acid proteins (GFAP) and Iba-1 in hippocampus, which was reversed by PRE-Ga (GFAP: 1.9 ± 0.23 vs PTZ: 3.1 ± 1.3 and Iba-1: 2.2 ± 0.8 vs PTZ: 3.2 ± 1.4). PRE-Ga presents neuroprotector effect in the mice model of epilepsy induced by pentylenetetrazole reducing seizures, gliosis, inflammatory cytokines and oxidative stress

    Eugenol as a Promising Molecule for the Treatment of Dermatitis: Antioxidant and Anti-inflammatory Activities and Its Nanoformulation

    Contact dermatitis produces an inflammatory reaction primarily via stimulation of keratinocytes and cells of the immune system, which promote the release of cytokines, reactive oxygen species (ROS), and other chemical mediators. Eugenol (EUG, phenylpropanoid of essential oils) has attracted attention due to its anti-inflammatory properties, as well as antioxidant effect. On the other hand, it is volatile and insoluble and is a skin irritant. In this case, nanostructured systems have been successfully employed as a drug carrier for skin diseases since they improve both biological and pharmaceutical properties of active compounds. The cytotoxic, antioxidant, and anti-inflammatory effects of EUG were assessed in human neutrophils and keratinocytes. Additionally, polymeric nanocarries (NCEUG) were prepared to improve the chemical and irritant characteristics of EUG. EUG presented apparent safety and antioxidant and anti-inflammatory effects on human neutrophils, but presented cytotoxic effects on keratinocytes. However, the nanocapsules were able to reduce its cytotoxicity. An in vivo experiment of irritant contact dermatitis (ICD) in mice induced by TPA showed that NCEUG reduced significantly the ear edema in mice when compared to the EUG solution, as well as the leukocyte infiltration and IL-6 level, possibly due to better skin permeation and irritancy blockage. These findings suggest that EUG is a promising bioactive molecule, and its nanoencapsulation seems to be an interesting approach for the treatment of ICD

    A demanda nutricional do abacaxizeiro é alta em relação à de outras culturas e depende do cultivar, do peso do fruto, do destino da produção, do sistema e da densidade de plantio, e a variação desses fatores nem sempre está contemplada nas tabelas de adubação. O objetivo deste trabalho foi propor um Sistema de Recomendação de Fertilizantes e Corretivos para a Cultura do Abacaxi (FERTCALC-Abacaxi), elaborado a partir da sistematização e modelagem de informações de literatura e do conhecimento atual sobre adubação e nutrição mineral da cultura. O FERTCALC-Abacaxi baseia-se no método do balanço nutricional e está subdividido em dois subsistemas: o subsistema requerimento, que contempla a demanda pela planta, considerando a eficiência de recuperação dos nutrientes aplicados e uma dose para atender o critério de "sustentabilidade", e o subsistema suprimento, que compreende a oferta de nutrientes pela calagem, pelo solo, pela matéria orgânica e pelos resíduos orgânicos. Simulações realizadas com o FERTCALC-Abacaxi mostraram que o Sistema recomenda maiores doses para o cultivar Pérola em relação ao Smooth Cayenne, para plantios mais adensados e para obtenção de frutos mais pesados. Diferentemente das tabelas, as doses variaram de forma contínua com a produtividade esperada e com o teor de nutrientes no solo, sendo maiores no primeiro em relação ao segundo cultivo. Por contemplar na sua estrutura maior número de fatores envolvidos na resposta da cultura à adubação, a exemplo da produtividade esperada, do cultivar, do peso do fruto, do sistema e da densidade de plantio, e considerar a contribuição dos resíduos orgânicos do cultivo anterior, o FERTCALC-Abacaxi constitui-se numa alternativa mais versátil para recomendar doses de fertilizantes e corretivos para o abacaxizeiro, embora precise ser aperfeiçoado em futuras versões e ter seu desempenho avaliado em condições práticas.<br>The nutritional demand of pineapple plants is higher than for other crops and IT depends on cultivar, fruit weight, production destination and planting density and cultivation system. However, the variation in these factors is not always taken into account in fertilization tables. The purpose of this study was to propose a fertilizer and lime recommendation system for pineapple designated FERTCALC-Abacaxi, based on systematization and modeling of literature data and the current knowledge concerning fertilization and mineral nutrition of the crop. FERTCALC-Abacaxi is based on the nutritional balance method and is divided into two subsystems: the requirement subsystem that contemplates the plant demand, considering the efficiency of recovery of the applied nutrients and a dose to meet the requirement "sustainability"; and the supply subsystem that comprises the nutrient supply by liming, soil, organic matter, and by organic residues. Simulations with FERTCALC-Abacaxi showed that the System recommends higher doses for the Pérola cv., than for Smooth Cayenne, for denser planting systems and to obtain heavier fruits; and differently from the tables, it varied continuously with the expected productivity and soil nutrient contents, being higher in the first than in the second growing cycle. In view of the higher number of factors taken into consideration that reflect the response of the crop to fertilization, for example the expected yield, cultivar, fruit weight, planting density and system, besides taking into account the contribution of organic residues from the previous cycle, the FERTCALC- Abacaxi represents a more versatile alternative for fertilizer and lime recommendation for the pineapple crop. However, improvements in future versions of the System as well as its validation under practical conditions are necessary
