    Variability of 137Cs and 40K soil-to-fruit transfer factor in tropical lemon trees during the fruit development period

    In this investigation we evaluate the soil uptake of 137Cs and 40K by tropical plants and their consequent translocation to fruits, by calculating the soil-to-fruit transfer factors defined as Fv = [concentration of radionuclide in fruit (Bq kg-1 dry mass)/concentration of radionuclide in soil (Bq kg-1 dry mass in upper 20 cm)]. In order to obtain Fv values, the accumulation of these radionuclides in fruits of lemon trees (Citrus limon B.) during the fruit growth was measured. A mathematical model was calibrated from the experimental data allowing simulating the incorporation process of these radionuclides by fruits. Although the fruit incorporates a lot more potassium than cesium, both radionuclides present similar absorption patterns during the entire growth period. Fv ranged from 0.54 to 1.02 for 40K and from 0.02 to 0.06 for 137Cs. Maximum Fv values are reached at the initial time of fruit growth and decrease as the fruit develops, being lowest at the maturation period. As a result of applying the model a decreasing exponential function is derived for Fv as time increases.     Cytokine Production but Lack of Proliferation in Peripheral Blood Mononuclear Cells from Chronic Chagas' Disease Cardiomyopathy Patients in Response to T. cruzi Ribosomal P Proteins

    Background:Trypanosoma cruzi ribosomal P proteins, P2β and P0, induce high levels of antibodies in patients with chronic Chagas' disease Cardiomyopathy (CCC). It is well known that these antibodies alter the beating rate of cardiomyocytes and provoke apoptosis by their interaction with β1-adrenergic and M2-muscarinic cardiac receptors. Based on these findings, we decided to study the cellular immune response to these proteins in CCC patients compared to non-infected individuals.Methodology/Principal findings:We evaluated proliferation, presence of surface activation markers and cytokine production in peripheral blood mononuclear cells (PBMC) stimulated with P2β, the C-terminal portion of P0 (CP0) proteins and T. cruzi lysate from CCC patients predominantly infected with TcVI lineage. PBMC from CCC patients cultured with P2β or CP0 proteins, failed to proliferate and express CD25 and HLA-DR on T cell populations. However, multiplex cytokine assays showed that these antigens triggered higher secretion of IL-10, TNF-α and GM-CSF by PBMC as well as both CD4+ and CD8+ T cells subsets of CCC subjects. Upon T. cruzi lysate stimulation, PBMC from CCC patients not only proliferated but also became activated within the context of Th1 response. Interestingly, T. cruzi lysate was also able to induce the secretion of GM-CSF by CD4+ or CD8+ T cells.Conclusions/Significance:Our results showed that although the lack of PBMC proliferation in CCC patients in response to ribosomal P proteins, the detection of IL-10, TNF-α and GM-CSF suggests that specific T cells could have both immunoregulatory and pro-inflammatory potential, which might modulate the immune response in Chagas' disease. Furthermore, it was possible to demonstrate for the first time that GM-CSF was produced by PBMC of CCC patients in response not only to recombinant ribosomal P proteins but also to parasite lysate, suggesting the value of this cytokine to evaluate T cells responses in T. cruzi infection.Fil: Longhi, Silvia Andrea.     Genome of Herbaspirillum seropedicae Strain SmR1, a Specialized Diazotrophic Endophyte of Tropical Grasses

    The molecular mechanisms of plant recognition, colonization, and nutrient exchange between diazotrophic endophytes and plants are scarcely known. Herbaspirillum seropedicae is an endophytic bacterium capable of colonizing intercellular spaces of grasses such as rice and sugar cane. The genome of H. seropedicae strain SmR1 was sequenced and annotated by The Paraná State Genome Programme—GENOPAR. The genome is composed of a circular chromosome of 5,513,887 bp and contains a total of 4,804 genes. The genome sequence revealed that H. seropedicae is a highly versatile microorganism with capacity to metabolize a wide range of carbon and nitrogen sources and with possession of four distinct terminal oxidases. The genome contains a multitude of protein secretion systems, including type I, type II, type III, type V, and type VI secretion systems, and type IV pili, suggesting a high potential to interact with host plants. H. seropedicae is able to synthesize indole acetic acid as reflected by the four IAA biosynthetic pathways present. A gene coding for ACC deaminase, which may be involved in modulating the associated plant ethylene-signaling pathway, is also present. Genes for hemagglutinins/hemolysins/adhesins were found and may play a role in plant cell surface adhesion. These features may endow H. seropedicae with the ability to establish an endophytic life-style in a large number of plant species

    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    This research work had as the aim of verifying the efficiency of five combined concentrations of hydrogen cyanamide (CH) in four consecutive pruning times of sprouting vine and harvest anticipation of ‘Niagara Rosada’ in the county of Ponta Grossa, PR. The experiment had a factorial design (4 x 5) with 3 replications and 3 plants per parcel. The analysis were sprouting percentage, number of clusters per plant, rate of production and average weight per cluster. The effect of CH was shown only at the first pruned time when sprouting but the occurrence of frosting decreased the yield. The delay in the process of pruning showed a higher number of cluster per plant, which consequently resulted in yield increase.O experimento foi conduzido com o objetivo de verificar a eficiência da combinação de cinco concentrações de cianamida hidrogenada (CH) e quatro épocas de poda, na quebra da dormência e antecipação da colheita da videira ‘Niagara Rosada’ no município de Ponta Grossa – PR. O delineamento experimental foi em blocos casualizados no esquema fatorial 4 x 5 (épocas de poda x concentrações de CH) , com 3 repetições e 3 plantas por parcela. Foram analisados a porcentagem de brotação, o número de cachos por planta, a produção por planta e a massa média dos cachos. Houve efeito da aplicação de cianamida hidrogenada no aumento da brotação das gemas apenas para a primeira época de poda (06/08/2007). A primeira época de poda antecipou em uma semana a colheita, embora a ocorrência de geadas tenha prejudicado a produção. O atraso na época de poda proporcionou um maior número de cachos por planta e a maior produção

    Concentrations of 137Cs, K and Na in fruits of lemon (Citrus limon B.) and of K and Na in fruits of coconut (Cocos nucifera L.) trees were measured by both gamma spectrometry and neutron activation analysis, with the aim to understand the behaviour of monovalent inorganic cations in tropical plants as well as the plant ability to store these elements. Similar amounts of K+ were incorporated by lemon and coconut trees during the growth and ripening processes of its fruits. The K concentration decreased exponentially during the growth of lemons and coconuts, ranging from 13 to 25 g kg− 1 dry weight. The incorporation of Na+ differed considerably between the plant species studied. The Na concentration increased linearly during the lemon growth period (0.04 to 0.70 g kg− 1 d.w.) and decreased exponentially during the coconut growth period (1.4 to 0.5 g kg− 1 d.w.). Even though radiocaesium is not an essential element to plants, our results have shown that 137Cs incorporation to vegetable tissues is positively correlated to K distribution within the studied tropical plant species, suggesting that the two elements might be assimilated in a similar way, going through the biological cycle together. A mathematical model was developed from the experimental data allowing simulating the incorporation process of monovalent inorganic cations by the fruits of such tropical species.
