    Leadership in the British civil service: an interpretation

    This article is essentially a polemic. The argument is that when politicians and officials now talk of ‘leadership’ in the British civil service they do not use that word in the way in which it was previously used. In the past leading civil servants, acting in partnership with ministers and within constitutional constraints, exercised leadership in the sense of setting example, inspiring confidence and encouraging loyalty. The loosening of traditional constitutional patterns, the marginalization of senior officials in the policy process and the emergence of business methods as the preferred model for public ­administration have led to a political and administrative environment in which leadership in the British civil service is now about encouraging patterns of behaviour which fit in with these changes. Leadership skills are now about ‘delivery’; they are not about motivation. It is time for politicians, officials and scholars to be open about this

    Elevated levels of inflammatory cytokines predict survival in idiopathic and familial pulmonary arterial hypertension

    BACKGROUND: Inflammation is a feature of pulmonary arterial hypertension (PAH), and increased circulating levels of cytokines are reported in patients with PAH. However, to date, no information exists on the significance of elevated cytokines or their potential as biomarkers. We sought to determine the levels of a range of cytokines in PAH and to examine their impact on survival and relationship to hemodynamic indexes. METHODS AND RESULTS: We measured levels of serum cytokines (tumor necrosis factor-alpha, interferon-gamma and interleukin-1beta, -2, -4, -5, -6, -8, -10, -12p70, and -13) using ELISAs in idiopathic and heritable PAH patients (n=60). Concurrent clinical data included hemodynamics, 6-minute walk distance, and survival time from sampling to death or transplantation. Healthy volunteers served as control subjects (n=21). PAH patients had significantly higher levels of interleukin-1beta, -2, -4, -6, -8, -10, and -12p70 and tumor necrosis factor-alpha compared with healthy control subjects. Kaplan-Meier analysis showed that levels of interleukin-6, 8, 10, and 12p70 predicted survival in patients. For example, 5-year survival with interleukin-6 levels of >9 pg/mL was 30% compared with 63% for patients with levels < or = 9 pg/mL (P=0.008). In this PAH cohort, cytokine levels were superior to traditional markers of prognosis such as 6-minute walk distance and hemodynamics. CONCLUSIONS: This study illustrates dysregulation of a broad range of inflammatory mediators in idiopathic and familial PAH and demonstrates that cytokine levels have a previously unrecognized impact on patient survival. They may prove to be useful biomarkers and provide insight into the contribution of inflammation in PAH

    Measles virus causes immunogenic cell death in human melanoma

    Oncolytic viruses (OV) are promising treatments for cancer, with several currently undergoing testing in randomised clinical trials. Measles virus (MV) has not yet been tested in models of human melanoma. This study demonstrates the efficacy of MV against human melanoma. It is increasingly recognised that an essential component of therapy with OV is the recruitment of host anti-tumour immune responses, both innate and adaptive. MV-mediated melanoma cell death is an inflammatory process, causing the release of inflammatory cytokines including type-1 interferons and the potent danger signal HMGB1. Here, using human in vitro models, we demonstrate that MV enhances innate antitumour activity, and that MV-mediated melanoma cell death is capable of stimulating a melanoma-specific adaptive immune response

    Titan's temporal evolution in stratospheric trace gases near the poles

    International audienceWe analyze spectra acquired by the Cassini/Composite Infrared Spectrometer (CIRS) at high resolution from October 2010 until September 2014 in nadir mode. Up until mid 2012, Titan’s Northern atmosphere exhibited the enriched chemical content found since the Voyager days (November 1980), with a peak around the Northern Spring Equinox (NSE) in 2009. Since then, we have observed the appearance at Titan’s south pole of several trace species for the first time, such as HC3N and C6H6, observed only at high northern latitudes before equinox. We investigate here latitudes poleward of 50°S and 50°N from 2010 (after the Southern Autumnal Equinox) until 2014. For some of the most abundant and longest-lived hydrocarbons (C2H2, C2H6 and C3H8) and CO2, the evolution in the past 4 years at a given latitude is not very significant within error bars especially until mid-2013. In more recent dates, these molecules show a trend for increase in the south. This trend is dramatically more pronounced for the other trace species, especially in 2013–2014, and at 70°S relative to 50°S. These two regions then demonstrate that they are subject to different dynamical processes in and out of the polar vortex region. For most species, we find higher abundances at 50°N compared to 50°S, with the exception of C3H8, CO2, C6H6 and HC3N, which arrive at similar mixing ratios after mid-2013. While the 70°N data show generally no change with a trend rather to a small decrease for most species within 2014, the 70°S results indicate a strong enhancement in trace stratospheric gases after 2012. The 663 cm−1 HC3N and the C6H6 674 cm−1 emission bands appeared in late 2011/early 2012 in the south polar regions and have since then exhibited a dramatic increase in their abundances. At 70°S HC3N, HCN and C6H6 have increased by 3 orders of magnitude over the past 3–4 years while other molecules, including C2H4, C3H4 and C4H2, have increased less sharply (by 1–2 orders of magnitude). This is a strong indication of the rapid and sudden buildup of the gaseous inventory in the southern stratosphere during 2013–2014, as expected as the pole moves deeper into winter shadow. Subsidence gases that accumulate in the absence of ultraviolet sunlight, evidently increased quickly since 2012 and some of them may be responsible also for the reported haze decrease in the north and its appearance in the south at the same time

    American political affiliation, 2003–43: a cohort component projection

    The recent rise and stability in American party identification has focused interest on the long-term dynamics of party bases. Liberal commentators cite immigration and youth as forces which will produce a natural Democratic advantage in the future while conservative writers highlight the importance of high Republican fertility in securing Republican growth. These concerns foreground the neglect of demography within political science. This paper addresses this omission by conducting the first ever cohort component projection of American partisan populations to 2043 based on survey and census data. A number of scenarios are modeled, but, on current trends, we predict that American partisanship will shift much less than the nation’s ethnic composition because the parties’ age structures are similar. Still, our projections find that the Democrats gain two to three percentage points from the Republicans by 2043, mainly through immigration, though Republican fertility may redress the balance in the very long term
