    Avaliação geoquímica de óleos brasileiros com ênfase nos hidrocarbonetos aromáticos

    Aromatic compounds are still poorly evaluated in geochemical studies of Brazilian oil samples. For this reason, the objective of this work was to analyze and evaluate 26 oil samples from four Brazilian sedimentary basins. Aromatic hydrocarbon fractions have been analyzed by gas chromatography coupled to mass spectrometry. The results were also compared with previously evaluated saturated hydrocarbon data. Maturation parameters obtained using the 4- and 1-methyl-dibenzothiophenes and trimethyl-dibenzothiophenes led to the best results because a better grouping was observed. For this correlation, the samples were grouped by maturity degree enabling the routine use of these parameters to evaluate oil maturity levels

    The GTPase TcRjl of the human pathogen Trypanosoma cruzi is involved in the cell growth and differentiation

    AbstractThe protozoan parasite Trypanosoma cruzi, the etiological agent of Chagas Disease, undergoes through a complex life cycle where rounds of cell division and differentiation occur initially in the gut of triatominae vectors and, after transmission, inside of infected cells in vertebrate hosts. Members of the Ras superfamily of GTPases are molecular switches which play pivotal regulatory functions in cell growth and differentiation. We have previously described a novel GTPase in T. cruzi, TcRjl, which belongs to the RJL family of Ras-related GTP binding proteins. Here we show that most of TcRjl protein is found bound to GTP nucleotides and may be locked in this stage. In addition, we show that TcRjl is located close to the kinetoplast, in a region corresponding possibly to flagellar pocket of the parasite and the expression of a dominant-negative TcRjl construct (TcRjlS37N) displays a significative growth phenotype in reduced serum medium. Remarkably, overexpression of TcRjl inhibits differentiation of epimastigotes to trypomastigote forms and promotes the accumulation of intermediate differentiation stages. Our data suggest that TcRjl might play a role in the control of the parasite growth and differentiation

    O ENSINO DA ANTROPOMETRIA NA ESCOLA: UMA PROPOSTA NA EDUCAÇÃO EM SAÚDE

    The aim was to discuss didactic-pedagogical possibility of the teaching of anthropometry knowledge as content of the School Physical Education. The textual construction considered the approach between education and health in a perspective of critical, autonomous and conscious formation of the participants of the classes. This is a study in the theoretical test format. Anthropometry is recognized as content that allows the understanding and analysis of the social phenomenon of obesity. The Physical Education classes aim at appropriating, experiencing and analyzing the knowledge related to the body. Then, a proposal was made for the use of the anthropometry content considering the conceptual, procedural and attitudinal perspectives for the school physical education. The possibility brings elements to extend the understanding and pedagogical organization of the knowledge of Physical Education at school, making the classes meaningful and able to transcend to the specific objectives, allowing orientation of behavior towards the theme, from an intentional intervention and anchored with the pedagogical orientations contemporary art.Se objetivó discutir posibilidad didáctico-pedagógica de la enseñanza del conocimiento antropometría como contenido de la Educación Física escolar. La construcción textual consideró la aproximación entre educación y salud en una perspectiva de formación crítica, autónoma y consciente de los participantes de las clases. Se trata de un estudio en el formato de ensayo teórico. La antropometría es reconocida como contenido que permite la comprensión y análisis del fenómeno social de la obesidad. Las clases de Educación Física apuntan a apropiación, vivencia y análisis de los conocimientos relativos al cuerpo. Entonces, se elaboró una propuesta para utilizar el contenido antropométrico considerando las perspectivas conceptuales, procedimental y actitudinal para la educación física escolar. La posibilidad trae elementos para ampliar la comprensión y organización pedagógica de los conocimientos de la Educación Física escolar, haciendo las clases significativas y capaces de trascender a los objetivos puntuales, posibilitando orientación de conductas frente al tema, a partir de una intervención intencional y anclada con las orientaciones pedagógicas contemporánea.Objetivou-se discutir possibilidade didático-pedagógica do ensino do conhecimento antropometria enquanto conteúdo da Educação Física escolar.     Aerobic exercise and telomere length in patients with systolic heart failure : protocol study for a randomized controlled trial

    Background: Heart failure (HF) with reduced ejection fraction (HFrEF) is a syndrome that leads to fatigue and reduced functional capacity due to disease-related pathophysiological mechanisms. Aerobic exercise (AERO) plays a key role in improving HF outcomes, such as an increase in peak oxygen uptake (VO2peak). In addition, HF promotes cell senescence, which involves reducing telomere length. Several studies have shown that patients with a worse prognosis (i.e., reduced VO2 peak) also have shorter telomeres. However, the effects of AERO on telomere length in patients with HFrEF are still unknown. In an attempt to fill this gap, we designed a study to determine the effects of 16 weeks of aerobic training (32 sessions) on telomere length in HFrEF patients. Methods: In this single-center randomized controlled trial, men and women between 50 and 80 years old will be allocated into two different groups: a moderate-intensity aerobic training and a control grouTelomere length, functional capacity, echocardiographic variables, endothelial function, and walking ability will be assessed before and after the 16-week intervention period. Discussion: Understanding the role of physical exercise in biological aging in HFrEF patients is relevant. Due to cell senescence, these individuals have shown a shorter telomere length. AERO can delay biological aging according to a balance in oxidative stress through antioxidant action. Positive telomere length results are expected for the aerobic training group

    Biocompatible Magic Sized Quantum Dots: Luminescent Markers and Probes

    Nanoscience and nanobiotechnology have aroused great academic and technological interest. Works relating biomaterials at the nanoscale can reach new biotechnologies and help in the development and use of tools for bioimage and diagnosis applications. In this work we demonstrated the advantages of magic sized quantum dots as luminescent markers and probes to bioimage applications. The visualization of MSQDs bioconjugated with biological probes in cells were performed at periods greater than 2 h, and visualization with no commercial dye would not be possible. Therefore, we demonstrated that theses biocompatible nanocrystals are luminescent markers and probes to diagnosis

    Biocompatibility of Doped Semiconductors Nanocrystals and Nanocomposites

    Exposure of humans and environment to nanocrystals are inevitable, and nanotoxicological analyses are a requirement. The wide variety of nanocrystals with different applications is increasing, and characterization of their effects after exposure includes their potential toxicity and uses. This review summarizes the characterization of doped nanocrystals and nanocomposites, Ca-doped ZnO, Ag- and Eu-doped ZnO and Ni-doped ZnO NCs, their biocompatibility and applications. This review uncovers how these nanocrystals present desirable biocompatible properties, which can be useful as antitumoral and antimicrobial inducing agents, which differ markedly from toxic properties observed in other general nanocrystals

    Comparative Proteomic Analyses Between Biofilm-Forming and Non-biofilm-Forming Strains of Corynebacterium pseudotuberculosis Isolated From Goats

    Caseous lymphadenitis (CLA) is a chronic disease that affects small ruminants and causes economic losses in the associated breeding system. The causative agent of CLA is Corynebacterium pseudotuberculosis, a Gram-positive bacterium that exhibits tropism for external and internal lymph nodes and induces abscess formation in the host. Bacterial communities often produce a biofilm matrix that serves various functions, including protection against hostile environmental conditions, antibiotics, and the host immune response. Although biofilm formation has been reported for C. pseudotuberculosis, not all strains demonstrate this property in culture. In this work, we report the first comparative proteomic analysis of one biofilm-forming (CAPJ4) and one biofilm-non-forming strain (CAP3W) of C. pseudotuberculosis isolated from goats. Bacterial whole cell protein extracts were obtained for mass spectrometry analyses. Using LC-MS/MS, our studies reveal three and four proteins exclusively found in the CAPJ4 and CAP3W proteome, respectively. In addition, label-free quantitative analysis identified 40 proteins showing at-least 2-fold higher values in CAPJ4 compared CAP3W proteome Notably, CAPJ4 differentially synthesized the penicillin-binding protein, which participates in the formation of peptidoglycans. CAPJ4 also exhibited upregulation of N-acetylmuramoyl-L-alanine amidase and galactose-1-phosphate uridylyltransferase, which are involved in biofilm formation and exopolysaccharide biosynthesis. Here, we demonstrate that biofilm formation in C. pseudotuberculosis is likely associated with specific proteins, some of which were previously shown to be associated with virulence and biofilm formation in other organisms. Our findings may drive studies related to the bacterial mechanisms involved in the biofilm formation, in addition to providing targets for the treatment of CLA.Fil: Sá, Maria Conceição Aquino de.
