    Quantification of MUCIN 1, cell surface associated and MUCIN16, cell surface associated proteins in tears and conjunctival epithelial cells collected from postmenopausal women

Molecular Vision, 19, 970–979.Purpose: To quantify the expression of mucin 1, cell surface associated (MUC1) and mucin 16, cell surface associated (MUC16) proteins and messenger ribonucleic acid (mRNA) in a cohort of postmenopausal women (PMW), to explore the relationship between mucin expression, dry eye symptomology, and tear stability.Methods: Thirty-nine healthy PMW (>50 years of age) were enrolled in this study. No specific inclusion criteria were used to define dry eye; instead, a range of subjects were recruited based on responses to the Allergan Ocular Surface Disease Index (OSDI) questionnaire and tear stability measurements as assessed by non-invasive tear breakup time (NITBUT). Tears were collected from the inferior tear meniscus using a disposable glass capillary tube, and total RNA and total protein were isolated from conjunctival epithelial cells collected via impression cytology. Expression of membrane-bound and soluble MUC1 and MUC16 were quantified with western blotting, and expression of MUC1 and MUC16 mRNA was assessed with real-time PCR.Results: OSDI responses ranged from 0 to 60, and NITBUT ranged from 18.5 to 2.9 s. Only two statistically significant correlations were found: soluble MUC16 protein concentration and MUC16 mRNA expression with OSDI vision related (−0.47; p=0.01) and ocular symptom (0.39; p=0.02) subscores, respectively. Post hoc exploratory analysis on absolute expression values was performed on two subsets of subjects defined as asymptomatic (OSDI ≤6, n=12) and moderate to severe symptomatic (OSDI ≥20, n=12). The only significant difference between the two subgroups was a significant reduction in MUC16 mRNA expression found in the symptomatic dry eye group (1.52±1.19 versus 0.57±0.44; p=0.03).Conclusions: A broad exploration of mucin expression compared to either a sign (NITBUT) or symptoms of dry eye failed to reveal compelling evidence supporting a significant relationship, other than a potential association between MUC16 with specific symptoms. Furthermore, comparison of mucin protein and expression levels between the asymptomatic and moderate to severe symptomatic subgroups revealed only one significant difference, a reduction in MUC16 mRNA expression in the symptomatic subgroup.Funding for this study was provided by Alcon Research Ltd and the Natural Sciences and Engineering Research Council of Canada (NSERC)

    En cuanto al sector salud la calidad está enfocada principalmente en la satisfacción del cliente, buscando brindar un servicio completo de bienestar. El Ministerio de Salud se ha esforzado y enfocado en aumentar la calidad de sus servicios, emitiendo a través de los años una serie de documentos relacionados con la gestión de la calidad en los servicios de salud, creando diversos protocolos para áreas especializadas de atención a pacientes y lineamientos dirigidos al personal de salud para el cumplimiento de sus deberes y la atención de pacientes con eficiencia, calidad y calidez. Como resultado de este esfuerzo el MINSAL ha obtenido una serie de reconocimientos por la mejora de sus servicios y la calidad que se brinda a los pacientes. De acuerdo a lo que plantea la misión, la visión y los objetivos estratégicos, el SEM tiene la responsabilidad de brindar servicios de atención en emergencias médicas, en coordinación con otras instancias involucradas para salvar vidas. En este trabajo se detalla en qué consiste la norma ISO 9001:2015, sus beneficios, los objetivos y la información documentada requerida para obtener la certificación. Se realizó un estudio de la situación actual del SEM, en los resultados de la metodología FODA se observa que el SEM debe reaccionar ante posibles amenazas que existen en el entorno externo y las debilidades que se tienen, y así realizar planes preventivos y desarrollar estrategias para minimizar los efectos que éstos tengan en su funcionamiento, asimismo, aprovechar las oportunidades que probablemente ocurran y las fortalezas con las que cuente. Con relación al cuestionario ISO se identificó que el SEM actualmente no cuenta con el cumplimiento de la totalidad de los requisitos de la norma ISO 9001:2015, sin embargo, sí realiza esfuerzos para brindar los servicios a la ciudadanía, cuenta con un avance en el contexto de la organización, sobretodo en el conocimiento del entorno y la relación con las partes relacionadas; en cuanto al liderazgo es el capítulo que presenta un mayor porcentaje de cumplimiento, ya que existe interés por la dirección dar seguimiento a los servicios que se brindan, el resto de ítems presentan un grado de cumplimiento menor al 20% lo que refleja que existe deficiencia; por todo lo anterior, se refleja una oportunidad para desarrollar en el SEM un proyecto de mejora e implementar un Sistema de Gestión que permita mejorar sus procesos y el servicio que brinda a sus usuarios. Como propuesta se elaboró el Manual de Calidad y el Manual de Registros y Procedimiento; asimismo, se presentan ejes estratégicos que son de suma importancia, ya que son parte del engranaje funcional de un Sistema de Gestión de la Calidad. Para finalizar, es necesario indicar que la esencia en el contenido de este trabajo viene dada por la estructura documentada del SGC del modelo ISO 9001:2015 abordado a detalle en el capítulo 4; que contiene aspectos que enriquecerán al servicio prehospitalario del SEM. El manual de calidad, sus requisitos y procedimientos; establecen mecanismos que pueden dar resultados de forma progresiva partiendo de resultados básicos a corto plazo

    THE SPRING MIGRATION OF ADULT NORTH AMERICAN OSPREYS

    Most North American Ospreys (Pandion haliaetus) are migratory, breeding in northern latitudes and migrating long distances to and from their wintering grounds in the tropics. Although fall migration patterns of North American Ospreys have been described and studied, very little has been published about the spring migration of these birds. We used satellite telemetry to: (1) determine the characteristics (timing, duration, migratory routes) of spring migrations of Ospreys; (2) determine if differences in spring migration patterns existed between sexes and among three breeding populations (east coast, midwestern, and western); and (3) compare consecutive fall and spring migrations of individual Ospreys. The median dates for departure from the wintering grounds and arrival on the breeding grounds did not differ significantly between adult male and female Ospreys. Compared to their fall migrations, all male and all east coast Ospreys spent fewer days on migration, fewer days in stopover periods along the migration route, traveled shorter distances overall, and traveled farther (on average) each day during spring. In contrast, fall and spring migration characteristics of all female and western Ospreys were similar. Our findings suggest that, although sex and breeding location might influence the spring migration strategy used by individual Ospreys, both males and females minimize the time spent on migration to ensure a timely arrival on the breeding grounds to establish or defend a nesting territory

    The Third wave in globalization theory

    This essay examines a proposition made in the literature that there are three waves in globalization theory—the globalist, skeptical, and postskeptical or transformational waves—and argues that this division requires a new look. The essay is a critique of the third of these waves and its relationship with the second wave. Contributors to the third wave not only defend the idea of globalization from criticism by the skeptics but also try to construct a more complex and qualified theory of globalization than provided by first-wave accounts. The argument made here is that third-wave authors come to conclusions that try to defend globalization yet include qualifications that in practice reaffirm skeptical claims. This feature of the literature has been overlooked in debates and the aim of this essay is to revisit the literature and identify as well as discuss this problem. Such a presentation has political implications. Third wavers propose globalist cosmopolitan democracy when the substance of their arguments does more in practice to bolster the skeptical view of politics based on inequality and conflict, nation-states and regional blocs, and alliances of common interest or ideology rather than cosmopolitan global structures

    Inorganic speciation of dissolved elements in seawater: the influence of pH on concentration ratios

    Assessments of inorganic elemental speciation in seawater span the past four decades. Experimentation, compilation and critical review of equilibrium data over the past forty years have, in particular, considerably improved our understanding of cation hydrolysis and the complexation of cations by carbonate ions in solution. Through experimental investigations and critical evaluation it is now known that more than forty elements have seawater speciation schemes that are strongly influenced by pH. In the present work, the speciation of the elements in seawater is summarized in a manner that highlights the significance of pH variations. For elements that have pH-dependent species concentration ratios, this work summarizes equilibrium data (S = 35, t = 25°C) that can be used to assess regions of dominance and relative species concentrations. Concentration ratios of complex species are expressed in the form log[A]/[B] = pH - C where brackets denote species concentrations in solution, A and B are species important at higher (A) and lower (B) solution pH, and C is a constant dependent on salinity, temperature and pressure. In the case of equilibria involving complex oxy-anions (MO(x)(OH)(y)) or hydroxy complexes (M(OH)(n)), C is written as pK(n )= -log K(n )or pK(n)* = -log K(n)* respectively, where K(n )and K(n)* are equilibrium constants. For equilibria involving carbonate complexation, the constant C is written as pQ = -log(K(2)(l)K(n )[HCO(3)(-)]) where K(2)(l )is the HCO(3 )(- )dissociation constant, K(n )is a cation complexation constant and [HCO(3)(-)] is approximated as 1.9 × 10(-3 )molar. Equilibrium data expressed in this manner clearly show dominant species transitions, ranges of dominance, and relative concentrations at any pH

    A preliminary evaluation of a single session behavioural activation intervention to improve well-being and prevent depression in carers

    Background: Major depressive disorder is predicted to be the lead cause of disease burden by 2030. Despite evidence suggesting that major depressive disorder can be prevented, little attention has been paid to developing interventions for this purpose. As research suggests that high levels of subjective well-being may protect against depression, an intervention that can enhance and maintain subjective well-being may assist in preventing major depressive disorder. Behavioural activation (BA) is a promising intervention that has been observed to both effectively treat depression and also enhance subjective well-being, even in a single session. Method: A randomised control design was used to investigate the efficacy of a single session of BA to boost well-being and reduce distress in a community sample of carers (N = 13), who may be at increased risk of major depressive disorder. Outcome measures assessed symptoms of depression, anxiety, stress, and well-being and the lifestyle factors of perceived environmental reward and the extent to which individuals lived in accordance with their personal values. Results: Generalised linear mixed modelling revealed significant group × time interactions for stress scores and valued living, indicating a treatment effect on these outcomes. Conclusions: Findings provide preliminary support for the effectiveness of a single session BA intervention to improve outcomes of carers

    RNA Polymerase II pausing temporally coordinates cell cycle progression and erythroid differentiation

    The controlled release of promoter-proximal paused RNA polymerase II (Pol II) into productive elongation is a major step in gene regulation. However, functional analysis of Pol II pausing is difficult because factors that regulate pause release are almost all essential. In this study, we identified heterozygous loss-of-function mutations in SUPT5H, which encodes SPT5, in individuals with β-thalassemia unlinked to HBB mutations. During erythropoiesis in healthy human cells, cell cycle genes were highly paused at the transition from progenitors to precursors. When the pathogenic mutations were recapitulated by SUPT5H editing, Pol II pause release was globally disrupted, and the transition from progenitors to precursors was delayed, marked by a transient lag in erythroid-specific gene expression and cell cycle kinetics. Despite this delay, cells terminally differentiate, and cell cycle phase distributions normalize. Therefore, hindering pause release perturbs proliferation and differentiation dynamics at a key transition during erythropoiesis, revealing a role for Pol II pausing in the temporal coordination between the cell cycle and differentiation

    Stellar Astrophysics and Exoplanet Science with the Maunakea Spectroscopic Explorer (MSE)

    The Maunakea Spectroscopic Explorer (MSE) is a planned 11.25-m aperture facility with a 1.5 square degree field of view that will be fully dedicated to multi-object spectroscopy. A rebirth of the 3.6m Canada-France-Hawaii Telescope on Maunakea, MSE will use 4332 fibers operating at three different resolving powers (R ~ 2500, 6000, 40000) across a wavelength range of 0.36-1.8mum, with dynamical fiber positioning that allows fibers to match the exposure times of individual objects. MSE will enable spectroscopic surveys with unprecedented scale and sensitivity by collecting millions of spectra per year down to limiting magnitudes of g ~ 20-24 mag, with a nominal velocity precision of ~100 m/s in high-resolution mode. This white paper describes science cases for stellar astrophysics and exoplanet science using MSE, including the discovery and atmospheric characterization of exoplanets and substellar objects, stellar physics with star clusters, asteroseismology of solar-like oscillators and opacity-driven pulsators, studies of stellar rotation, activity, and multiplicity, as well as the chemical characterization of AGB and extremely metal-poor stars.Comment: 31 pages, 11 figures; To appear as a chapter for the Detailed Science Case of the Maunakea Spectroscopic Explore
