    This document presents the data analysis procedures proposed for use with the COMPTEL instrument aboard the Compton Gamma Ray Observatory (GRO) in the search for radiative neutrino decay from supernovae. The proposed analysis methodology is an extension of a standard procedure used by the COMPTEL team in searching for a variety of source types. We have applied the procedures to a set of simulated data to demonstrate the feasibility of the method to this project

    Hollow fiber clinostat for simulating microgravity in cell culture

    A clinostat for simulating microgravity on cell systems carried in a fiber fixedly mounted in a rotatable culture vessel is disclosed. The clinostat is rotated horizontally along its longitudinal axis to simulate microgravity or vertically as a control response. Cells are injected into the fiber and the ends of the fiber are sealed and secured to spaced end pieces of a fiber holder assembly which consists of the end pieces, a hollow fiber, a culture vessel, and a tension spring with three alignment pins. The tension spring is positioned around the culture vessel with its ends abutting the end pieces for alignment of the spring. After the fiber is secured, the spring is decompressed to maintain tension on the fiber while it is being rotated. This assures that the fiber remains aligned along the axis of rotation. The fiber assembly is placed in the culture vessel and culture medium is added. The culture vessel is then inserted into the rotatable portion of the clinostat and subjected to rotate at selected rpms. The internal diameter of the hollow fiber determines the distance the cells are from the axis of rotation

    Analytic Methods for Optimizing Realtime Crowdsourcing

    Realtime crowdsourcing research has demonstrated that it is possible to recruit paid crowds within seconds by managing a small, fast-reacting worker pool. Realtime crowds enable crowd-powered systems that respond at interactive speeds: for example, cameras, robots and instant opinion polls. So far, these techniques have mainly been proof-of-concept prototypes: research has not yet attempted to understand how they might work at large scale or optimize their cost/performance trade-offs. In this paper, we use queueing theory to analyze the retainer model for realtime crowdsourcing, in particular its expected wait time and cost to requesters. We provide an algorithm that allows requesters to minimize their cost subject to performance requirements. We then propose and analyze three techniques to improve performance: push notifications, shared retainer pools, and precruitment, which involves recalling retainer workers before a task actually arrives. An experimental validation finds that precruited workers begin a task 500 milliseconds after it is posted, delivering results below the one-second cognitive threshold for an end-user to stay in flow.Comment: Presented at Collective Intelligence conference, 201

    The development of a space continuous flow electrophoresis system (CFES) is discussed. The objectives of the experiment were: (1) to use a model sample material at a high concentration to evaluate the continuous flow electrophoresis process in the McDonnell Douglass CFES instrument and compare its separation resolution and sample throughput with related devices on Earth, and (2) to expand the basic knowledge of the limitations imposed by fluid flows and particle concentration effects on the electrophoresis process by careful design and evaluation of the space experiment. Hemoglobin and polysaccharide were selected as samples of concentration effects. The results from space show a large band spread of the high concentration of the single species of hemoglobin that was principally due to the mismatch of electrical conductivity between the sample and buffer

    Dynamic removal of replication protein A by Dna2 facilitates primer cleavage during Okazaki fragment processing in Saccharomyces cerevisiae

    Eukaryotic Okazaki fragments are initiated by an RNA/DNA primer, which is removed before the fragments are joined. Polymerase d displaces the primer into a flap for processing. Dna2 nuclease/helicase and flap endonuclease 1 (FEN1) are proposed to cleave the flap. The single-stranded DNA binding protein, replication protein A (RPA), governs cleavage activity. Flap-bound RPA inhibits FEN1. This necessitates cleavage by Dna2, which is stimulated by RPA. FEN1 then cuts the remaining RPA-free flap to create a nick for ligation. Cleavage by Dna2 requires that it enter the 5'-end and track down the flap. Since Dna2 cleaves the RPA-bound flap, we investigated the mechanism by which Dna2 accesses the protein-coated flap for cleavage. Using a nuclease-defective Dna2 mutant, we showed that just binding of Dna2 dissociates the flap-bound RPA. Facile dissociation is specific to substrates with a genuine flap, and will not occur with an RPA-coated single strand. We also compared the cleavage patterns of Dna2 with and without RPA to better define RPA stimulation of Dna2. Stimulation derived from removal of DNA folding in the flap. Apparently, coordinated with its dissociation, RPA relinquishes the flap to Dna2 for tracking in a way that does not allow flap structure to reform. We also found that RPA strand melting activity promotes excessive flap elongation, but it is suppressed by Dna2-promoted RPA dissociation. Overall, results indicate that Dna2 and RPA coordinate their functions for efficient flap cleavage and preparation for FEN1

    Laboratory Rearing of \u3ci\u3eLycaeides Melissa Samuelis\u3c/i\u3e (Lepidoptera: Lycaenidae), An Endangered Butterfly in Michigan

    The Karner blue butterfly (Lycaeides melissa samuelis) is listed as a federally endangered species in the United States. It occurs in oak savanna and pine barren habitats from eastern Minnesota to New Hampshire. In 1994, we successfully reared Karner blue larvae under controlled laboratory conditions for experimental purposes, and report on those rearing methods here. We collected 20 female Karner blue adults of the spring generation from two areas in Michigan, and housed them in cages in an environmental chamber at 240 -26°C for 5 days. The female butterflies produced 154 eggs, of which 72 hatched in an average of 4.5 days, and 68 first instars survived. Eggs, larvae and pupae were kept in a growth chamber at 24°C. Developmental time from egg to adult averaged 26 days; the average duration of each instar ranged from 3 to 4 days, and the average pupal duration was 8 days. Thirty three lab- oratory-reared Karner blue larvae successfully completed the 4 instars, and were released as adults into maternal collection sites. Laboratory rearing may be a viable means of providing Karner blue individuals for reintroduction into areas where the species is extirpated, for supplementation of small populations, or for research. Ultimately, such methods may become an integral part in the recovery of this and other rare invertebrate species
