    Exercise, protein metabolism, and muscle growth

    Exercise has a profound effect on muscle growth, which can occur only if muscle protein synthesis exceeds muscle protein breakdown; there must be a positive muscle protein balance. Resistance exercise improves muscle protein balance, but, in the absence of food intake, the balance remains negative (i.e., catabolic). The response of muscle protein metabolism to a resistance exercise bout lasts for 24-48 hours; thus, the interaction between protein metabolism and any meals consumed in this period will determine the impact of the diet on muscle hypertrophy. Amino acid availability is an important regulator of muscle protein metabolism. The interaction of postexercise metabolic processes and increased amino acid availability maximizes the stimulation of muscle protein synthesis and results in even greater muscle anabolism than when dietary amino acids are not present. Hormones, especially insulin and testosterone, have important roles as regulators of muscle protein synthesis and muscle hypertrophy. Following exercise, insulin has only a permissive role on muscle protein synthesis, but it appears to inhibit the increase in muscle protein breakdown. Ingestion of only small amounts of amino acids, combined with carbohydrates, can transiently increase muscle protein anabolism, but it has yet to be determined if these transient responses translate into an appreciable increase in muscle mass over a prolonged training period

    Increased net muscle protein balance in response to simultaneous and separate ingestion of carbohydrate and essential amino acids following resistance exercise

    Relative to essential amino acids (EAAs), carbohydrate (CHO) ingestion stimulates a delayed response of net muscle protein balance (NBAL). We investigated if staggered ingestion of CHO and EAA would superimpose the response of NBAL following resistance exercise, thus resulting in maximal anabolic stimulation. Eight recreationally trained subjects completed 2 trials: combined (COMB - drink 1, CHO+EAA; drink 2, placebo) and separated (SEP - drink 1, CHO; drink 2, EAA) post-exercise ingestion of CHO and EAA. Drink 1 was administered 1 h following an acute exercise bout and was followed 1 h later by drink 2. A primed, continuous infusion of l-[ring-13C6]-phenylalanine was combined with femoral arteriovenous sampling and muscle biopsies for the determination of muscle protein kinetics. Arterial amino acid concentrations increased following ingestion of EAA in both conditions. No difference between conditions was observed for phenylalanine delivery to the leg (COMB: 167 ± 23 μmol·min-1·(100 mL leg vol)-1 × 6 h; SEP: 167 ± 21 μmol·min-1·(100 mL leg vol)-1 × 6 h, P > 0.05). In the first hour following ingestion of the drink containing EAA, phenylalanine uptake was 50% greater for the SEP trial than the COMB trial. However, phenylalanine uptake was similar for COMB (110 ± 19 mg) and SEP (117 ± 24 mg) over the 6 h period. These data suggest that whereas separation of CHO and EAA ingestion following exercise may have a transient physiological impact on NBAL, this response is not reflected over a longer period. Thus, separation of CHO and EAA ingestion is unnecessary to optimize post-exercise muscle protein metabolism

    Constrained Multistate Sequence Design for Nucleic Acid Reaction Pathway Engineering

    We describe a framework for designing the sequences of multiple nucleic acid strands intended to hybridize in solution via a prescribed reaction pathway. Sequence design is formulated as a multistate optimization problem using a set of target test tubes to represent reactant, intermediate, and product states of the system, as well as to model crosstalk between components. Each target test tube contains a set of desired “on-target” complexes, each with a target secondary structure and target concentration, and a set of undesired “off-target” complexes, each with vanishing target concentration. Optimization of the equilibrium ensemble properties of the target test tubes implements both a positive design paradigm, explicitly designing for on-pathway elementary steps, and a negative design paradigm, explicitly designing against off-pathway crosstalk. Sequence design is performed subject to diverse user-specified sequence constraints including composition constraints, complementarity constraints, pattern prevention constraints, and biological constraints. Constrained multistate sequence design facilitates nucleic acid reaction pathway engineering for diverse applications in molecular programming and synthetic biology.     Suomi NPP VIIRS Prelaunch and On-orbit Geometric Calibration and Characterization

    The Visible Infrared Imager Radiometer Suite (VIIRS) sensor was launched 28 October 2011 on the Suomi National Polarorbiting Partnership (SNPP) satellite. VIIRS has 22 spectral bands covering the spectrum between 0.412 m and 12.01 m, including 16 moderate resolution bands (M-bands) with a spatial resolution of 750 m at nadir, 5 imaging resolution bands (I-bands) with a spatial resolution of 375 m at nadir, and 1 day-night band (DNB) with a near-constant 750 m spatial resolution throughout the scan. These bands are located in a visible and near infrared (VisNIR) focal plane assembly (FPA), a short- and mid-wave infrared (SWMWIR) FPA and a long-wave infrared (LWIR) FPA. All bands, except the DNB, are co-registered for proper environmental data records (EDRs) retrievals. Observations from VIIRS instrument provide long-term measurements of biogeophysical variables for climate research and polar satellite data stream for the operational communitys use in weather forecasting and disaster relief and other applications. Well Earth-located (geolocated) instrument data is important to retrieving accurate biogeophysical variables. This paper describes prelaunch pointing and alignment measurements, and the two sets of on-orbit correction of geolocation errors, the first of which corrected error from 1,300 m to within 75 m (20 I-band pixel size), and the second of which fine tuned scan angle dependent errors, bringing VIIRS geolocation products to high maturity in one and a half years of the SNPP VIIRS on-orbit operations. Prelaunch calibration and the on-orbit characterization of sensor spatial impulse responses and band-to-band co-registration (BBR) are also described

    Stimulation of muscle protein synthesis with low-dose amino acid composition in older individuals

    Essential amino acid (EAA)-based compositions have been shown to be effective stimulators of muscle protein synthesis, but the lower limit of effective dosage is not clear. We have used stable isotope tracer methodology to quantify the response of muscle protein fractional synthetic rate (FSR) to a dose of 3.6 g of a high-leucine composition of EAAs plus arginine in older subjects. Muscle protein FSR increased 0.058%/hour over 3 h following consumption. When account was taken of the total muscle mass, this increase in muscle protein FSR represented approximately 80% of ingested EAAs. We conclude that a low dose of an EAA-based composition can effectively stimulate muscle protein synthesis

    Quantifying the climate impacts of albedo changes due to biofuel production: a comparison with biogeochemical effects

    Lifecycle analysis is a tool widely used to evaluate the climate impact of greenhouse gas emissions attributable to the production and use of biofuels. In this paper we employ an augmented lifecycle framework that includes climate impacts from changes in surface albedo due to land use change. We consider eleven land-use change scenarios for the cultivation of biomass for middle distillate fuel production, and compare our results to previous estimates of lifecycle greenhouse gas emissions for the same set of land-use change scenarios in terms of CO2e per unit of fuel energy. We find that two of the land-use change scenarios considered demonstrate a warming effect due to changes in surface albedo, compared to conventional fuel, the largest of which is for replacement of desert land with salicornia cultivation. This corresponds to 222 gCO2e/MJ, equivalent to 3890% and 247% of the lifecycle GHG emissions of fuels derived from salicornia and crude oil, respectively. Nine of the land-use change scenarios considered demonstrate a cooling effect, the largest of which is for the replacement of tropical rainforests with soybean cultivation. This corresponds to − 161 gCO2e/MJ, or − 28% and − 178% of the lifecycle greenhouse gas emissions of fuels derived from soybean and crude oil, respectively. These results indicate that changes in surface albedo have the potential to dominate the climate impact of biofuels, and we conclude that accounting for changes in surface albedo is necessary for a complete assessment of the aggregate climate impacts of biofuel production and use.Federal Aviation AdministrationUnited States.
