    The Disciplined Use of Simplifying Assumptions

Columbia, Maryland, April 19-21, 1982.Simplifying assumptions — everyone uses them but no one's programming tool explicitly supports them. In programming, as in other kinds of engineering design, simplifying assumptions are an important method for dealing with complexity. Given a complex programming problem, expert programmers typically choose simplifying assumptions which, though false, allow them to arrive rapidly at a program which addresses the important features of the problem without being distracted by all of its details. The simplifying assumptions are then incrementally retracted with corresponding modifications to the initial program. This methodology is particularly applicable to rapid prototyping because the main questions of interest can often be answered using only the initial program. Simplifying assumptions can easily be misused. In order to use them effectively two key issues must be addressed. First, simplifying assumptions should be chosen which simplify the design problems significantly without changing the essential character of the program which needs to be implemented. Second, the designer must keep track of all the assumptions he is making so that he can later retract them in an orderly manner. By explicitly dealing with these issues, a programming assistant system could directly support the use of simplifying assumptions as a disciplined part of the software development process.MIT Artificial Intelligence Laborator

    Formalizing Reusable Software Components

    This paper has been accepted by the ITT Workshop on Reusability in Programming, Newport RI, September 7-9, 1983.There has been a long-standing desire in computer science for a way of collecting and using libraries of standard software components. Unfortunately, there has been only limited success in actually doing this. We believe that the lack of success stems not from any resistance to the idea, nor from any lack of trying, but rather from the difficulty of choosing an appropriate formalism for representing components. In this paper we define five desiderata for a good formalization of reusable software components and discuss many of the formalisms which have been used for representing components in light of these desiderata. We then briefly describe a formalism we are developing — the Plan Calculus — which seeks to satisfy these desiderata by combining together the best features of prior formalisms.MIT Artificial Intelligence Laborator

    English workers have reported that doubling the feed from 4 to 8 lb. for one feeding after breeding tended to increase the number of ovulation sites and the number of pigs born. Workers at Kentucky have also found similar results when gilts were fed ad libitum for a 12-hour period following breeding. Previous work done at this station has failed to confirm these reports. The experiment reported here is a further attempt to substantiate this work under South Dakota conditions

    The Metal-Poor Halo of the Andromeda Spiral Galaxy (M31)

    We present spectroscopic observations of red giant branch (RGB) stars over a large expanse in the halo of the Andromeda spiral galaxy (M31), acquired with the DEIMOS instrument on the Keck II 10-m telescope. Using a combination of five photometric/spectroscopic diagnostics -- (1) radial velocity, (2) intermediate-width DDO51 photometry, (3) Na I equivalent width (surface gravity sensitive), (4) position in the color-magnitude diagram, and (5) comparison between photometric and spectroscopic [Fe/H] estimates -- we isolate over 250 bona fide M31 bulge and halo RGB stars located in twelve fields ranging from R = 12-165kpc from the center of M31 (47 of these stars are halo members with R > 60 kpc). We derive the photometric and spectroscopic metallicity distribution function of M31 RGB stars in each of these fields. The mean of the resulting M31 spheroid (bulge and halo) metallicity distribution is found to be systematically more metal-poor with increasing radius, shifting from = -0.47+/-0.03 (sigma = 0.39) at R = -0.94+/-0.06 (sigma = 0.60) at R ~ 30 kpc to = -1.26+/-0.10 (sigma = 0.72) at R > 60 kpc, assuming [alpha/Fe] = 0.0. These results indicate the presence of a metal-poor RGB population at large radial distances out to at least R = 160 kpc, thereby supporting our recent discovery of a stellar halo in M31: its halo and bulge (defined as the structural components with R^{-2} power law and de Vaucouleurs R^{1/4} law surface brightness profiles, respectively) are shown to have distinct metallicity distributions. If we assume an alpha-enhancement of [alpha/Fe] = +0.3 for M31's halo, we derive = -1.5+/-0.1 (sigma = 0.7). Therefore, the mean metallicity and metallicity spread of this newly found remote M31 RGB population are similar to those of the Milky Way halo.Comment: Accepted for publication in ApJ on May 4th, 2006 (submitted on Jan 30, 2006). 16 pages, 13 figures, 3 table

    Roles of GRK and PDE4 Activities in the Regulation of β2 Adrenergic Signaling

    An important focus in cell biology is understanding how different feedback mechanisms regulate G protein–coupled receptor systems. Toward this end we investigated the regulation of endogenous β2 adrenergic receptors (β2ARs) and phosphodiesterases (PDEs) by measuring cAMP signals in single HEK-293 cells. We monitored cAMP signals using genetically encoded cyclic nucleotide-gated (CNG) channels. This high resolution approach allowed us to make several observations. (a) Exposure of cells to 1 μM isoproterenol triggered transient increases in cAMP levels near the plasma membrane. Pretreatment of cells with 10 μM rolipram, a PDE4 inhibitor, prevented the decline in the isoproterenol-induced cAMP signals. (b) 1 μM isoproterenol triggered a sustained, twofold increase in phosphodiesterase type 4 (PDE4) activity. (c) The decline in isoproterenol-dependent cAMP levels was not significantly altered by including 20 nM PKI, a PKA inhibitor, or 3 μM 59-74E, a GRK inhibitor, in the pipette solution; however, the decline in the cAMP levels was prevented when both PKI and 59-74E were included in the pipette solution. (d) After an initial 5-min stimulation with isoproterenol and a 5-min washout, little or no recovery of the signal was observed during a second 5-min stimulation with isoproterenol. (e) The amplitude of the signal in response to the second isoproterenol stimulation was not altered when PKI was included in the pipette solution, but was significantly increased when 59-74E was included. Taken together, these data indicate that either GRK-mediated desensitization of β2ARs or PKA-mediated stimulation of PDE4 activity is sufficient to cause declines in cAMP signals. In addition, the data indicate that GRK-mediated desensitization is primarily responsible for a sustained suppression of β2AR signaling. To better understand the interplay between receptor desensitization and PDE4 activity in controlling cAMP signals, we developed a mathematical model of this system. Simulations of cAMP signals using this model are consistent with the experimental data and demonstrate the importance of receptor levels, receptor desensitization, basal adenylyl cyclase activity, and regulation of PDE activity in controlling cAMP signals, and hence, on the overall sensitivity of the system
